2.Establishment of a cisplatin-induced multidrug resistance cell line SK-Hep1/DDP.

BACKGROUND AND OBJECTIVEMultidrug resistance (MDR) is a major obstacle in the chemotherapy of cancer patients. The aim of this study was to establish a mutidrug-resistant cell line SK-Hep1/DDP and explore its molecular mechanism of the MDR.
METHODSSK-Hep1/DDP cell line was induced by pulse treatment using a high concentration of cisplatin (DDP) in vitro. The chemoresistance indexes of cells were evaluated by CCK-8 assays. The protein of MDR1 (ABCB1), MRP1 (ABCC1), MRP2 (ABCC2) and Bax were detected by Western blotting, and the effect of MDR1 inhibitor cyclosporine A (CsA) on expression of MDR1 proteins in SK-Hep1 and SK-Hep1/DDP cell lines. Flow cytometry was performed to determine the distribution of the cell cycle and cell apoptosis ratio.
RESULTSThe SK-Hep1/DDP cells were 13.76 times more resistant to DDP in comparison with SK-Hep1 cells, and SK-Hep1/DDP cells also exhibited cross-resistance to many other chemotherapeutic agents (adramycin and 5-fuorouracil). MDR1, MRP1, and MRP2 protein expressions were significantly higher in the SK-Hep1/DDP than in the SK-Hep1 (P < 0. 01), but Bax was lower in the SK-Hep1/DDP than in the SK-Hep1(P < 0. 01). There was no obvious influence between SK-Hep1 and SK-Hep1/DDP cells in the expression of MDR1 by MDR1 inhibtor CsA (P > 0. 05). The percentages of cells in G(2)/M and S phase were significantly increased in SK-Hep1/DDP in comparison with those in SK-Hep1 [(20.67 +/- 5.69)% vs. (12.14 +/- 3.36)%; (42.20 +/- 2.65)% vs. (27.91 +/- 2.16)%; P < 0. 01]. After the cells were exposed to 10 μg/mL DDP for 24 h, the cell apoptosis rate of SK-Hep1/DDP was decreased in comparison with SK-Hep1, but it was increased in those with pretreatment of MDR1 inhibitor CsA as compared with those without pretreatment.
CONCLUSIONSA reliable multi-drug resistant human hepatoma cell line SK-Hep1/DDP is successfully established. The MDR mechanisms of this cell lines are closely related to the over-expression of MDR1 MRP1 and MRP2, lower expression of Bax and the attenuated cell apoptosis induced by chemotherapeutic agents.

5.Current attitudes and knowledge about suicide in community members: a qualitative study.

OBJECTIVEUnderstand the public's current attitudes and knowledge about suicide and, thus, provide essential information to the development of targeted public education programs-important components of the suicide prevention effort.
METHODSSeventeen mental health professionals who were extensively trained in the methods of conducting focus groups used a pre-tested focus group outline on attitudes and knowledge about suicide to conduct 101 focus groups and 18 individual in-depth interviews with a total of 842 community respondents from 6 regions in northern China. The focus groups and in-depth interviews were audio-taped, transcribed and analyzed using the QSR Nvivo text analysis software.
RESULTSMost respondents believed that suicide was a greater problem in rural areas and among women and identified physical illnesses, economic problems and interpersonal conflicts (particularly family conflicts) as the main causes of suicide. Rural residents and women were believed to exhibit impulsive suicidal behavior because of their personal limitations and over-sensitiveness. Most thought that suicide was understandable and a small proportion felt that it was acceptable behavior in certain circumstances. Almost all felt that suicide resulted in the stigmatization and a loss of 'face' for the family. Most believed that one should show concern for persons who have suicidal behavior and their family members and expressed a willingness to have superficial social relationships with them but were unwilling to establish close personal relations with them. The vast majority believed that suicide was either very difficult or impossible to prevent.
CONCLUSIONSIn China the community is tolerant, sympathetic and, in some cases, accepting of suicide but there remains a substantial underlying stigmatization of suicide. Community members have some misunderstandings about suicide; the most obvious misunderstanding is the underestimation of the importance of mental illness as a cause of suicide. The content of public health messages used in suicide prevention programs should be developed by combining findings from both qualitative and quantitative research.

6.Relationship between mitochondrial DNA instability and interleukin-8 activity in gastric mucosa.

OBJECTIVETo evaluate the relationship between mitochondrial DNA instability (mtMSI) and interleukin-8 (IL-8) activity in gastric mucosa of various lesions.
METHODSIL-8 level in gastric mucosa was assayed using ELISA method. The mtMSI was detected by PCR-SSCP techniques.
RESULTSmtMSI was observed in 11 out of 30 (36.7%) gastric cancers, 2 of 15 (13.3%) intestinal metaplasia, 2 of 10 dysplasia and 1 of 10 chronic atrophic gastritis. IL-8 level in mtMSI+ group [(76.8 +/- 3.8) pg/mg] was significantly higher than that in mtMSI- group [(48.3 +/- 3.6) pg/mg, P < 0.05].
CONCLUSIONmtMSI closely correlates with IL-8 level in gastric mucosa and is involved in gastric carcinogenesis.

7.Clinical value and safety of total laparoscopic gastro-duodenal triangle anastomosis

Objectives:To analyze the clinical value and safety of total laparoscopic gastro-duodenal triangle anastomosis in radical surgery of distal gastric cancer.Methods:Sixty patients with gastric cancer treated from May 2013 to August 2016 were selected as study subjects and randomly divided into experimental group and control group,with 30 cases in each group.The experimental group was given laparoscopic radical gastrectomy for gastro-duodenal anastomosis,while the control group was given laparoscopic auxiliary distal gastrectomy for radical gastrectomy.The clinical efficacy and safety of the two groups were compared.Results:Compared with the control group,the operation time of the experimental group was longer,the blood loss was less,the distal margin of the tumor was longer,the postoperative pain score was lower and the dosage of analgesics was less (P<0.05);There was no significant difference in TNM stage and Lauren classification between the two groups(P>0.05).The incidence of postoperative complications in the experimental group was 6.67%,which was lower than that in the control group(10.0%),but the difference was no Statistical significance (P>0.05).Conclusion:Total laparoscopic distal gastric cancer Gastro-duodenal triangle anastomosis is effective and safe,and worthy of clinical application.

OBJECTIVETo investigate the effect of different magnitudes of tensile strain on human osteoblasts differentiation.
METHODSAccording to the strain amplification mechanism at cellular level and a data calculated by finite element analysis, the cellular level strain of 0.8%, 1.6%, 2.4% and 3.2% was respectively applied to human osteoblasts for 48 h at a frequency of 1 Hz. Alkaline phosphatase activity and the expression of osteoblast-related genes were detected by Semi-Quantitative RT-PCR and densitometric analysis.
RESULTSAlkaline phosphatase activity significantly increased at 0.8% and 1.6%. The level of osteocalcin mRNA increased at 2.4% and 3.2%. Cbfa1/Runx2 gene expression only increased at 3.2%. Comparing to static control, mRNA level of type I collagen increased at every magnitude. The mRNA level decreased at 0.8% and increased at 3.2% when compared to the group with 1.6% elongation.
CONCLUSIONSHigher magnitudes of strain enhance expression of osteocalcin, type I collagen gene and Cbfa1/Runx2 in human osteoblasts, but lost the ability to increase ALP activity which is remained by lower magnitudes of strain. Type I collagen gene expression increases in a strain magnitude dependent manner.

Objective:To screen and identify the protein that interacts with the adhesion protein of Mycoplasma genitalium (MgPa)from T7-phage display cDNA library of human uroepithelial cells(SV-HUC-1).Methods:Recombinant adhesion protein of My-coplasma genitalium(rMgPa)was used as target molecule to biopan the T7 phage display cDNA library of SV-HUC-1 cell,the selected positive clones were analysed using DNA sequencing and BLAST analysis and identified by means of indirect ELISA,Dot immunoblot and Far-western blot.Results:After four rounds of biopanning,positive phages were obviously enriched.According to the results of DNA sequencing and BLAST analysis,the selected randomly 32 positive clones included 7 kinds different sequences,of which the number of RPL35 repeats was the most.The results of indirect ELISA,Dot immunoblot and Far-western blot showed that 7 representative phages could bind specifically with rMgPa.Conclusion:60S ribosomal protein L35(RPL35) may be the interacting protein of MgPa,which lays the experimental foundation for understanding the function of MgPa and the pathogenesis of Mycoplasma genitalium.
