1.Diurnal variation, moon phase and seasonal variation in onset of stroke

ObjectiveTo determine the diurnal variation,moon phase,and seasonal variation in the onset of stroke. MethodsThe time parameters,including the time,the moon phase and the season of the stroke occurrence for 905 in-patients from 1989-2001 were investigated.ResultsThere was a significant difference (D=0.155,P<0.01) for circadian rhythm in the onset of 321 cases with cerebral infarction,and the incidence frequency at dawn was much higher than that at other time,but the difference of the moon phase and seasonal rhythm in the onset of cerebral infarction were not significant(P>0.05).There were not diurnal variation,moon phase and seasonal rhythms in the onset for cerebral haemorrhage(P>0.05).ConclusionsThe cerebral infarction occur more frequently at dawn time.
5.The special values of the time phases in liver enhancement scanning with multi-slices helical CT

Objective:To study the special values of the time phases in liver enhancement scanning with multi-slices helical CT.Methods:The relationship between the image features and the time phases was retrospectively analyzed with 143 cases that underwent liver enhancement scanning with multi-slices helical CT.Results:The time phases of liver enhancement scanning included hepatic artery earlier phase,hepatic artery metaphase,hepatic artery advanced phase,hepatic portal venous phase,hepatic portal venous-hepatic venous phase,hepatic venous phase and lag period.Conclusion:The different image features have the special value in the different time phases when liver was scanned with multi-slices helical CT.They ought to be differently treated and used according to the different objectives of liver enhancement scanning.
6.Preliminary study on cancer stem-like cells(side population)in human colorectal cancer cell lines

Objective:To investigate the content of cancer stem-like cells in different human colorectal cancer ce11 lines and explore the methods for enriching cancer stem-like cells in SW480.Methods:The percentage of side population cells(SP)in human colorectal cancer cell lines LOVO,SW480,HCT116 were detected by flow cytometry.SW480 cell line was cultivated in serum-free medium(SFM)supplemented with growth factors and cancer stem-like cells reforming into floatingspheres were isolated.The isolated cancer stem-like cells were identified by limiting-dilution assay,differentiation assay,self-renewal assay,alternative cultivation assay.Results:The percentage of SP cells was 1.0% in LOVO,1.2% in SW480 and 1.3% in HCT116.In the absence of serum,a minority(0.54% ̄0.62%)of cancer stem-like cells in SW480 cells survived,proliferated and assembled into the suspended tumor cell spheres.SW480 cancer stem-like cells possessed proliferative,self-renewal and differentiation potential,which were responsible for the floating tumor clone.Serum addition into SFM resulted in the proliferation of cancer stem-like cells;after several generations and alternated cultivation in SSM and SFM,cancer stem-like cells maintained their characters.Conclusion:LOVO,SW480 and HCT116 cell lines contain a tiny minority of SP cells with stem cell properties.The cancer stem-like cells in SW480 line can be maintained in SFM using a floating culture method.
7.Construction and identification of lentiviral vector of RNA interference of GF?1 gene

Objective: To construct a lentiviral vector of RNA interference(RNAi) of TGF-?1 gene. Methods: Three target sequences were selected according to TGF?1 mRNA sequence firstly,then three pairs of oligonucleotide sequences according to these target sequences and one pair of negative control oligonucleotide sequence were designed and synthesized. The annealed oligonucleotide fragments were subcloned into pRNA-U6/Lenti plasmid. After identified by restriction enzyme digestion and sequencing ,these plasmids were cotransfected along with ViraPowerTM Lentiviral Packaging Mix into 293FT cells for the package of lentiviral particles. Then the lentiviral vector particles were transfected into Hela cells and TGF-?1 expression in the transfected cells was assayed by Real-Time PCR and ELISA. Results: It was confirmed by digestion and sequencing that lentiviral vectors had the correct structure and could express high titer of virus. After transfected into Hela cells,TGF-?1 expression was decreased significantly by two of these lentiviral vectors at both the protein and mRNA level and the first vector had the best interefence efficiency. Conclusion: The lentivirus RNAi vector of TGF-?1 was constructed successfully.
8.Prospects and research situation of insect-drugs

The cognition,applications and research of insect-drugs were reviewed in this paper from the Spring and Autumn Period to modern time. From this we can appreciate the development of application of insect-drugs by physicians of past generations. This paper,based on textual research of ancient books,excavated the therapeutic effects and extended the clinical application. And to explore the mechanism of insect drugs from multi-angle,multi-channel,multi-target by making the best of the modern biological technology. The application of insect drugs would make new progress and play a more outstanding role in prevention and treatment of human disease.
9.Establishment of an experimental model of autogenousoxygenator cardiopulmonary bypass in dogs

Objective:To establish an experimental autogenous oxygenator cardiopulmonary bypass in dogs.Methods:Sixteen mongrel dogs were used.Under intravenous anesthesia and artificial respiration,thoracotomy and cardiac cannulations were performed:right atrium cannulation and pulmonary artery cannulation,connecting to reservoir and pump,the right heart bypass system was established;left atrium and ascending aotra cannulation,connecting to reservoir,heat exchanger and pump,the left heart bypass was established.Results:The experimental model was established successfully in fifteen dogs and one failed.Conclusion:The susscessful establishment of this experimental model depends on skillful cannulation,balance of left and right circulation,appropriate body temperature.
10.Repair of ventricular septal defect through subaxillary minithoractomy

Objective:To introduce the clinical experiences of repairing ventricular septal defect through right subaxillary minithoracotomy and to evaluate it's clinical effect.Methods:46 cases of VSD were repaired through right subaxillary minithoracotomy.The sizes of the defects varied from 0.4~0.8cm in diameter and all of them were membranous or conventricular defects.The average age was 7.1 ys.Results:There was no operative death or complication.The lenth of incision varied from 5~10cm.Cardiopulmonary bypass time were 21~42 minutes.Postoperative hospitalized time were 6 to 10 days.Conclusion:Repairing of ventricular septal defect through subaxillary minithoracotomy have the advantages of minimal trauma,hidden incision and quicker recovery.
