1.Hypoxia-inducible factor-1α expression in the collagen induced arthritis model and its significance

Objective To study the hypoxia-inducible factor-1α (HIF-1α) expression in the col-lagen induced arthritis(CIA) model and its role in rheumatoid arthritis (RA) pathogenesis. Methods We established CIA model, and taked CIA model of the ankle joint to have them HE staining and HIF-1α immu-nohistochemistry staining, then the HIF-1α expression in the tissues of RA was observed. Results The ex-pression of CIA synovial membrane of rat and the lower synodal both was expressed in HIF-1α. The positive expression of HIF-1α reached the highest volume in the first 21 days. Then, with the disease progression, the expression gradually decreased, which was in significantly positive relation with the synovial pathology score, synovial hyperplasia and angingenesis rating score, however, the expression was no positively correla-ted with the inflammatory infiltration. Conclusion The expression of HIF-1α in RA tissue is positively re-lated with inflammation severity, It indicates that HIF-1α is closely related to the occurrence and develop-ment of RA.
2.Epidemic characteristics and influencing factors of influenza-like illness outbreaks in schools in Nanchang City from 2011 to 2022

Abstract: Objective To analyze the epidemic characteristics and influencing factors of school influenza-like cases in 2011-2022 in Nanchang, so as to provide reference for the prevention and control of school influenza outbreaks. Methods The epidemiological relevant data of school influenza-like cases from 2011 to 2022 in Nanchang and the pathogen test results of respiratory tract samples were collected for epidemiological and etiology analysis. Results From 2011 to 2022, a total of 142 influenza-like cases were reported in schools in Nanchang, with a cumulative morbidity of 2 880 cases and a morbidity rate of 1.89%. A total of 1 263 samples were collected, with an overall positive influenza nucleic acid detection rate of 58.91%. The highest proportion of outbreaks occurred in 2017-2019, while the lowest incidence was in 2011-2013. Outbreaks mainly occurred from November to March of the following year (accounting for 79.58%), presenting obvious seasonal characteristics. The distribution was mainly in primary schools, accounting for 70.42% (100/142) of all outbreaks. From 2011 to 2022, all types of influenza viruses were tested, and more than 2 types of influenza viruses were prevalent each year. The dominant strains alternated between influenza A H1N1, influenza A H3N2 and Victoria lineages of type B influenza viruses. From 2011 to 2019, the influenza epidemic in schools in Nanchang showed a continuous upward trend. During COVID-19 in 2020-2022, Nanchang adopted a variety of non-drug prevention and control measures for COVID-19, and both the number of influenza epidemics in various schools and the number of influenza virus nucleic acid positive cases decreased. The average number of classes involved in the epidemic was (3±2), and the average duration of the epidemic was (8±4) days. Conclusions The outbreaks of influenza-like illness in Nanchang schools is highly prevalent in winter and spring, , with urban primary schools being the high incidence locations. Non-drug prevention and control measures for COVID-19 have an impact on the epidemic trend of influenza, so the continuous monitoring of the school influenza virus activities and improving the timeliness of the report will be conducive to the rapid control of the epidemic.
3.Fusion expression of D-amino acid oxidase from Trignoposis variabilis with maltose binding protein and Vitreoscilla hemoglobin.

D-amino acid oxidase (DAAO) is one of important industrial enzymes. To increase the solubility and activity of the TvDAAO from Trignoposis variabilis expressed in recombinant Escherichia coli (E. coli), a maltose binding protein (MBP) and Vitreoscilla hemoglobin (VHb) was introduced to fuse with N-terminal of the TvDAAO, respectively. Fusion protein of MBP-TvDAAO was constitutively expressed in JM105/pMKC-DAAO and inductively expressed in JM105/pMKL-DAAO. With respect to the control strain of BL21 (DE3)/pET-DAAO without MBP fusion, the constitutive fusion expression obtained 28% of soluble protein with 3.7 folds of solubility improvement. As for the inductive fusion expression, corresponding results changed to 17% and 1.8 folds, respectively. However, the DAAO activity significantly decreased in the MBP-fusing expression. Fusion protein of VHb-TvDAAO was constructed and inductively expressed in BL21 (DE3)/pET-VDAAO. Its DAAO activity highly reached 3.24 u/mL in flask culture, about 90% increase in contrast to the control without VHb.

4.Effect of different estrogen receptor subtypes on vascular endothelial growth factor protein expression in human breast cancer samples.

OBJECTIVESTo evaluate the relationship of vascular endothelial growth factor expression with estrogen receptor (ER) subtypes in fresh human breast cancer samples, and study the potential effect of ER subtypes on tumor angiogenesis.
METHODSWestern blot was used to detect the VEGF and ERbeta protein expression in 86 fresh samples of human breast cancer. ERalpha was analyzed by immunohistochemistry routinely.
RESULTSAmong 86 samples, 42 (48.8%) showed low expressed VEGF and 44 (51.2%) high expressed VEGF. The level of VEGF protein was correlated with ERbeta. In high expressed VEGF group, ERbeta was also highly expressed (chi(2) = 7.36, P < 0.01). But there was no significant difference between VEGF protein level and ERalpha (P > 0.05).
CONCLUSIONIn human breast cancer samples, VEGF may be related to ERbeta protein expression.

5.Efficacy of ivabradine combined with levosimendan in patients with acute myocardial infarction compli-cated with heart failure

Objective To observe the efficacy of ivabradine combined with levosimendan in patients with acute myocardial infarction complicated with heart failure.Methods A total of 78 patients with acute myocardial infarction complicated with heart failure admitted to the hospital from May 1,2020 to December 31,2022 were selected and divided into control group(n = 39)and study group(n = 39)by random number table method.In addition to basic treatment,the control group received levosimendan,and the study group ivabradine and levosi-mendan.The treatment period of both groups was 4 weeks.Compare two groups of patients'vital signs,clinical curative effect and heart function index,serological indexes,inflammatory factors and security.Results Compared with before treatment,the systolic blood pressure,diastolic blood pressure and heart rate of the two groups were decreased after treatment(P<0.05),and the systolic blood pressure,heart rate,diastolic blood pressure and heart rate of the study group were significantly decreased compared with the control group(P<0.05).Compared with the control group,the total effective rate of the study group was significantly higher(P<0.05).Compared with before treatment,the levels of CO and LVEF were increased(P<0.05),while the levels of LVEDV and LVESV were decreased(P<0.05).Compared with the control group,the levels of CO and LVEF in the study group were higher(P<0.05),and the levels of LVEDV and LVESV in the study group were lower(P<0.05).Compared with before treatment,the levels of cTnI,sST2 and NT-proBNP in both groups were decreased after treatment(P<0.05),and the serum levels of cTnI,sST2 and NT-proBNP in the study group were significantly decreased compared with the control group(P<0.05).Tumor necrosis factor α(TNF-α),myeloperoxidase(MPO)and highly sensitive C-reactive protein(hs-CRP)were all decreased after treatment in the two groups(P<0.05),and those in the study group were much lower(P<0.05)No difference between the incidence of adverse reactions to the total contrast the two groups(P>0.05).Conclusion Levosimendan combined with ivabadinehave a definite effect on patients with acute myocardial infarction complicated with heart failure for the improved cardiac function,reduced inflammation,regulated serum sST2,cTnI,and NT-proBNP level.It is also safe and reliable
6.Construction of Meridian and Collateral Homeostasis Model from Phenomics

By applying "homeostasis" to the study of the meridian and collateral system, the concept of meridian and collateral homeostasis has been proposed which refers to a balanced and stable state of meridian and collateral system, and plays an important role in maintaining body health and can provide a reference for the diagnosis and treatment of diseases. Phenomics realizes the cross-scale correlation from micro-phenotypic data, such as genome, proteome, and metabolome, to macro-phenotypic data, such as physiological state, behavioral activities, and external manifestations. From the perspective of phenomics, this paper proposes a meridian and collateral homeostasis dynamic mapping model of "macroscopic signs and microscopic expression". This model combines macro signs such as the four examinations of traditional Chinese medicine (TCM), biophysical indicators of acupoints, and micro expression information such as genes, proteins, and metabolism, and systematically investigates the relationship between meridian and collateral homeostasis and health and disease, thereby providing ideas and references for the identification of pre-disease states as well as precise diagnosis and treatment in TCM.
