3.The relationship between angle of puncture and distribution of bone cement of unilateral percutaneous kyphoplasty for the treatment of thoracolumbar compression fractures.

OBJECTIVETo explore the relationship of bone cement distribution and the puncture angle in the treatment of thoracolumbar compression fractures with unilateral percutaneous kyphoplasty (PKP).
METHODSThe clinical data of 37 patients with thoracolumbar osteoporotic compression fractures underwent PKP between January 2013 to March 2014 were retrospectively analyzed, all punctures were performed unilaterally. There were 6 males, aged from 65 to 78 years old with an average of (71.83 ± 6.15) years; and 31 females, aged from 57 to 89 years old with an average of (71.06 ± 7.89) years. Imaging data were analyzed and puncture angle and puncture point were measured before operation. According to the measured data, the puncture were performeds during the operation. Distribution area of bone cement were calculated by X-rays data after operation. The effect of bone cement distribution on suitable puncture angle was analyzed; VAS score was used to evaluate the clinical effects.
RESULTSThe puncture angle of thoracic vertebrae in T8-T12 was from 28° to 33° with an average 30.4°; and the puncture angle of lumbar vertebrae in L1-L5 was from 28° to 35° with an average of 31.3°. Postoperative X-rays showed the area ratios of bilateral bone cement was 0.97 ± 0.15. Bilateral diffuse area were basic equal. Postoperative VAS score decreased significantly (1.89 ± 1.29 vs 7.03 ± 1.42).
CONCLUSIONThrough measure imaging data before operation with PKP,the puncture point and entry point can be confirmed. According the measured data to puncture during operation, unilateral puncture can reach the distribution effect of the bilateral puncture in the treatment of thoracolumbar compression fractures.

4.Effect of glucocorticoid on the expression of PGC-1α mRNA in brown adipose tissue and skeletal muscle of obese rats fed with high-fat diet

Real-time fluorescent quantitative PCR was used to examine PGC-1α mRNA expression in brown adipose tissue and skeletal muscle of rats. The results showed that the expression levels of PGC-1α mRNA in brown adipose tissue and skeletal muscle of obese rats were lower than those of the normal ones (all P<0.01). After high dose glucocorticoid treatment, the levels of PGC-1α mRNA expression in brown adipose tissue and skeletal muscle, both in normal and obese rats, were decreased significantly.
5.Effects of xuefu zhuyu decoction on antioxidant and drug-metabolizing enzymes in liver of rats.

Xuefu Zhuyu decoction (XFZYD) is a famous traditional Chinese medicine (TCM) formula, is widely used in the treatment of cardiovascular and cerebrovascular diseases in China over one hundred years. But its effect on antioxidant and drug-metabolizing enzymes are unknown. This study was to observe the effects of Xuefu Zhuyu decoction (XFZYD) on the activities of antioxidant and drug metabolism enzymes (DMEs) in liver of rats. Male SD rats, treated with XFZYD at the dosage of 3.51, 7.02 and 14.04 g x kg(-1) per day for 15 days, serum were collected, tissue fluid, cytosols and microsomes isolated from liver tissues were prepared by centrifugation according to the standard procedure, the activities of antioxidant enzymes and drug-Metabolizing Enzymes were determined by UV-V is spectrophotometer. In serum, the activities of AST was not significantly affected by the treatment with XFZYD, at the high- est dose, the levels of ALT, Cr and BUN were significantly decreased (P < 0.05). GPX were significantly increased at the dose of 7.02, 14.04 g x kg(-1) (P < 0.05), CAT were significantly increased at the highest dose (P < 0.05). T-SOD was not significantly af- fected by this treatment. In the liver tissue, GPX was significantly increased at the dose of 3.51, 7.02 g x kg(-1) (P < 0.05), GST, CAT and T-SOD were not significantly affected following this treatment. In cytosols, GST was significantly increased at the dose of 3.51 g x kg(-1) (P < 0.05), T-SOD was remarkable induced at the dose of 3.51 and 7.02 g x kg(-1) (P < 0.05). In microsomes, XFZYD had no significant effect on Cytochromeb5, NADPH-Cytochrome P450 reductase, CYP3A, CYP2E1 and UGT, XFZYD significantly in- duced GST at the dose of 3.51 and 7.02 g x kg(-1) (P < 0.05), and the level of GSH were significantly increased by XFZYD at the dose of 3.51, 7.02 and 14.04 g kg(-1) (P < 0.05). These findings suggest XFZYD can induce the activities of GPX, CAT, SOD, GST and increase GSH level in liver of rats, which indicate XFZYD may have detoxification and antioxidant functions.

6.Telomerase Activity in Condyloma Acuminatum:Correlation with Decreased Apoptosis

Objective To study the telomerase activity,the keratinocyte apoptosis in condyloma ac-cuminatum(CA)and the correlation between them.Methods CA specimens from30patients were stud-ied,and compared with normal tissue and tumor cell lines.Telomerase activity was detected with telomeric repeat amplification polymerase chain reaction(TRAP)-ELISA.Terminal deoxynucleotidyl transferase(TdT)-mediated biotin dUTP nick end-labeling(TUNEL)was used to evaluate apoptotic cells.Results Increased telomerase activity was detected in27(90%)patients with CA,with A 450 ranging between0.50and2.76(mean1.3022).Apoptotic keratinocytes were found in24out of30CA cases(80%).A statistically signifi-cant inverse correlation was found between telomerase activity and apoptotic index(r =-0.52,P=0.004).Conclusion Keratinocyte telomerase is activated in condyloma acuminata,which is correlated with the downregulation of apoptosis,thus they might be involved in the pathogenesis of CA.
8.Mechanical properties of thermodynamic nickel-titanium archwires in a simulated oral environment in vitro.

OBJECTIVEThis study was to investigate the mechanical properties of thermodynamic nickel-titanium (NiTi) archwires of different brands under controlled temperature and in artificial saliva to provide some reference for its clinical application.
METHODSUsing a modified partial dental arch bending system, four types of thermodynamic NiTi archwires were tested under controlled temperature (25, 33, 37, 60 degrees C) and in artificial saliva. An Instron Universal Testing Machine was used to apply 3 mm deflections of archwires in the lateral incisor area. The load-deflection curves were determined from the passive position to deflections of 3 mm. The following variables from unloading curves were calculated: Stiffness between 2.5 mm UDP (unloading deflection point) to 0.5 mm UDP after unloading. Any statistically significant differences in these variables for the different brands, temperature and moments levels were analyzed using one-way analysis of variance.
RESULTSUnloading values of four types thermodynamic nickel-titanium archwires increased with controlled temperature rising. The unloading stiffness of the same brand archwires were approximate at 25, 33, 37 degrees C, while their unloading stiffness increased obviously at 60 degrees C.
CONCLUSIONDifferent brands thermodynamic NiTi archwires demonstrated different mechanical behaviours, but all of them exhibited super elasticity and practical shape memory effect. The four thermodynamic NiTi archwires displayed temperature sensitivity at the four test temperatures.

9.Effects of acupuncture at "Sifeng" (EX-UE 10) on serum zinc and gastric emptying rate in the rat of experimental spleen deficiency.

OBJECTIVETo probe the mechanism of acupuncture at Sifeng (EX-UE 10) for treatment of infant anorexia.
METHODSHealthy rats were randomly divided into a control group, a model group and an acupuncture group. The model rats were made by intramuscular injection of Reserpine. The acupuncture group were treated with pricking at Sifeng (EX-UE 10) once daily for 2 weeks. After that, the gastric emptying rate and contents of Zn in the red cell and serum were observed in all the groups.
RESULTSCompared with the model group, the gastric emptying rate was accelarated, and the serum Zn content was increased in the acupuncture group (P<0.01, P<0.05).
CONCLUSIONAcupuncture at Sifeng (EX-UE 10) has significant therapeutic effect on the rat with spleen deficiency, accelerating the gastric emptying rate and increasing the serum Zn content.

10.Inhibitory effect of resveratrol on cardiac fibroblast proliferation induced by angiotensin II.

OBJECTIVETo observe the effect and mechanism of resveratrol on cardiac fibroblast (cFs) proliferation induced by angiotensin II (Ang II).
METHODSThe in vitro cFs proliferation model was established by stimulating cultured cFs of new born rats with Ang II by differential attachment method. Cell proliferation was measured by MTT assay, and the effect of resveratrol, L-NAME and ODQ on cell proliferation were observed respectively. Besides, the hypertrophic response of cFs was estimated by measuring expressions of atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP) mRNA, with the levels of ANP and BNP in culture medium determined by radioimmunoassay and ELISA respectively; and their mRNA expressions determined by reverse transcription polymerase chain reaction (RT-PCR). Level of nitric oxide (NO) in the culture medium was measured by Griess reagent; nitric oxide synthase (NOS) level by chemical colorimetric method; and cGMP by radioimmunoassay.
RESULTSResveratrol at the dose of 25-100 micromol/L inhibited cFs proliferation in a time and dose dependent manner, which could be partially blocked by pretreatment with L-NAME or ODQ. NO and cGMP levels increased, ANP, BNP levels and their mRNA expression lowered after resveratrol treatment.
CONCLUSIONResveratrol in a definite concentration range could inhibit cFs proliferation and hypertrophic response induced by Ang II, up-regulating the signal pathway of NO and cGMP might be one of the acting paths of the inhibitory effects.
