2.Research of mechanism jinxueyuan granules increased saliva secretion of xerostomia model rats.

To analyze the specific mechanism of Jinxueyuan granules, the relationship between the Jinxueyuan granules increased the saliva secretion of xerostomia model SD rats and excitement of receptors were studied in this experiment. In the study, three groups of xerostomia model rats were successfully established by using M-receptor blockers-4-diphenyl-acetoxy-N-methyl-piperidine (4-DAMP) and atropine, or adrenergic receptor blocker phentolamine; after the modeling, the medicine Jinxueyuan granules were gavaged. According to the clinical dose of Jinxueyuan granules and SD rats body surface area, the rats in atropine group were divided three dose groups respectively, namely low, medium and high dose of Jinxueyuan granules groups. The 4-DAMP group and phentolamine group were gavaged medium dose of Jinxueyuan granules. And the amount of salivary secretion for 150 minutes in all groups continuously were measured, and the effect of Jinxueyuan granules increased salivation and the relationship between characteristics and the receptors were observed; and submandibular gland tissue of the rats was isolated, then the effect of Jinxueyuan Granules for expression of the water channel protein aquaporin-5 (AQP5) in submandibular gland cells was analyzed by the Western blot technology. It was found that the saliva secretion of Jinxueyuan Granules groups was increased significantly, and compared with the saline control group, phentolamine group, 4-DAMP group and atropine group, difference was significant, P < 0.05. There was no significant difference between the low-dose of Jinxueyuan granules group and the saline group, but the medium dose of Jinxueyuan granules group had a significant difference, compared with the saline group (P < 0.05). In the time distribution of increasing saliva secretion, there was a significant difference between the saline and Jinxueyuan granules group in the saliva secretion (P < 0.05). After administration of Jinxueyuan granules, the expression of AQP5 protein in the submandibular gland cells expressing of treatment groups was increased, and compared with the blocker groups, there was a significant difference, P < 0.05. Except the atropine group, there was no significant difference in Jinxueyuan granules relieving the inhibition induced by blocks in phentolamine group and 4-DAMP group, compared with the saline group. Compared the AQP5 expression in three blockers groups, there was no significant difference in the efficacy of Jinxueyuan granules between phentolamine group and 4-DAMP group; but there was a significant difference between the atropine group and other groups (P < 0.05). Therefore, it was considered that the mechanism of Jinxueyuan granules increasing saliva secretion (effectiveness of nourishing Yin and generating body fluid ) possibly through the pathway mediated by muscarinic M receptor, especially M3 receptor, or adrenergic receptor, and increased expression of salivary gland AQP5 membrane, and then stimulate saliva production.

3.Professor ZHOU Yun-xian's experience of acupuncture combined with Chinese material medica for xerophthalmia.

Professor ZHOU Yun-xian's clinical experience of acupuncture combined with Chinese material medica for xerophthalmia is introduced. Her treatment for xerophthalmia is firstly focused on syndrome differentiation, indicating that different syndrome has different treatment of acupuncture and Chinese material medica. It is emphasized that by following the holistic concept of TCM, the local acupoints and whole-body acupoints cooperate closely, forming her special combination method of acupoints. In clinic, attention is focused on the application of many acupuncture techniques and extra points. With the combination of acupuncture and Chinese material medica, the treatment is flexible with superior clinical efficacy.

4.Effect of short pulse gastric electrical stimulation on the c-fos expression in nucleus tractus solitarius and hypothalamus and its possible mechanism

Objective To investigate whether enteric nervous system(ENS)involve in mediating the effect of short pulse gastric electrical stimulation on c-fos expression in nucleus tractus solitarius (NTS)and hypothalamus.Methods Fifteen Wistar rats were divided into control group,gastric electrical stimulation(GES)group and ENS free group with 5 rats each.A pair of electrodes was implanted on the gastric serosa.In ENS free group,0.5%benzalkonii chloridum was applied on the serosa during the surgery.A set of short pulse GES with frequency of 20 cpm,pulse width of 300 μs and amplitude of 2 mA was applied via the electrodes for 30 rain in the stimulation group and ENS free group.The c-fos expression in NTS/hypothalamus was detected 1 h after the stimulation.Results The number of neuron with c-fos expression was(71.6±7.4)/HP in NTS group and(63.4±10.8)/HP in ENS free group,both were significantly higher than those in control group(36.4±8.6/HP,P<0.05).The number of neuron with c-fos expression in hypothalamus was higher in GES[(224.2±47.3)/HP]and ENS free groups[(249.1±44.0)/HP]compared with control group[(90.2±47.3)/HP,P<0.05].There was no significant difference in c-fos expression in NTS/hypothalamus between GES group and ENS free group(P>0.05).Conclusions Short pulse gastric electrical stimulation significantly increases c-fos expression in NTS/hypothalamus,which suggests that NTS/hypothalamus may participate in mediating the effect of GES on gastric function.This effect may be not mediated by ENS.

7.Expression of microRNA-126 in Colon Cancer and Its Clinical Significance

Objective To investigate the expression and biological significance of microRNA-126 in colon cancer tissue.Methods The expression of microRNA-126 in colon cancer tissues and adjacent tissues of colon cancer patients was detected by real-time quantitative polymerase chain reaction (RT-PCR) in 104 patients with colon cancer surgery (total of 104 pairs).The lentiviral vector was used to construct microRNA-126 cell line,and the effects of microRNA-126 on proliferation,migration and invasion of colon cancer cells were further studied in vitro.Results The relative expression of microRNA-126 in colon cancer tissues (0.63±0.11) was significantly lower than that in adjacent tissues (1.08±0.15),the difference was statistically significant(t=14.561,P<0.01).The positive rate of microRNA-126 expression in colon cancer tissues (61.5%) was significantly lower than that in adjacent cancer tissues (86.5 %) the difference was statistically significant(x2=16.908,P<0.05).The expression of microRNA-126 was significantly correlated with Dukes stage,depth of invasion,lymph node metastasis and distant metastasis (P<0.05).In the Transwell experiment with matrix glue,the number of cell migration in transfection group (11.26±4.85) was significantly lower than that in blank control group (264.37±32.15),the difference was statistically significant (t=23.418,P<0.01).In the Transwell experiment without matrix glue,the number of cell migration in transfection group (83.75 ± 13.74) was significantly lower than that in blank control group (339.64 ± 26.38),the difference was statistically significant(t=12.682,P<0.01).MicroRNA-126 could inhibit the proliferation and invasion of SW480 cells.Conclusion MicroRNA-126 can significantly inhibit the development of colon cancer cells and affect the biological behavior of colon cancer cells.
