1.Study on correlation between aquaporin 3, 4 gene expression in gastric mucosa and severity of Pi-Wei damp-heat syndrome in patients with chronic superficial gastritis.

OBJECTIVETo explore the relationship between aquaporin 3,4 (AQP3, AQP4) gene expression in gastric mucosa and severity of Pi-Wei damp-heat syndrome (PWDHS) in patients with chronic superficial gastritis (CSG).
METHODSGastric mucosa taken from the upper part of gastric corpus was collected under gastroscope and preserved in liquid nitrogen. The gene expression of AQP3 and AQP4 was determined quantitatively by fluorescent PCR.
RESULTSThe gene expression of AQP3 and AQP4 in patients with PWDHS of moderate and severe degree was higher than that in those of mild degree and in healthy persons respectively (P <0.05 and P <0.01); and the gene expression of AQP3 in patients with PWDHS of severe degree was higher than that in those of moderate degree (P<0.05).
CONCLUSIONThe gene expression of AQP3 and AQP4 in gastric mucosa was correlative with the severity of PWDHS in patients with chronic superficial gastritis, the severer the syndrome, the higher the gene expression.

3.Protective effects of imperatorin against cerebral ischemia/reperfusion-induced oxidative stress through Nrf2 signaling pathway in rats

OBJECTIVE To investigates the effects of imperatorin on the oxidative stress in the cerebral cortex and hippocampus after focal cerebral ischemia/reperfusion injury. METHODS Transient focal cerebral ischemia/reperfusion model in male Sprague-Dawley rats was induced by 2 h middle cerebral artery occlusion followed by 24 h reperfusion. Imperatorin (1.25 and 2.5 mg·kg- 1) or vehicle were administered intraperitoneally at 1, 5 and 9 h after the onset of ischemia. At 24 h after reperfusion, the biomarkers of oxidative stress such as the levels of reactive oxygen species (ROS), lipid peroxidation products malondialdehyde (MDA), nitric oxide (NO) and total antioxidant capacity (T-AOC), the activities of inducible nitric oxide synthase (iNOS), superoxide dismutase (SOD) and catalase (CAT) in the cerebral cortex and hippocampus were observed. We also assessed the nuclear factor erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and the NAD(P)H-quinone oxidoreductase 1 (NQO-1) protein expression by Western blot. RESULTS As compared to vehicle-treated animals, imperatorin treatment significantly reduced the ROS, MDA, NO levels and iNOS activity, increased T-AOC and the activities of SOD and CAT. Furthermore, imperatorin treatment also significantly induced the nuclear translocation of Nrf2, enhanced the protein expression of HO-1 and NQO-1 in the cerebral cortex and hippocampus. CONCLUSION Our findings indicate that imperatorin can protect the brain against the excessive oxidative stress induced by cerebral ischemia/reperfusion through activation of Nrf2 signaling pathway.
4.The alterations of nitric oxide synthase activity of ventricular cardiac muscle of rats in two septic shock models.

AIMTo observe the differences of hemodynamics and nitric oxide synthase(NOS) activity of ventricular cardiac muscle in two septic shock models and explore the possible mechanism.
METHODSTwo rat models of septic shock[lipopolysaccharide(LPS)-induced and cecal ligation and puncture (CLP)-induced septic shock] were used. The hemodynamic parameters and nitric oxide synthase activity of ventricular cardiac muscle were measured.
RESULTSThe hemodynamic parameters in CLP-induced model were increased in the early stage and decreased in the late stage while in LPS-induced model the parameters showed the same change of the CLP late stage. Both LPS model and CLP model (late stage) showed significant increase in NOS activity, but there was no difference between the two models. After treatment of the NOS inhibitor N-nitro-L-arginine methyl ester (L-NAME), the parameters of CLP-late stage and LPS model increased significantly. The NOS activity reached the highest level in the CLP-middle stage. The production of nitrite/nitrate decreased significantly in LPS model and CLP model(late stage) after treatment of L-NAME, but the nitrite/nitrate produced by constitutive NOS in LPS model was higher than CLP model(late stage).
CONCLUSIONThe increase of the NOS activity may be the main reason to lead to the depression of the hemodynamic parameters. Inducible NOS may play the leading role in the LPS model while cNOS and iNOS have the same effect in the CLP model.

Objective:To improve the genetic stability of HBV gene in transgenic mice.Methods:HBV transgenic mice were bred by backcross and double cross.The HBV gene expression and replication were studied with real-time PCR,ELISA and chemiluminescence.Results:The HBV transgenic mice have stably bred to 23rd generation.The serum HBsAg level is 4122.31?2044.74IU/ml;The rate of HBV transgenic mice whose serum HBV DNA reach 104~106copies/ml was 93.93%.The HBV replication and expression were improved markedly.There is no difference between male and female mice about serum HBsAg level.Conclusion:After breeding the HBV gene was expressed stably with high-level in transgenic mice.

7.Observation on therapeutic effect of picking therapy on Graves' disease.

OBJECTIVETo observe therapeutic effect of picking therapy on Graves' disease (GD) and its effects on thyroid function.
METHODSSixty cases of GD were randomly divided into a treatment group (pricking therapy group) and a control group (medication group). They were treated respectively with pricking thyroid gland high point or oral administration of tapazole. And changes of total triiodothyronine (TT3), total thyroxine (TT4), free T3 (FT3), free T4 (FT4), thyroxine receptor antibody (TRAb), supersensitive thyrotropin (S-TSH) after treatment were investigated.
RESULTSThe total effective rate was 93.33% in the treatment group and 76.67% in the control group with a significant difference between the two groups (P < 0.05). After treatment, serum TT3, TT4, FT3, FT4 and S-TSH contents had very significant change in the two groups (P < 0.01), TRAb had very significant change in the treatment group (P < 0.01) and no significant change in the control group (P > 0.05) there were significant differences in decrease of TT3, FT3, TRAb (P < 0.05) and no significant difference in decrease of TT4, FT4 and increase of S-TSH between the two groups (P > 0.05).
CONCLUSIONPricking therapy has a definite therapeutic effect on Graves' disease, which is carried out through regulating thyroid function.

Objective To detect the mutations of ATP2C1 gene in patients with Hailey-Hailey dis- ease (HHD).Methods PCR and direct sequencing were performed in 17 patients and 120 healthy controls to screen the mutations in the exons of ATP2C1 gene.Results Eight mutations were identified in nine probands, including three deletion mutations (nt1464-1487 del/nt1462-1485del,1523delAT,2375delTTGT),three splice site mutations (360—2A→G,1415—2A→T,2243+2T→C) and two missence mutations (C920T and G1942T).None of the above mutations was found in the controls.Conclusion Eight specific novel mutations were identified in nine probands of HHD,which could be causative factors of the disease.
9.Dynamic accumulation regulation of curcumin, demethoxycurcumin and bisdemethoxyeurcumin in three strains of curcuma longae rhizome.

The paper is aimed to study the dynamic accumulation regulation of curcumin (Cur), demethoxycurcumin (DMC) and bisdemethoxyeurcumin (BDMC) in three strains of Curcuma longa, and provide scientific references for formalized cultivation, timely harvesting, quality control and breeding cultivation of C. longa. The accumulation regulation of the three curcumin derivatives was basically the same in rhizome of three strains. The relative contents decreased along with plant development growing, while the accumulation per hectare increased with plant development growing. The accumulation of curcuminoids per hectare could be taken as the assessment standard for the best harvest time of C. longa. A3 was the best strain in terms of Cur and BDMC content.

10.HPLC fingerprint of ethyl acetate extraction of Saxifraga stolonifera.

OBJECTIVETo establish an HPLC fingerprint of ethyl acetate extraction of Saxifraga stolonifera.
METHODThe HPLC analysis was performed on a Diamonsil C18 (4.6 mm x 250 mm, 5 microm) column with isocratic elution of acetonitrile-0.05% phosphoric acid at a flow rate of 1.0 mL x min(-1). The detection wavelength was set at 256 nm and the column temperature was set at 30 degrees C.
RESULTThe HPLC fingerprint of ethyl acetate extraction of S. stolonifera has been established. There were fifteen common peaks, seven of which were identified by reference substances. The RSD of relative retention time was less than 3% in the precision and repeated tests. Eleven samples from different area can be distinguished from their fingerprints.
CONCLUSIONThis method is reasonable and reliable and can be used for quality control of S. stolonifera.
