1.Fabrication of drug-loaded tissue engineering scaffold and its curative effect for chronic osteomyelitis in rabbits

Objective To investigate the anti-infection and bone repair effects of composite scaffolds encapsulated with vancomycin and bone morphogenetic protein-2 (BMP-2).Methods Chitosan (CS) solution and nano-hydroxyapatite (n-HAP) mixed in a ratio of 1∶1 were made into solid materials by freeze drrying technology,and immerged into BMP-2/polylactic acid (PLA) and vancomycin/PLA microsphere solution to fabricate drug-loaded scaffolds.Efficiency of drug release was measured.After the induction of chronic infectious tibial bone defect,60 New Zealand white rabbits were assigned to isolate debridement group (Group Ⅰ),n-HAP/CS scaffold group (Group Ⅱ),n-HAP/CS + vancomycin/PLA group (Group Ⅲ) and n-HAP/CS + vancomycin/PLA + BMP-2/PLA group (Group Ⅳ) according to the random number table,with 15 rabbits per group.After 8 weeks of treatment,X-ray filming,Norden score,HE staining and narrow culture were performed to evaluate the anti-infection ability and bone repair ability in each group.Results Pore size of the n-HAP/CS scaffold constructed in this study ranged from 80 to 270 μm (mean,146.53 μm).Biomechanical test showed the elastic modulus of the scaffold material was (27.50 ± 1.58)MPa and the compressive strength was (0.75 ± 0.04)MPa.BMP-2/PLA and vancomycin/PLA microspheres into ball shape uniformly distributed on the scaffold pores.Drug-loaded scaffold showed relatively stable release of vancomycin and BMP-2 in a period of 18 days.With the extended time amount of the drug release gradually reduced.On the X-ray films 8 weeks after the treatment,sinus formation was visible in groups Ⅲ and Ⅳ,and emergence of periosteal reaction with focal low density shadow was demonstrated in groups Ⅰ and Ⅱ.Norden scores of groups Ⅰ,Ⅱ,Ⅲ and Ⅳ were (2.64 ±0.33) points,(2.63 ±0.36) points,(1.14 ±0.29) points and (0.89 ±0.42) points respectively,indicating group Ⅳ ranked the lowest compared with other three groups (P<0.05) and group Ⅲ showed significant difference in comparison with groups Ⅰ and Ⅱ (P <0.05).HE staining showed massive infiltration of inflammatory cells and destroyed bone trabeculas in groups Ⅰ and Ⅱ,trabecular bone arranged neatly in groups Ⅲ and Ⅳ,and thicker trabecular bone in group Ⅳ than in group Ⅲ.Marrow culture showed obvious pale yellow colony formation in groups Ⅰ and Ⅱ,but no colony formation in groups Ⅲ and Ⅳ.Conclusions Vancomycin/BMP-2-loaded tissue engineering scaffolds are constructed successfully.Drug-loaded n-HAP/CS composite scaffold is identified as an ideal implant material for treatment of chronic osteomyelitis in rabbits
2.Quantitative changes of anterior segment before and after implantable collamer lens implantation for high myopia with Pentacam anterior segment system

?AlM: To analyze the quantitative changes of anterior segment by Pentacam anterior segment system and to evaluate the safety and effectiveness after implantable collamer lens ( lCL) implantation for high myopia.
?METHODS:Twenty- one patients with 39 eyes were implanted lCL in our hospital from September 2011 to February 2013. The follow-up was 6 ~ 12mo. Central anterior chamber depth ( ACD ) , anterior chamber angle ( ACA) , anterior chamber volume ( ACV) were measured by Pentacam preoperatively, 1wk, 1 and 6mo postoperatively. The distance between crystal lens and lOL ( lCL- vault ) were measured at 1wk, 1 and 6mo postoperatively. Preoperative uncollected visual activity ( pre - UCVA ) , best corrected visual acuity ( BCVA ) , postoperative uncorrected visual acuity ( post-UCVA) and intraocular pressure ( lOP) were recorded. The paired t-test was used to access the data preoperatively and postoperatively in the statistical way by using SPSS 18. 0 statistical software.
? RESULTS: Preoperatively, at 1wk, 1 and 6mo postoperatively, ACD were 3. 27±0. 22, 3. 02±0. 33, 2. 98±0. 31, and 2. 98±0. 32mm. ACA were (40. 39±5. 40)o, (26. 70±4. 47)o, (26. 96±4. 48)o, and (26. 95±4. 45)o. ACV were 207. 74±25. 43, 122. 87±17. 58, 128. 05±17. 84, and 128. 64±17. 50mm3 , respectively. There were statistically significant differences between preoperative and postoperative data in ACD, ACA, ACV (P<0. 05). The distances between crystal lens and lOL ( lCL-vault) were 0. 49±0. 13, 0. 46±0. 13, and 0. 46±0. 14 at 1wk, 1 and 6mo postoperatively. There were statistically significant difference between 1wk and the others (P<0. 05). There were statistically significant different between the preoperative BCVA and postoperative BCVA(P<0. 05). There was no contact between lOL and the crystal len. The anterior chamber angle was all open. There were no serious complications such as cataract, glaucoma, cystoid macular edema, retinal detachment, etc.
?CONCLUSlON:ACD, ACA, ACV by Pentacam anterior segment system are found to be decreased after the operation, but there is still a relative safe distance between lOL and the crystal len. lCL implantation is a safe, efficacy method in correcting high myopia, but the long-term effects will require further observation.
4. Antitumor efficacy of photodynamic therapy-generated glioma vaccine from dendritic cells

Objective: To investigate the killing activity of cytotoxic T lymphocytes (CTLs) stimulated by dendritic cells (DCs) loaded with C 6 glioma antigen eluted by mild acid after photodynamic therapy(PDT) and test the direct influence of PDT on the immunogenicity of C6 glioma cells. Methods: Progenitors of DCs were isolated from rat bone marrow and cultured with rat recombinant interleukin-4 and rat recombinant granulocyte-macrophage colony stimulating factor to differentiate mature DC. After PDT, the surface antigen peptides were eluted by mild acid from the still adherent C 6 cells and the whole cell antigens were extracted from the supernatant. For the PDT-untreated C 6 cells, tumor antigens were made by freeze/thaw or mild acid elution method. Then the four types of antigens were used to pulse DCs to generate DC vaccines. SD rats were inoculated with the four DC vaccines to prepare antigen-specific CTLs. The killing activities of spleen CTLs were determined in vitro. Results: The mild acid-eluted surface antigens from PDT-treated C6 cells promoted the maturation of DCs most efficiently. CTLs stimulated by the DC vaccine had highest cell-killing activity in vitro (95.5 ± 1.6)%. The cell-killing rate was (90.2 ± 2.4) % for the CTLs pulsed by whole cell antigens from supernatant of PDT-treated C 6 glioma cells, (73.3 ± 2.7)% for the CTLs pulsed by acid-eluted antigen from PDF-untreated C 6 glioma cells, (63.6 ± 4.9)% for the CTLs pulsed by antigen from PDF-untreated and frozen-thawed C 6 glioma cells, and 0 for PBS control group. Conclusion: PDT directly enhancs immunogenicity of tumor cells. The DC vaccine generated by the antigen peptides eluted by mild acid from PDT-treated tumor cells has a wide prospect of application and research values.
