1.Effects of altered metabolic enzymes on epigenetic status in gliomas

Alteration of metabolism and epigenetic status are two new focuses in the field of cancer re -search.Recently,more and more reports have demonstrated that the changes of both metabolic enzymes and al -tered epigenetic status have close interrelationship with the pathogenesis and development of gliomas in human . We discuss here the effects of altered metabolic enzymes on epigenetic status in gliomas based on two important altered metabolic enzymes ,with the aim to unlock the molecular mechanism on how gliomas occur and progress . This newfound molecular mechanism of how altered metabolic enzymes impact on epigenetic status in gliomas may contribute to the identification of novel therapeutic targets to gliomas in the future .
3.Correlation of maternal serum vitamin D status and pregnancy outcome

Objective To investigate the effects of 25‐hydroxy vitamin D [25(OH)D] status on the pregnancy outcome in the second and third trimester pregnant women living .Methods A total of 143 pregnant women (12 -42 weeks gestational age) were participated in the study .Their serum 25(OH)D concentrations were detected by liquid chromatography‐mass spectrometry method .Finally ,vitamin D status was assessed according to sufficiency group(n= 32) ,inadequacy group(n= 49) ,insufficiency group(n=51) and deficiency group(n=11) .Retrospective analysis were adopted to follow up the pregnancy outcome of pregnant women .Results (1)The average serum 25(OH)D level was(22 .49 ± 9 .51)ng/mL in the second and third trimester of pregnancy . (2)Abortion rate were 3 .1% (1/32) in sufficient group ,2 .0% (1/49)in inadequacy group ,11 .8% (6/51) in insufficiency group , 36 .4% (4/11) in deficiency group ,fetal distress incidence were 3 .1% (1/32) ,8 .2% (4/49) ,11 .8% (6/51) ,36 .4% (4/11) in each group .Abortion ,fetal distress rate were higher in deficiency group compared with the other three groups(P<0 .05) .(3)Bacterial vaginosis incidence were 0(0/32) ,4 .1% (2/49) ,13 .7% (7/51) ,27 .3% (3/11)in each group ,deficiency group and insufficiency group were higher(P<0 .05) .(4)Otherwise ,the gestation period were (39 .48 ± 1 .44) weeks in sufficient group ,(39 .00 ± 1 .70) weeks in inadequacy group ,(38 .09 ± 2 .01) weeks in insufficiency group ,(37 .29 ± 0 .49) weeks in deficiency group ,pregnancy cycle was significantly shorter when 25(OH)D<20 .00 ng/mL (P<0 .05) .Conclusion Vitamin D deficiency in pregnant women could increase the risk of abortion ,fetal distress ,bacterial vaginosis ,and significantly shorten the gestation period .
4.Construction of three lentiviral vectors expressing HPV16 E5, E6 or E7 oncogene and respective infection of oral epithelial cells with the vectors

Objective To provide the basis for the study of HPV16 E5 gene inthe pathogenesis of oral squamous cell carcinoma by constructing three lentiviral vectors expressing HPV 16 E5, E6 or E7 oncogene and transfecting o-ral epithelial cells with the vectors.Methods HPV16 E5, E6 or E7 oncogene was amplified using PCR and liga -ted into the lentiviral vector pLVX-AcGFP-N1 respectively to construct vectors pLVX -AcGFP-E5, pLVX-AcGFP-E6 and pLVX-AcGFP-E7, then respectively cotransfected 293T cells with packaging plasmids , viral supernatant was collected to respectively transfect oral epithelial cells .Afterpuromycin screening,oral epithelial cells with HPV 16 E5, E6,or E7 oncogene transfection were constructed , then reverse transcription PCR and western blotassays were performed for verifying HPV16 E5, E6 or E7 expression.Results pLVX-AcGFP-E5, pLVX-AcGFP-E6 and pLVX-AcGFP-E7 were successfully constructed, oral epithelial cells expressing HPV 16 E5, E6 or E7 oncogene wereacquired, HPV16 E5,E6 or E7 expression was confirmed in oral epithelial cellsthrough reverse transcription PCR and western blot assays.Conclusion Three lentiviral vectors expressing HPV16 E5, E6 or E7 oncogene can successfully infect oral epithelial cells .

7.Efficacy observation of long-time needle retaining at Baihui(GV20)combined with multidirectional point-toward-point needle insertion with needle shaking at Fengchi(GB20)for cervical vertigo

Objective:To observe the clinical curative effect of long-time needle retaining at Baihui(GV20)combined with multidirectional point-toward-point needle insertion with needle shaking at Fengchi(GB20)for the treatment of cervical vertigo(CV)and its influence on the blood flow velocity of vertebrobasilar arteries. Methods:Seventy patients with CV were randomly divided into a treatment group(35 cases,1 dropout)and a control group(35 cases,2 dropouts)according to the random number table method.Those in the treatment group were treated with long-time needle retaining at Baihui(GV20)combined with multidirectional point-toward-point needle insertion with needle shaking at Fengchi(GB20),and those in the control group were treated with conventional acupuncture.The treatment was performed every other day,7 sessions as a treatment course,for a total of 2 courses.The clinical efficacy was compared between the two groups by observing changes in the evaluation scale for symptoms and functions of cervical vertigo(ESCV)and the mean blood flow velocity(Vm)of vertebrobasilar arteries. Results:The total effective rate and the cured plus markedly effective rate were 91.2%and 79.4%,respectively,in the treatment group,versus 78.8%and 54.5%in the control group,respectively,with a statistically significant difference in the cured plus markedly effective rate between the two groups(P<0.05).The ESCV score and the Vm of vertebrobasilar arteries in the two groups improved significantly after treatment.The Vm of the left vertebral artery(LVA),right vertebral artery(RVA),and basilar artery(BA)increased in patients with low and normal flow velocities(P<0.05),and the Vm of the LVA,RVA,and BA decreased in patients with a high flow velocity(P<0.05);the results in the treatment group were significantly better than those in the control group(P<0.05). Conclusion:Long-time needle retaining at Baihui(GV20)combined with multidirectional point-toward-point needle insertion with needle shaking at Fengchi(GB20)can significantly reduce the clinical symptoms of CV and regulate the blood flow rate of vertebrobasilar arteries bidirectionally,and thus is an effective therapy for CV.
10.Sterol composition in field-grown and cultured mycelia of Inonotus obliquus

Sterols are one of the active classes of compounds in Inonotus obliquus for their effective therapy of many diseases. In field environment, this fungus accumulates large amount of sterols. In cultured mycelia, however, this class of compounds is less accumulated. For analyzing the factors responsible for differing sterol composition, the field-grown and cultured mycelia were extracted with 80% ethanol at room temperature and total sterols were prepared using silicon gel column chromatography followed by identification using either GC-MS or spectroscopic methods. For culturing Inonotus obliquus, the seed culture was grown either in basic medium consisting of glucose (2%), yeast extract (0.5%), KH2PO4 (0.01%), MgSO4·7H2O (0.05%) and distilled water at pH 6.5, or the basic medium supplemented with serial concentrations of AgNO3. The results indicated that field-grown mycelia contained lanosterol and inotodiol (comprised 45.47% and 25.36% of the total sterols, respectively) and other 10 sterols (comprising the remaining 30.17%) including ergosterol biosynthetic intermediates such as 24-methylene dihydrolanosterol, 4,4-dimethylfecosterol, 4-methyl fecosterol, fecosterol and episterol. Column chromatography also led to the isolation of lanosterol, Inotodiol, trametenolic acid, foscoparianol B and a new triterpenoid foscoparianol D in field-grown mycelia. In comparison, the cultured mycelia only contained three sterols with ergosterol as the predominant one (82.20%). Lanosterol only accounted for 3.68%. Supplementing Ag+ into the culture at 0.28 μmol·L-1 greatly enhanced content of lanosterol (accounting for 56.81%) and decreased the content of ergosterol (18.5%) together with the presence of intermediates for ergosterol biosynthesis. These results suggested that the sterol composition in mycelia of the fungus can be diversified by supplementing substances inhibiting enzymatic process towards the synthesis of ergosterol. Harsh growth conditions in field environment (I.e. temperature variation, UV irradiation etc.) can delay the synthesis of ergosterol and hereby diversify the sterol composition in the mycelia of Inonotus obliquus.
