2.Advances development of 64CuCl 2 in the diagnosis and treatment of tumor and disease of abnormal copper metabolism

Copper is an essential metal element for human metabolism. Copper homeostasis can lead to inherited disorders of copper metabolism and also play an important role in tumor proliferation. In recent years, great progress has been made in the research and development of radioactive metal nuclide tracers, and molecular imaging quickly becomes a new inspection technology for detecting copper metabolism. 64CuCl 2 carries out PET imaging diagnosis of diseases with abnormal copper metabolism and tumors with high copper metabolism through real-time tracking of changes of copper ions in the body. At the same time, β - decay and electron capture of 64CuCl 2 have a therapeutic effect, and can be directly used for tumor nuclide therapy. Therefore, 64CuCl 2, as an integrated radiopharmaceutical for diagnosis and treatment, is the current research focus of molecular imaging. This article reviews 64CuCl 2 preparation, biological metabolism and its application in the diagnosis and treatment of abnormal copper metabolism diseases and tumors.

4.Research on UKF control of epileptic-form spikes in neural mass models.

Neural mass models are able to produce epileptic electroencephalogram (EEG) signals in different stages of seizures. The models play important roles in studying the mechanism analysis and control of epileptic seizures. In this study, the closed-loop feedback control was used to suppress the epileptic-form spikes in the neural mass models. It was expected to provide certain theory basis for the choice of stimulus position and parameter in the clinical treatment. With the influence of measurement noise taken into account, an unscented Kalman filter (UKF) was added to the feedback loop to estimate the system state and an UKF controller was constructed via the estimated state. The control action was imposed on the hyper-excitable population and all populations respectively in simulations. It was shown that both UKF control schemes suppressed the epileptic-form spikes in the model. However, the control energy needed in the latter scheme was less than that needed in the former one.

6.Clinical observation on the treatment of premature ventricular beats with Zaobo decoction combined with bisoprolol

Objective To observe the clinical effects of Zaobo decoction combined bisoprolol treating premature ventricular contractions.Methods 108 patients of ventricular premature beat were recruited intoa control group and an observation group (n=54) according to the random number table method.The control group was treated with bisoprolol, while the observation group was treated with Zaobo decoction on the basis of the control group, and both groups were treated for 8 weeks.24 h dynamic electrocardiogram (ECG), renin activity plasma (PRA), angiotensin Ⅱ (angiotensionⅡ) and ALD (Ang) were observed before and after treatment.The clinical effects were evaluated.Results The total effective rate showed significant difference between the observation group and the control group (75.9% vs.57.4%;x2=4.167, P=0.041) after the treatment.After treatment, Ang-Ⅱ (56.22 ± 12.7 pg/ml vs.68.45 ± 12.7 pg/ml, t=5.004) in the observation group was significantly lower than the control group (P<0.01);24 h sinus RR interval standard deviation (129.16 ± 28.56 ms vs.116.13 ± 17.38 ms, t=2.864), every 5 min sinus RR interval mean standard deviation within 24 h (123.57 ± 25.24 ms vs.112.46 ± 18.23 ms, t=2.622), and within 24 h of sinus RR interval difference rms (31.76 ± 11.42 ms vs.22.64 ± 10.32 ms, t=4.354) in the observation group were significantly higher than the control group (P<0.01).Conclusion Zaobo decoction combined with bisoprolol can effectively improve heart rate variability, regulate rernin vascular angiotensin system, and improve the clinical efficacy of the patients with ventricular premature beat.
7.Investigation for the Mechanism of Vascular Smooth Muscle Cell Calcification Induced by Calcium and Phosphorus in Experimental Rats

Objective: To explore the effect of oxidative stress injury on the mechanism of vascular smooth muscle cell (VSMC) calciifcation induced by calcium and phosphorus in experimental rats.
Methods: The VSMC calcification was induced by incubating the cells with calcium chloride (CaCl2) andβ-sodium glycerophosphate (β-GP) for 8 days, and the cells were divided into 4 groups: ① Control group, ② Calcification group,③ Calciifcation+H2O2 group, ④ Calciifcation+catalase group. The calcium nodule formation and calcium deposition in VSMC were detected by Alizarin red staining and o-cresolphthalein complexone method, the reactive oxygen species (ROS) was detected by DCFH-DA probe staining and the protein expression of Runx2 was examined by Western blot analysis.
Results: Compared with Control group, Calciifcation group showed the higher ROS production, more calcium nodule and calcium deposition, higher Runx2 protein expression;while compared with Calciifcation group, the above indexes were even higher in Calciifcation+H2O2 group, P<0.05. The ROS production, calcium nodule, calcium deposition and Runx2 protein expression were lower in Calciifcation+catalase group than those in Calciifcation group and Calciifcation+H2O2 group, but still higher than that in Control group. The protein expression of Runx2 was similar between Calciifcation+catalase group and Control group, P>0.05.
Conclusion: CaCl2 andβ-GP treatment may induce VSMC calciifcation via activating ROS-Runx2 signal pathway in experimental rats.
8.Effect of Vitamin K2 on Theaortic Artery Calcification in Experimental Rats

Objective: To explore the effects of Vitamin K2 (VK2) on theaortic artery calciifcation and oxidative stress injury in experimental rats.
Methods: A total of 24 rats were divided into 4 groups:①Control group,②6-week calciifcation group,③12-week calciifcation group and④6-week calciifcation + 6-week VK2 group;n=6 in each group. The arterial calciifcation was induced by warfarin (WFN) treatment. The calcium nodule and deposition in rat’s theaortic artery were detected by Alizarin red staining and o-cresolphthalein complexone method, the reactive oxygen species (ROS) were measured by DHE probe staining and the morphological changes of mitochondria in smooth muscle cells were detected by transmission electron microscopy.
Results: Calciifcation nodule formed in both 6-week and 12-week calciifcation groups, the calciifcation deposition and ROS were higher than Control group,P<0.01. Compared with both calcification groups, the above indexes were decreased in 6-week calciifcation + 6-week VK2 group,P<0.01. Both calciifcation groups showed mitochondria swelling with unclear structure and cytoplasm vacuoles degeneration in vascular smooth muscle cells. The vascular smooth muscle cell volumes were similar between Control group and 6-week calcification + 6-week VK2 group, and no cytoplasm vacuoles degeneration was observed.
Conclusion: Warfarin induced aortic calciifcation is related to oxidative stress injury which may cause the ultra-micro structural damage in smooth muscle cells; VK2 may reduce the oxidative stress injury and improve the condition of vessel calciifcation in experimental rats.

10.Caffeoylquinic acid derivatives from leaves of Lonicera japonica.

OBJECTIVETo study the caffeoylquinic acid derivatives from the leaves of Lonicera japonica.
METHODThe compounds were isolated by column chromatography and identified on the basis of physico-chemical constants and spectral analysis.
RESULTFive caffeoylquinic acid derivatives were isolated,and their structures were identified as 3,4-di-O-caffeoyl quinic acid methyl ester (1), 5-O-caffeoyl quinic acid methyl ester (2), 3,4-di-O-caffeoyl quinic acid (3), 1,3-di-O-caffeoyl quinic acid (4) and chlorogenic acid (5), respectively.
CONCLUSIONCompounds 2, 4 were obtained from this genus for the first time. Compounds 1, 3 were obtained from this plant for the first time.
