1.Inhibition of berberine on growth of human nasopharyngeal carcinoma cells CNE-2 in vivo and in vitro

Objective To study the inhibitory effect of berberine on human nasopharyngeal cancer cell line CNE-2 in vivo and in vitro. Methods CNE-2 cell proliferation was measured by MTT assay and cell cycle was analyzed by flow cytometry. Cell morphology was observed with transmission electron microscopy. The cell cycle relative protein was detected by Western blotting. DNA and protein syntheses were assessed by the cellular incorporation of ~ 3 H-TdR and ~ 3 H-Leu, respectively. Anti-tumor activity of berberine in the experimental transplantation tumor CNE-2 was evaluated by relative tumor growth ratio. Results Berberine inhibited CEN-2 cells growth in a time-and dose-dependent manner. MTT Assay showed that the IC_ 50 values of 48 and 72 h were (49.5?5.8) and (13.3?2.0) ?mol/L, respectively. Cell cycle analyses of 50.0 ?mol/L berberine-treated CNE-2 cells by flow cytometry showed the accumulation of cells in the G_2/M phase while 25.0 ?mol/L berberine treatments for 48 h induced apoptosis with the index of (48.9?10.4)%. The inhibition of CNE-2 cell growth by berberine was associated with suppression of cyclin B1, CDK1, and cdc25c proteins. 2.Effect of Eucommiae ulmoides on the Microstructural and Ultrastructural Structure of Femur and Lumbar Vertebrae in Ovariectomized Rats

OBJECTIVE: To evaluate the effect of Eucommiae ulmoides on the microstructural and ultrastructural change of femur and lumber vertebrae in ovariectomized rats.METHODS: 80 4-month-old female SD rats were randomized to 4 groups: sham operation group,model group,alfacalcidol group,E.ulmoides group.In the sham operation group,the abdominal cavities of the rats were cut open to expose ovaries before being sutured.However,the rats in the other 3 groups were ovariectomized bilaterally.At 10 days after operation the rats were administered intragastrically with corresponding drug.After 3 months,femur and lumber vertebrae were dissected and fixed in 4% glutaral for 3 days,then decalcified in 4% EDTA for 4 weeks,and then the structure of femur and lumber vertebrae was observed under light microscope and transmission electron microscope(TEM).RESULTS: Observed under light microscope,the femur and lumber vertebrae were full of fat in the ovariectomized rats and in which the density of bone trabecula decreased significantly;however,in alfacalcidol-treated rats and E.ulmoides-treated rats,the amount of fat decreased markedly while the density and thickness of bone trabecula increased.Observed under TEM,in the ovariectomized rats,rough endoplasmic reticulum(RER),cytolysosome and mitochondrial were plentiful inside osteoclasts in femur and lumber vertebrae,partial osteoblasts atrophied,the amount of RER and other cellular organ decreased significantly and apoptosis appeared for partial cells;however,in alfacalcidol-treated group and E.ulmoicdes-treated group,the above changes improved significantly.CONCLUSION:E.ulmoides has remarkable beneficial effect on osteoporosis in ovariectomized rats.The mechanism might be attributed to its inhibition on the synthesis of fat and the activity of osteoclasts,promotion on the activity of osteoblasts and inhibition on the apoptosis.

In this paper authors explored the clinical development mode of IM, extracted connotation and extension of IM clinics, hoping to promote the development of IM clinics.

Vascular disrupting agents (VDAs) have presented a new kind of anti-cancer drug in recent years. VDAs take advantage of the weakness of established tumor endothelial cells and their supporting structures. In contrast to anti-angiogenic therapy, which inhibits the outgrowth of new blood vessels, vascular targeting treatments selectively attack the existing tumor vasculature. Here we summarized the anti-tumor activities, mechanisms and clinical applications of small molecule VDAs.
8.RP-HPLC determination of valproate concentration in serum

Objective To establish an RP-HPLC method for determining serum level of valproate.Methods A symmetryshield RP18 column (5?m,3.9mm?150mm) was used.The mobile phase,methanol-water(80:20) was used at a flow rate of 1.0ml/min.The detector was Waters 2487 dual?absorbance detector,which was set at the wavelength of 248nm.Serum sample was extracted with 2ml acetic n-hexane,then valproic acid in organic layer was derived by adding into?-bromoacetophenone,and cyclohcxanecarboxyiic acid was selected as an internal stan- dard.Results The calibration curve was linear in the range of 5.0 to 250.0vtg/ml.The average recovery rate was 98.23%,and the RSD within-day and between-day were both less than 3 %.Conclusion This method is suitable for therapeutic drug monitoring of phenytoin.
