1.The Bionomics of Streptococcus gordonii and Its Application in Mucosa Vaccine

Streptococcus gordonii is a nonpathogenic gram-positive commensal bacterium and component of the normal microbial flora of the human oral cavity. It is suitable to be as a mucosa vaccine vector due to its special bionomics. knowing the bionomics of Streptococcus gordonii,the general expressing system,and the application of it in mucosa vaccine,will provid important reference for the further development of its mucosa vaccine.
2.Construction of the Recombinant Plasmid with esat6 Gene and Its Expression in Streptococcus gordonii

To construct expressing vector carrying esat6 gene and express this protein in Streptococcus gor-donii GP251. esat6 gene was amplified by PCR with specific primer from genome of Mycobacterium tuber-culosis (MTB)H37Rv. Inserted esat6 into the pMD18-T vector by T/A clone to get recombinant vector pMD18-esat6. Then digested pMD18-esat6 with restriction enzyme, esat6 was cloned to vector PSMB104 and expressed in Streptococcus gordonii GP251. The expression of esat6 protein was detected by Tricine-SDS-PAGE and Western-blot, ELISA technique was also used to detect its secretory volume. Re-striction endonuclease, PCR, Tric ine-SDS-PAGE and Western-blot confirmed that esat6 gene was cloned into expressing vector successfully, and a 10 kD protein secreted in Streptococcus gordonii GP251, this pro-tein has a good immunogenicity. The expression vector of esat6 gene was constructed, and esat6 protein ex-pressed in Streptococcus gordonii1 successfully, it will be benefit for future study.
4.Relative factors on the delayed encephalopathy after acute carbon monoxide poisoning and the study of P_(300)

Objective To explore the relative factors on delayed encephalopathy after acute carbon monoxide poisoning(DEACMP)and the signifiance of P 300 for judging the severity and prognosis of DEACMP.Methods Thirteen aspects such as contact time with carbon monoxide (CO)?coma time et al were analysed between 44 patients with DEACMP (DEACMP group)and 42 patients with acute CO poisoning but without DEACMP (ACMP group). 5.EFFECT OF SELENIUM ON GENERATION OF SUPEROXIDERADICAL IN RAT HEART MITOCHONDRIA

Weanling rats were fed with a low-selenium diet (Se 0.009mg/kg, — Se group) and the diet supplemented with sodium selenite (Se 0. 232mg/kg,+ Se group) for 1, 2 and 3 months respectively. The generation rate of superoxide radical(O_2~-), activity of manganese dependent superoxide dismutase (Mn-SOD) in heart mitochondria and blood Se concentration from the rats were determined. The results showed that the mitochondrial O_2~- generation rate from — Se rats was significantly higher than those from + Se rats at 2 and 3 months. During all the feeding periods, no significant difference in mitochondrial Mn-SOD activity was found between the—Se and + Se groups,while the blood Se concentration in — Se group was significantly lower than those in + Se group at each feeding time and continually decreased in the 2 and 3 month. The results suggested that selenium could depress O_2~- generation in heart mitochondria.

7.Three-dimensional reconstruction and virtual hepatectomy for laparoscopic precise liver resection of right liver tumor

Objective To evaluate the clinical value of three-dimensional reconstruction and virtual hepatectomy for laparoscopic precise liver resection of right liver tumor.Methods 31 cases of right liver tumor from June 2012 to October 2015 at Department of Hepatobiliary Surgery in Affiliated Hospital of Nantong University,were analyzed retrospectively.Patients were divided into three dimensional reconstruction group (n =15) and control group (n =16),Operation time,bleeding volume during operation,postoperative complications and days of hospitalization after operation in two groups were observed.Results All operation was successfully performed with pure laparoscopic right liver tumor resection in three dimensional reconstruction group.In control group,1 case was completed by hand-assisted laparoscopic liver resection,1 case was converted to open operation,the remaining 14 cases underwent total laparoscopic right liver tumor resection.Operation time in 3-dimensional reconstruction group was (141 ± 36)min vs.(207 ± 66)min in control group,bleeding volume was (274 ±88)ml vs.(418 ± 189)ml,hospitalization after operation was (9 ± 3)days vs.(13 ±6)days (all P <0.05).Conclusions Preoperative three-dimensional reconstruction and virtual hepatectomy can ensure the localization of parenchyma tumor during the laparoscopic precise liver resection,clarifing the relationship between tumor operation section and peritumoral vascular or biliary ducts,fulfilling the idea of precise liver resection.
10.Effects of octreotide on the apoptosis of human HSCs and expression of Bcl-2/Bax in HSCs

Objective To investigate the effects of octreotide on the apoptosis of human hepatic stellate cells (HSCs) and expression of Bcl-2/Bax in HSCs,and to reveal the mechanism underlying octreotide against hepatic fibrosis. Methods HSCs lines (HSC-LX2) were incubated with different concentrations of octreotide for 24 and 48 hours. Cell apoptosis was evaluated by Fitc-tunel fluorescence staining. Bcl-2 and Bax protein exoression in HSC-LX2 was detected by immunocytochemistry. Meanwhile, Bcl-2 protein of HSC-LX2 were detected by Western blot assay. Results Octreotide could promote the apoptosis of HSC-LX2, and the apoptosis rate was significantly increased with the concentration of octreotide(P < 0.05). The HSC-LX2 were incubated with the same concentration of octreotide for 24 and 48 hours, the cell apoptosis rate of 48-hour octreotide treatment was significantly higher than that of 24-hour octreotide treatment (P < 0.05). The immunocytochemistry result indicated that octreotide could significantly decrease Bcl-2 expression and increase Bax expression in HSC-LX2 (P<0.05); Western blot assay showed that octreotide could also significantly inhibit Bcl-2 expression in HSC-LX2 (P<0.05). Conclusions Octreotide could induce the apoptosis of HSCs in a dose-and time-dependent manner, the mechanism of octreotide inducing HSCs apoptosis might be associated with down-regulation of Bcl-2 and upregulation of Bax in HSC.
