1. Effect of resveratrol on apoptosis induction of fibroblast synovial cells and its possible mechanism

Aim To explore the effect of resveratrol (Res) on fibroblast synovial cells (FLS) inadjuvant arthritis (AA) rats treated with low concentration of H2O2, and the role of mitochondrial oxidative stress proteins deacetylase 3 (SIRT3) and manganese superoxide dismutase (MnSOD). Methods Twenty SD rats were randomly divided into normal group and model group. The AA model was induced by subcutaneous injection of the complete adjuvant in toes of model group. On 28th day after modeling, the rats were killed and the changes in serum oxidative stress index in two groups were detected. The expression of oxidative stress protein of SIRT3 and MnSOD in synovial tissues of knee joint of AA rats were detected by immunohistochemistry stainning. The effect of different concentrations of H2O2or Res treatment on FLS proliferation was observed by CCK-8 method. The effect of Res on apoptosis and the expression proteins SIRT3 and MnSOD of FLS treated with low H2O2concentration were detected by Hoechst 33258 and Western blot. Results Compared with normal group, the serum of AA rats showed oxidative stress. The expression of SIRT3 and MnSOD increased in synovial tissues of AA model rats. Low concentration of H2O2promoted FLS proliferation, and Res could dose-dependently inhibit proliferation treated with low concentration of H2O2, and increased FLS apoptosis. Moreover, with the increase of Res dose, the expression of SIRT3 and MnSOD protein decreased in FLS treated with low concentration H2O2. Conclusions AA rats are in a state of oxidative stress, low concentration of H2O2can promote FLS growth in AA ras, and Res treatment can inhibit FLS proliferation, promoting FLS apoptosis. The mechanism may be related to the reduction of anti-oxidative stress effect.
2.Identification of Schisandra chinensis with white fruits based on ITS2 sequences.

OBJECTIVETo analyse a special kind of Schisandra chinensis with the white fruit using ITS2 barcode at molecular levels.
METHODITS2 regions were sequenced bidirectionally. Sequence assembly and consensus sequence generation were performed using the CodonCode Aligner, MEGA 5.0 software was used to align the sequences. The ITS2 secondary structure was predicted using ITS2 web server, BLAST 1 method was used to identify the S. chinensis with the white fruit.
RESULTThe length of the ITS2 sequence was 231 bp. And the sample was identified as S. chinensis using the method of BLAST 1. Their mean interspecific genetic distance (K2P distance) among the populations of the S. chinensis with the white fruit and S. chinensis was far lower than the mean interspecific genetic distance between the S. chinensis and S. sphenanthera.
CONCLUSIONBy using ITS2 the S. chinensis with the white fruit was identified as S. chinensis, and the ITS2 barcode could be used to identify S. chinensis and S. sphenanthera.

3.Effect of forchlorfenuron on fruit morphology and lignans content of Schisandra chinensis.

The effect of plant growth regulator forchlorfenuron (CPPU) 1 x 10(-6), 0.67 x 10(-6), 0.5 x 10(-6) on fruit morphology and effective components lignans was studied. Those morphologies were the combination of four basic morphological changes. The result showed, diametre were increased and longitudinal diametre of fruits were inhibited by foliage fertilizers including CPPU. At the same time, 1 000-grain weight and yield showed the varying degrees increase under CPPU. The order of the degree was 0.5 x 10(-6) > 1 x 10(-6) > 0.67 x 10(-6). Six lignans content of Schisandra chinensis of different harvest time and different CPPU processing groups were determined, the results showed that lignans accumulation occurred mainly in periods of premature the half mature fruiting stages. Under the 0.67 x 10(-6) CPPU treatment, schisandrol B, schisandrin B, schisandrin C content of S. chinensis showed different increase.

4.Clinical efficacy of compound periploca liquid in treating condyloma acuminatum and its effect on human papilloma virus DNA expression in vitro.

OBJECTIVETo investigate the clinical efficacy of compound periploca liquid (CPL) in treating condyloma acuminatum (CA), and to explore its mechanisms at molecular level.
METHODSEighty-one patients with CA were randomly divided into three groups, 30 patients in Group A were treated with CPL, 21 in Group B were treated with periploca syrup and 30 in Group C were treated with Youtuoxin (YXG). The clinical efficacy and adverse reaction occurred in the three groups was evaluated respectively. Besides, change of human papilloma virus (HPV) DNA in two patients with CA was determined by polymerase chain reaction (PCR) in vitro after being treated with CPL and periploca, the monarchic drug of CPL.
RESULTSThe cure rate obtained in group A, B, and C was 56.67%, 42.86% and 63.33% respectively, the total effective rate in them was 83.33%, 71.43% and 86.67% respectively, the difference of therapeutic efficacy was insignificant among the three groups. But the adverse reaction occurrence in them (6.67%, 4.76% and 86.67%) was significantly different (P < 0.01). The recurrent rate in them was 14.29%, 12.5% and 47.1% respectively. PCR showed negative expression of HPV in the two samples of CA of same concentration after the verrucous homogenate suspension being treated with CPL or periploca syrup of different concentration, while it was positive after the suspension was treated with the group of normal saline without any drug.
CONCLUSIONThe therapeutic efficacies of CPL, periploca syrup and Youtuoxin are not significantly different, but the former two have advantages of less adverse effects and lower recurrent rate. And they are possibly having germicidal action on HPV-DNA in CA tissue in vitro.

5.HPLC determination of six lignans in different parts of Schisandra chinensis.

OBJECTIVETo compare the content of six lignans of different parts of Schisandra chinensis.
METHODAgilent TC-C18 (4.6 mm x 250 mm, 5 microm) was used with acetonitrile-water gradient system as mobile phase. Wave length was 250 nm. The flow rate was 1 mL x min(-1). Column temperature was 30 degrees C.
RESULTThe total lignans content of wild Schisandra chinensis was higher than that of the cultivated varieties. The total lignans content of different parts varied significantly, wherein the root > main branch > side branches > leaf.
CONCLUSIONThis method is stable, reliable, can be used for the quality evaluation of different parts of Schisandra.

8.Clinical experience of 70 cases of cerebral arteriovenous malformations embolization with Onyx, a novel liquid embolic agent.

OBJECTIVETo report our clinical experience of using Onyx, a new liquid embolic agent, to treat cerebral arteriovenous malformations (AVMs) as well as its efficacy.
METHODSSeventy cases were placed with 6F sheath in the femoral artery after Seldinger puncture and 6F guiding catheter was introduced into the internal carotid artery or vertebral artery, then a microcatheter was navigated into the nidus of AVMs. Slow injection of Onyx under fluoroscopic control was performed to embolize cerebral AVMs using the "plug and push" technique.
RESULTSThirteen AVM cases (18.6%) were totally occluded by Onyx and 5 cases of which didn't recurrence at 6-month after operation. Thirty-eight cases (54.3%) were subtotally occluded, while another 19 cases (27.1%) were partially embolized. Severe cerebral hemorrhage occurred in 4 cases, 2 of which had mild to severe hemiplegia after operation, and one died. Mild hemiplegia was also found in 1 case due to functional area embolization, and visual field deficit in 2 cases.
CONCLUSIONSOnyx has unique and distinctive superiority in treating cerebral AVMs. Nonetheless, the correct embolization technique should be learned to achieve good clinical results and avoid complications.

9.Risk factors for intracranial aneurysm in a Chinese ethnic population.

BACKGROUNDIntracranial aneurysm (IAN) is a protruding bubble or a sac on a brain artery that balloons out over time, which may lead to spontaneous subarachnoid hemorrhage (SAH), ultimately disability and mortality. Current research indicates that the disease is due to multiple causes, including environmental factors and various congenital abnormalities of blood vessels. Apart from congenital predisposition, various high-risk factors such as sex, age, hypertension, and atherosclerosis are involved in the formation of intracranial aneurysms. The aim of this study was to investigate the risk factors associated with the formation of sporadic intracranial aneurysms in Chinese Han ethnic patients.
METHODSA total of 251 patients with intracranial aneurysm and 338 patients with other cerebral diseases (control group) were enrolled in this study. Single factor and logistic regression model were used to analyze the association of intracranial aneurysms with age; sex; cigarette smoking; alcohol or cocaine consumption; history of hypertension, coronary artery disease, diabetes mellitus and inherited connective tissue disease; and the levels of fasting blood glucose and blood fat. The data expressed as mean +/- standard deviation were processed with the statistical software SPSS13. Quantitative and qualitative data were analyzed by the independent-sample t test, and the chi-square test respectively. Logistic regression method was used to analyze the multiple factors.
RESULTSIn the 251 patients, 163 (64.94%) were at age of 40 to 60 years. Sex (OR, 1.41; 95% CI, 1.01 - 1.96), cigarette smoking (OR, 1.81; 95% CI, 1.06 - 3.10), hypertension (OR, 2.32; 95% CI, 1.30 - 4.16) and fasting blood glucose were significantly associated with intracranial aneurysm (P < 0.05). Intracranial aneurysm was correlated with alcohol consumption, coronary artery disease, and the level of blood lipids (P > 0.05). Using logistic regression analysis, we identified female sex and advanced age as significant risk factors for sporadic intracranial aneurysms.
CONCLUSIONSSporadic intracranial aneurysms mostly occur in people aged 40 to 60 years. Feminine, cigarette smoking, and hypertension are independent risk factors for the disease, and the gender is the most significant factor. Advanced age can increase the effect of these risk factors.

10.Soluble guanylate cyclase stimulators:research advances

The soluble guanylate cyclase (s GC) is a key signal transduction enzyme in the nitric oxide (NO) -sGC-cyclic guanosine monophosphate (cGMP) pathway, which can be activated by NO and thus catalyzes the conversion of guanosine triphosphate (GTP) into the c GMP. As a second messenger, cGMP modulates the activity of downstream effectors, including the cGMP-dependent protein kinases (cGK), cGMP-dependent phosphodiesterases (PDE) and ion-gated channels, and ultimately participates in the regulation of several physiological functions, including the function of cardiovascular, gastrointestinal and central nervous systems. sGC stimulators could not only directly stimulate s GC but also synergistically act with NO to increase the sGC enzyme activity, which has shown a great potential to treat various diseases via regulating the NO-s GC-cGMP signaling pathway. Here, we give an overview of NOindependent sGC stimulators in clinical trial.
