Objective ATRA can restrain proliferation and promote differentiation in various tumor cells. The aim of the study is to investigate the differentiation characteristics of K562 induced by ATRA. Methods Morphology (Benzidine staining, Wright's staining, NSE staining and NBT recovery test) and flow cytometry were used to observe the differentiation characteristics of K562 after co-incubation with 1 μmol/L and 2.5 μmol/L ATRA for 1 d, 4 ds, and 5 ds. Results Co-incubated for 4 ds, 61.5% K562 cells in the 1 μmol/L ATRA group and 39% K562 cells in the 2.5 μmol/L ATRA group showed some evidence of myeloid maturation, but no evidence of erythroid or monocytoid maturation. Co-incubated for 5 ds, the percentage of differentiated K562 cells was much higher than that in the control group. One day after induction by 1 μmol/L ATRA or 2.5 μmol/L ATRA, the expression of CD13 was 8.0% and 6.7%, respectively, which was higher than that in the control group (2.1%). Five days after induction by 1 μmol/L ATRA or 2.5 μmol/L ATRA, the expression of CD13 increased to 28.1% and 37.8%, respectively, while the expression of CD71 decreased to 1.2% and 0.9% respectively. The differences between the ATRA groups and the control group were significant (P<0.05). CD71 decreased from 9.7% and 10.8% in the 1 μmol/L and 2.5 μmol/L ATRA groups on day 1 to 1.2% and 0.9% on day 5, while the CD13 expression level increased from 8.0% and 6.7% to 28.1% and 37.8%, respectively. Conclusions ATRA can induce K562 to differentiate into myeloid linage.

To understand the maintenance and transmission of SFTS virus, the potential vector ticks were collected from sheep, cattle and dogs in the endemic areas of SFTSV in Shandong Province. Among the collected ticks, the dominant species was H. longicornis ticks. Real-time PCR for RNA detection, virus isolation and characterization, genomic sequencing, phylogenetic and antigenic analysis were performed in this investigation. The results showed that the SFTS viral RNA was detected in 2.14% H. longicornis, and a SFTS virus was isolated from one of viral RNA positive ticks collected from sheep. Whole genome analysis of the SFTSV isolates with 11 human-origin SFTS virus revealed a highly pairwise similarity, and the growth curve analysis showed nearly identical in virus yield and the dynamic of virus reproduction compared to human derived viral isolates. Immunofluorescence and neutralization test showed identical serological reaction character of the two different origin viral strains. In this study, the characters of a SFTSV isolate was firstly described, which suggested that the tick species H. longicornis acting important vector role in the transmission of SFTS virus.

OBJECTIVETo explore an approach to rapidly and accurately identify the compounds as biomarkers of Chinese medicine (CM) syndromes.
METHODThe Fourier transform infrared (FT-IR) spectrometry was applied to investigate the characteristic components of a mice model of Kidney (Shen)-yang deficiency syndrome (KDS), and the remedial effect of a typical CM formula Shenqi Pill (). Thirty-six females and 18 males of Balb/c mice were randomly divided into KDS, Shenqi or control group. The females and males of the same group freely were mated for 96 h, and the males were taken out and only the female mice were raised. Females of the KDS group were threatened by a ferocious cat every other day for 14 d. After delivery, the KDS, or gestational threatened, offspring were raised at standard condition for 11 weeks. Then 10 male offspring were randomly selected, anaesthetized and their representative organs, i.e. testes, kidneys, lungs and feet were collected, for the FT-IR scan. Mice of the Shenqi group were intragastric administered Shenqi Pill; while mice in the KDS and control groups were given the same volume of saline.
RESULTSThe attenuated birth outcomes of the KDS group were displayed. The remarkable FT-IR differences of all organs between KDS mice and healthy control were mainly at 1,735-1,745 cm(-1) (indicating the increased levels of lipids) and at 1,640-1,647 cm(-1) and 1,539-1,544 cm(-1) (displaying the decreased proteins). No statistic FT-IR difference between Shenqi and control mice was observed.
CONCLUSIONIn accordance with major traits of KDS, prenatal stress extensively impaired the building up of proteins and resulting in the excessive lipid storage, and FT-IR could effectively identify the biomarkers of KDS.

Objective To observe the expression of extracellular matrix metalloproteinase inducer (EMMPRIN) in the unstable plaque of patients with acute coronary syndrome (ACS),and the impact of leukotriene B4 (LTB4) on the EMMPRIN expression in macrophages. Methods The EMMPRIN expression was detected by immunohistochemistry in 11 unstable plaques from patients with ACS.Protein expression of EMMPRIN was evaluated by Western blot on macrophages differentiated from THP-1 which were stimulated with LTB4 in the absence or presence of LTB4 antagonist U75302.There are 8 study groups:1-THP-1,2 -8-the macrophages derived from THP-1,2 - 6- macrophages were stimulated by LTB4 (0,10-10,10-9,10-8 and 10-7mol/L) for 24 h,7 -8-the macrophages were pretreated by 10-6 mol/L or 10-7 mol/L U75302 2 h before the LTB4 ( 10-7 mol/L) stimulation. Results Abundant EMMPRIN expression was detected in macrophages and smooth muscle cells of unstable plaques from ACS patients. As to the THP-1 derived macrophages,EMMPRIN expression was significantly upregulated in a concentration-dependent manner in LTB4 stimulated groups, which was significantly higher in group 3 -6 than in the THP-1 group ( group 1 ) and macrophages group ( group 2 ) ( all P < 0.05 ) and pretreatment with U75302 significantly reduced the LTB4 induced upregulation of EMMPRIN in a dose-dependent manner (P < 0.05).Conclusion EMMPRIN expression is enhanced in macrophages and smooth muscle cells on unstable coronary artery plaques from ACS patients.LTB4 could stimulate EMMPRIN expression on THP-1 derived macrophages suggesting that LTB4 and EMMPRIN might be both involved in the formation and progression of unstable plaques, future studies are warranted to explore if LTB4 and EMMPRIN antagonists are effective or not for treating patients with ACS.

【Objective】To investigate the expression of FosB in invasive breast cancer and its correlation to the ultrasound findings and clinical significance.【Methods】Immunohistochemical and ultrasound examination were performed for 155 cases with invasive breast cancer,36 cases with paracancerous tissue and 30 cases with normal breast tissue. To investigate the correlations between the different expression of FosB with histological grade ,lymph nodes, and ultrasound findings.【Results】①The expression of FosB in paracancerous tissue and normal breast tissue was higher than that in invasive breast cancer(P<0.01). ②Based on immunohistochemical staining, high positive expression rate of FosB was associated with low histological grade,ER positive and PR positive(P<0.05). There was no correlation between FosB and age, tumor diameter, lymph nodes,Her2 and clinical stages(P>0.05)③The expression of FosB was higher in microcalcification positive and lymph nodes bigger tissues(P<0.05). There was no correlation between FosB and tumor size, regular shape, boundary, posterior attenuation ratio and blood flow(P>0.05).【Conclusion】The expression of FosB in invasive breast cancer was lower than that in paracancerous tissue and normal breast tissue. The different expression of FosB was correlated with histological grade,ER positive and PR positive. To study the expression of FosB in invasive breast cancer may be helpful for differential diagnosis and targeted treatment. The different expression of FosB was correlated with microcalcification, lymph nodes, and combined FosB with ultrasound findings may contribute to prognostic evaluation of breast cancer.
