1.Effect of ATRA on the expression of genes Hoxb2 and Hoxb4 in cord blood erythroid progenitors.

This study was aimed to investigate the expressions of genes hoxb2 and hoxb4 after interference of the proliferation and differentiation of hematopoietic stem cells (HSC) to the erythroid progenitors (CFU-E) in vitro by using all-trans retinoic acid (ATRA). The cord blood was collected from 12 cases of fetal placenta umbilical vein and cultured by using culture technique of HSC in vitro. The proliferation and differentiation of HSC to CFU-E were interfered with 6 x 10(-8) mol/L of ATRA. The expression levels of genes hoxb2 and hoxb4 in blank control and ATRA groups were detected by FQ-RT-PCR on day 3, 7 and 10 of culture. The results showed that the expressions of genes Hoxb2 and hoxb4 were a little on day 3, obviously increased on day 7 and reached highest level on day 10 in 2 groups. The expression level of hoxb4 on day 3, 7 and 10 in blank control group was obviously higher than expression level of hoxb2. As compared with blank control group, the expressions of genes hoxb2 and hoxb4 in the ATRA group were significantly up-regulated. It is concluded that the genes hoxb2 and hoxb4 all expressed in process of proliferation and differentiation to erythroid progenitors, which suggests that hoxb2 and hoxb4 relate to erythroid hematopoiesis, and the hoxb4 has more great relevance to erythroid hematopoiesis as compared with hoxb2. The ATRA (6 x 10(-8) mol/L) can up-regulate the expression of hoxb2 and hoxb4 significantly.

2.Epidemic trend of hepatitis B in 18 ethnic minorities of Yunnan Province from 2009 to 2018

Objective To analyze the epidemic trend of hepatitis B virus (HBV) in 18 minority nationalities in Yunnan Province from 2009 to 2018, so as to explore the ethnic differences in the incidence of HBV in Yunnan Province. Methods Based on the reported incidence data of hepatitis B in China's disease prevention and control information system from 2009 to 2018, descriptive epidemiology method was used to describe and analyze the incidence of hepatitis B in different ethnic groups, and K-means clustering method was used to explore and analyze the annual average incidence of hepatitis B in different ethnic groups. Results From 2009 to 2018, the average incidence of hepatitis B in Yunnan Province was 44.26/100 000, which was much lower than the overall level of China every year; the average incidence of hepatitis B in ethnic groups was 41.27/100 000, slightly lower than the overall level of Yunnan every year. The prevalence of hepatitis B was different in different ethnic groups. The average incidence of Wa was significantly higher than others (95.26/100 000), and Jingpo was the lowest (22.51/100 000). According to the incidence of hepatitis B, different ethnic groups were divided into three categories: high incidence ethnic group, middle incidence ethnic group and low incidence ethnic group. Conclusion There are ethnic differences in the incidence of hepatitis B in Yunnan Province. The incidence of hepatitis B in some ethnic groups is higher than that in the whole country all the year round, which is the key population in the prevention and control of hepatitis B.

OBJECTIVETo study organochlorine pesticide and heavy metal residuals of Chrysanthemum morifolium.
METHODThe contents of organochlorine pesticide residuals was determined by chromatography, Cu, Pb, Cd was determined by AAS, and As, Hg was determined by AFS.
RESULT AND CONCLUSIONThe contents of organochlorine pesticide and heavy metals residuals are up to "Green Trade Standards of Importing & Exporting Medicinal plants & Preprations" (MOFTEC, 2001), but the contents are different among four cultivars clearly.

5.Research on human life signal separation and reconstruction method based on optimized variational modal decomposition

Objective To propose a human life signal separation and reconstruction method based on optimized variational mode decomposition(VMD)to improve the accuracy and timeliness of the life detection radar in extracting and separating human life signals such as heartbeat and respiration.Methods Firstly,the particle swarm optimization algorithm was used to optimize the parameters of VMD,and the human life signal was decomposed into a series of intrinsic mode functions(IMFs);secondly,the alignment entropy of each IMF was calculated,the noise was removed based on the alignment entropy threshold,and the remaining components were reconstructed to form human life signals;finally,the method proposed was compared with infinite impulse response(IIR)filtering,VMD and complete ensemble empirical mode decomposition with adaptive nosie(CEEMDAN)to verify its performance.Results Under different noise levels the proposed method outperformed IIR filtering,VMD and CEEMDAN in evaluation metrics of signal-to-noise ratio and root-mean-square error,and behaved better than CEEMDAN in terms of computational time-consumption.Conclusion The proposed method realizes rapid separation and reconstruction of vital signals such as heartbeat and respiration while effectively filtering out the noise,which has broad application prospects in the fields of non-contact vital signs detection of bum/scald patients,infectious disease patients and newborns and the search and rescue of buried casualties after a disaster.[Chinese Medical Equipment Journal,2024,45(3):9-15]
6.Analysis of relationship between sexual concept and psychological health among college famale students majored in nursing

Objective To esplore the relationship between sexual concept and psychological health among college famale students majored in nursing and provide scientific evidence for adolescent sex education. Methods 1 093 college famale students majored in nursing were investigated with sexual concept questionnnaire and SCL-90. Results 35.1% subjects were in love, 38.2% agreed with sexual behavior before marriage, 9.7% agreed with extramarital sex. The score of all factors in Scl-90 were higher than the norm. The results showed the positive relationship between paranoid ideation and loving behavior, between hostility and paranoid ideation and premarital sex, between paranoid ideation and psychosis and extramarital sex, and the negtive relationship between interpersonal relationship and extramarital sex. Conclusions We should not ignore the effects of psychological factors on college famale students majored in nursing when carrying out the sexual education, especially for the students with higher paranoid ideation, hostility and psychosis who will be needed other education.
7.Transplantation of neural stem cells overexpressing glial cell line-derived neurotrophic factor enhances Akt and Erk1/2 signaling and neurogenesis in rats after stroke.

BACKGROUNDOur previous studies have indicated that the beneficial effects of grafting neural stem cells (NSCs) overexpressing glial cell line-derived neurotrophic factor (GDNF) in rats after stroke. However, the underlying mechanisms are highly debatable. In this study, we investigated whether neurogenesis, Akt, and extracellular signal-regulated kinase 1/2 (Erk1/2) signaling were involved in this process.
METHODSTransient ischemic stroke were induced by occluding middle cerebral artery for 2 hours and reperfusion. At 3 days after reperfusion, GDNF/NSCs, NSCs, and vehicle were administered. Immunohistochemical staining was used to evaluate neurogenesis by nestin antibody; phosphorylation of Akt and Erk1/2 was investigated by Western blotting analysis.
RESULTSTransplantation of GDNF/NSCs and NSCs significantly increased nestin-positive cells compared to control group (vehicle) from 1 to 7 weeks after reperfusion, and GDNF/NSCs showed stronger effect than NSCs at 2 and 3 weeks after reperfusion. Meanwhile, enhanced phosphorylation level of Erk1/2 was observed in the GDNF/NSCs and NSCs groups compared with control group, and phosphorylation level of Erk1/2 in GDNF/NSCs group was remarkably higher than that of NSCs group at any given time. In contrast, expression of mitogen-activated protein kinase phosphatase-1 (MKP-1), known as inhibitor of Erk1/2 signaling, was significantly decreased in the GDNF/NSCs and NSCs groups compared with the control group. Moreover, much enhanced and prolonged phosphorylation level of Akt of GDNF/NSCs group was detected compared with control and NSCs group.
CONCLUSIONGrafting GDNF/NSCs enhances neurogenesis and activates Akt and Erk1/2 signaling, that may provide the potential for GDNF/NSCs in stroke treatment.

8.The Clinical Efficacy of Danlou Tablet in Non-revascularization Patients With Acute Coronary Syndrome and Phlegm and Blood Stasis Syndrome Type

OBJECTIVE To investigate the clinical efficacy of Danlou tablet in Non-revascularization Patients with acute cor-onary syndrome(ACS)and phlegm and blood stasis syndrome type(PBSST).METHODS 70 ACS patients with PBSST for various reasons failed revascularization therapy were randomly assigned to the control group and the treatment group.The con-trol group was given a standardized western medicine treatment and the treatment group was given Danlou tablets (every time 1.5g,3 times a day)based on standardized western medicine treatment for a month.Angina episodes(intensity,frequency, duration),nitroglycerin use (consumption,angina relief time after taking nitroglycerin ) and the score of angina,traditional Chinese Medicine(TCM)clinical syndrome (including:chest pain or tightness and its frequency and duration,palpitations, shortness of breath,fatigue and total syndrome score),lipids including total cholesterol(TC),low-density lipoprotein choles-terol(LDL-C),high density lipoprotein cholesterol(HDL-C),triglycerides(TG)and hypersensitive C-reactive protein(hs-CRP) were observed in the two groups of patients before and after treatment.The clinical efficacy of angina,the score of TCM clinical syndrome and blood biochemical changes were recorded and compared between the two groups.RESULT The total ef-fective rate of angina(85.29%)was significantly better than that of the control group(48.48%)(P<0.01).Compared with the same group before treatment,the frequency and duration of angina,the angina relief time after taking nitroglycerin,the a-mount of nitroglycerin,the score of angina and TCM clinical syndromes and the total score of syndrome,all these were signifi-cantly lower(P<0.01)in the two groups of patients after treatment.Compared with the control group,the frequency and du-ration of angina,the angina relief time after taking nitroglycerin,the amount of nitroglycerin,the total score of TCM clinical syndrome(P<0.05)and the score of angina(P<0.01)were significantly lower in the treatment group after treatment.The improvement of TCM clinical syndromes in the treatment group(the total efficiency of syndrome score was 79.41%)was sig-nificantly better than the control group (the total efficiency of syndrome score was 45.45%)(P<0.01).TC,LDL-C(P<0.01)and hs-CRP(P<0.05) levels after treatment were significantly lower than before.After treatment,hs-CRP in the treatment group was significantly lower compared with the control group(P<0.05).CONCLUSION Danlou tablet can pro-mote a significant improvement to angina and TCM clinical syndromes in Non-revascularization Patients with ACS and PBSST, inhibit the inflammation,and show plaque stabilizing effects.
9.Comparison on efficacy and safety between domestic levosimendan versus dobutamine for patients with acute decompensated heart failure

Objective To compare the efficacy and safety of domestic levosimendan versus dobutamine for patients with acute decompensated heart failure (ADHF).Methods ADHF patients from 8 medical centers were recruited in this multicenter,blind,positive-controlled,randomized study and received 24 h intravenous levosimendan (n =114) or dobutamine (n =114) therapy.SWAN-GANZ catheter was performed in patients with pulmonary capillary wedge pressure (PCWP) ≥ 15 mm Hg ( 1 mm Hg =0.133kPa) and cardiac index (CI) ≤ 2.5 L · min-1 · m-2 ( n =39 each).Results Compared with baseline level,LVEFincreased [(31.56 ±9.69)% vs.(28.44 ±7.08)%,P<0.01] at 24 h in both groups.LVEF increase at 24 h was similar between two groups [ ( 3.11 ± 6.90 ) % vs.( 3.00 ± 6.63 ) %,P >0.05].The PCWP decrease at 24 h was significantly greater in levosimendan group than in dobutamine group [ ( - 8.90 ± 7.14) mm Hg vs.( - 5.64 ± 6.83) mm Hg,P =0.04 ].Decrease in NT-proBNP at 3 days was also more significant in levosimendan group than in dobutamine group [ the percentage change compared to baseline:( - 22.36 ± 38.98) % vs.( - 8.56 ± 42.42) %,P < 0.01 ].Dyspnea improvement at 24 h was more significant in levosimendan group than in dobutamine group.The incidences of adverse reactions and events were similar between two groups.Conclusion LVEF improvement is similar between dobutamine and domestic levosimendan while greater decreases in PCWP and NT-proBNP are achieved with domestic levosimendan in patients with ADHF.

OBJECTIVETo study the value of immunomarkers CXCL13, CD10, bcl-6 in pathologic diagnosis of angioimmunoblastic T-cell lymphoma (AITL).
METHODSOne hundred and fifteen cases of AITL, 30 cases of peripheral T-cell lymphoma, not otherwise specified (PTCL, NOS) and 30 cases of reactive lymph nodes with paracortical hyperplasia (RH) encountered during the period from January, 1990 to January, 2008 were retrieved from the archival files of the Department of Pathology, West China Hospital of Sichuan University, China. The morphologic features were reviewed and compared. Immunohistochemical study was performed by SP method for CXCL13, CD10, bcl-6, CD21, CD3epsilon, CD3, CD45RO, CD20 and Ki-67. TCR-gamma gene rearrangement study was also carried out.
RESULTSRegressed follicles were evident in 7.8% (9/115) of AITL cases, 6.7% (2/30) of PTCL, NOS cases and 83.3% (25/30) of RH cases, respectively. A marked increase of number of arborizing venules was shown in 98.3% (113/115) of AITL cases, 63.3% (19/30) of PTCL, NOS cases and 76.7% (23/30) of RH cases, respectively. In lymph nodes with paracortical hyperplasia, the expression of CXCL13, CD10 and bcl-6 were restricted to the germinal centers. In AITL, 96.5% (111/115) of cases showed CXCL13 expression, in contrast to 26.7% (8/30) of PTCL, NOS. Expression of CD10 and bcl-6 were found in the neoplastic cells in 50.4% (58/115) and 78.3% (90/115) of AITL, and 3.3% (1/30) and 3.3% (1/30) of PTCL, NOS, respectively. Irregular meshworks of CD21-positive follicular dendritic cells were found in all the AITL cases. Clonal TCR-gamma rearrangement was detected in 83% (83/100) of the AITL cases.
CONCLUSIONSAITL is a type of lymphoma originated from the follicular helper T cells. Detailed morphologic assessment and use of immunohistochemical markers are essential for accurate diagnosis.
