1.The mechanical simulation analysis of instantaneous distal moving trend of the maxillary first molar in forsus
Journal of Chongqing Medical University 1986;0(02):-
Objective:To simulate the instantaneous distal moving trend of the maxillary first molar based on the three-dimensional finite element mode(l3-FEM)of Forsus distalizing the maxillary first molar with some visco-elastic-plastic characters and explore the favorable loading mode of molar distal movement in Forsus in order to provide biomechanical basis for clinical practice. Methods:A male patient with ClassⅡmalocclusion was selected. The 3-FEM of Forsus distalizing the maxillary first molar was built through spiral CT scanning,Mimics8.0,and Abaqus6.5 software. This study simulated instantaneous distal moving process of the maxillary first molar under three dif-ferent loading modes(loading force was 300 g,point of force was on the center of the crown's buccal surface, loading time was 1s),which included a distalizing force,a distalizing force+counterrotation moment,a distalizing force+countertipping moment+counterrotation moment. Results:When Forsus distalized the maxillary first molar, different loading modes lead to different PDL stress distributions. When the model was added by a distalizing force, the maxillary first molar would be relatively seriously inclined;when added by a distalizing force +counterrotation moment(Mt/F=9),the maxillary first molar would be relatively seriously rotated and when added by a distalizing force+ countertipping momen(tMt/F=9)+counterrotation momen(tMr/F=5),the distal surface of the PDL would bear the lightest stress, and the first molar would achieve approximate translation movement. Conclusion:When mandible is guided forward by Forsus, the maxillary first molar would achieve better molar translation effect if added certain countertipping and counterrotation moment
2.Effects of Fas, NF-κB and caspases on rat microvascular endothelial cell apoptosis induced by TNFα
Chinese Journal of Pathophysiology 2001;17(8):793-794
AIM: To study the apoptotic effect of TNFα on rat pulmonary microvascular endothelial cells (PMVEC) and the influences of Fas, NF-κB in its mechanism. METHODS: Apoptosis of PMVEC was analyzed and quantitated with TUNEL, flow cytometer. The distribution of NF-κB was detected via histoimmunochemical staining in TNF-treated cells and the control. Northern blot was applied to assess the influence of TNF on PMVEC Fas expression. Fas antibody was used to investigate the apoptotic effect of Fas on PMVEC. Activation of caspase-8 was detected with Western blot. Expression of caspase-3 was analyzed with histoimmunochemical staining. RESULTS: After treatment with 5×108 U/L TNF for 24 hours, viable PMVEC significantly diminished. Apoptosis rate was 14.0%±3.1% detected with TUNEL, and 13.1% with flow cytometer. Histoimmunochemical staining showed that NF-κB relocated from cytoplasm to the nuclear. When the cells were co-cultured with TNF and APDC, an NF-κB inhibitor, less cells were viable and more cells were positively stained with TUNEL. Fas expression in PMVEC was elevated treated with TNF. Apoptosis in PMVEC was found aggravated, when the cells were co-cultured with TNF and anti-Fas antibody. The positive rate was 24.1%±1.5% with TUNEL. Increase of caspase-8 activation was manifested by Western blot following TNF stimulation. Caspase-3 expression was found elevated using histoimmunochemical staining. Cell permeable caspase-3 inhibitor significantly ameliorated PMVEC apoptosis induced by TNF. CONCLUSION: 1. Large dose of TNF(5×108 U/L) can induce apoptosis in rat PMVEC. 2. NF-κB has a protective effect on PMVEC apoptosis. 3. TNF up-regulates Fas expression in PMVEC. And the latter takes a part in apoptosis. 4. TNF induced caspase-8 activation in PMVEC, and more caspase-3 was expressed. These may be involved in PMVEC apoptosis induced by TNF.
3.Portable instrument for arteriosclerosis assessment.
Chinese Journal of Medical Instrumentation 2014;38(1):6-10
A portable instrument for arteriosclerosis assessment containing sensor module, acquisition board and embedded module was developed for home care in this paper. The sensor module consists of one ECG module and three pulse wave extraction modules, synchronously acquiring human ECG and pulse wave signal of carotid, radial, and dorsal, respectively. The acquisition board converts the sensor module's analog output signals into digital signals and transmits them to the embedded module. The embedded module realizes the functions including signal display, storage and the calculation and output of pulse wave velocity. The structure of the proposed portable instrument is simple, easy to use, and easy to expand. Small size, low cost, and low power consumption are also the advantages of this device. Experimental results demonstrated that the proposed portable instrument for arteriosclerosis assessment has high accuracy, good repeatability and can assess the degree of atherosclerosis appropriately.

4.High-mobility group protein B1 and ischemic stroke
International Journal of Cerebrovascular Diseases 2016;24(3):263-267
High mobility group protein box 1 (HMGB1) is a typical nonhistone chromosomal protein. It has many celular functions in nucleus. Studies in recent years have showed that HMGB1 can be released to the outside of cels to exert a wide range of cytological effects. Ischemic stroke is one of the diseases with the highest morbidity and disability. More and more evidence has shown that HMGB1 plays a variety of important roles in the occurrence and development process of ischemic stroke. This article reviews the roles of HMGB1 in ischemic stroke.
5.Chloride channels and ischemic stroke
International Journal of Cerebrovascular Diseases 2017;25(3):285-288
Ischemic stroke is one of the diseases with the highest morbidity and disability.Hypertension is recognized as the most important independent risk factor for ischemic stroke.Vascular remodeling during the development of hypertension is the pathological basis of causing ischemic stroke.Studies have shown that vascular smooth muscle cell proliferation and apoptosis will lead to vascular remodeling. In addition, cerebral ischemia-reperfusion can result in neuronal damage and apoptosis.Recent research has shown that vascular remodeling and neuronal apoptosis are associated with chloride channels. At least 3 chloride channels including volume regulated chloride channel, calcium activated chloride channel and cystic fibrosis transmembrane conductance regulator are involved in these processes. This article reviews the roles of the 3 chloride channels in vascular remodeling, neuronal apoptosis, and ischemic stroke.
6.Effects of Fas, NF-?B and caspases on microvascular endothelial cell apoptosis induced by TNF?
Chinese Journal of Pathophysiology 2000;0(08):-
AIM:To study the apoptotic effect of TNF? on rat pulmonary microvascular endothelial cells (PMVEC)and the role of Fas, NF-?B in its mechanism. METHODS: Apoptosis of PMVEC was analyzed and quantitated with TUNEL, flow cytometer. Northern blot was applied to assess the influence of TNF? on PMVEC Fas expression. Fas antibody was used to investigate the apoptotic effect of Fas on PMVEC. Activation of caspase-8 was examined by Western blot. Expression of caspase-3 was analyzed with histo-immunochemical staining. RESULTS:Growth curve showed that TNF? suppressed endothelial cell proliferation in a dose-dependent manner. After treatment with 5?10 6 U/L TNF?, apoptotic rate was 14.0%?3.1% detected with TUNEL, and 13.1% with flow cytometer. When the cells were co-cultured with TNF? and APDC, an NF-?B inhibitor, less cells were viable and more cells were positively stained with TUNEL. Fas expression in PMVEC was elevated after TNF? treatment. Co-culturing with Anti-Fas antibody aggravated PMVEC apoptosis. Caspase-8 activity and caspase-3 expression was elevated. Caspase-3 inhibitor significantly ameliorated PMVEC apoptosis. CONCLUSION: Large dose of TNF? (5?10 6 U/L) can induce apoptosis in rat PMVEC. NF-?B has an anti-apoptotic effect in PMVEC. TNF? up-regulates Fas expression in PMVEC, and the latter takes a part in apoptosis. Caspase-8 and caspase-3 are involved in PMVEC apoptosis induced by TNF?.

Aim To investigate whether 8-bromo-7-methoxychrysin ( BrMC ) can reverse the epithelial-mesenchymal transition ( EMT) in sphere-forming cells (SFCs) derived from cervical cancer SiHa cell line. Methods Human cervical cancer SiHa cell line was cultured in vitro and then SFCs were obtained from Si-Ha cell line using suspension culture with the stem-condition culture system. After treatment with BrMC for 72 h, the morphology of SFCs of SiHa cell line was observed via monolayer adherence culture. The expres-sion levels of E-cadherin, N-cadherin and Twist1 pro-teins were analyzed using western blot assay. Results SFCs of SiHa cell line presented the mesenchyme-like( spindle-shaped) cell shape and exhibited reduced E-cadherin and elevated N-cadherin protein expres-sion, compared with the parental cells. After treatment with BrMC(1. 0 μmol·L-1 ) for 72 h, SFCs of SiHa cell line changed from spindle-shaped cells in to poly-gon shaped cells under monolayer adherence culture. After treatment with BrMC for 24 h, the expression lev-els of E-cadherin protein were up-regulated and the ex-pression levels of N-cadherin and Twist1 proteins were decreased in SFCs derived from SiHa cell line. Con-clusion BrMC effectively reverses EMT of SFCs de-rived from cervical cancer SiHa cell line, and its mechanism is involved in down-regulating the expres-sion of Twist protein.
