1.PRESSOR EFFECT OF NALOXONE IN ACUTE EXPERI-MENTAL RABBITS

Experiments were carried out on the anesthetized, operated and paralyzed rabbits to study if endogenous opioids are involved in the cardiovascular activity in the acute experimental conditions under which many experiments of pharmacology & physiology were performed. In 6 rabbits, low dose naloxone (0.4 mg/kg, i.v. ) caused sevral temporary pressor waves in 2.9?1.1 min after the injection & apperance of the arrhythmias in 2 animals. In other 9 rabbits, high dose naloxone ( 5 mg/kg, i.v.) raised basal blood pressure in 13.9?7.4 min after the injection, and caused arrhythmias in 5 animals. When the pressor effect was the highest, monoamine metabolites did not change significantly in CSF sampled from the terminal of midbrain aquaduct. Our other experiment has showed that NA & MHPG of the CSF increased during the central stimulation-induced pressor effect. Thus, it is supposed that endogenous opioids play an inhibitory effect on the cardiovascular system in the conditions like present experiment, and the peripheral opioid receptors which are poorly sensitive to naloxone may be involved in the effect.
2.Content Comparison of Polydatin and Emodin between Polygonum Cuspidatum Pieces and Dispensing Gran-ules

Objective:To compare the content of polydatin and emodin between polygonum cuspidatum pieces and dispensing gran-ules to provide theoretical basis for the improvement of preparation process of the granules. Methods:An HPLC method with linear gra-dient elution was used to determine the content of polydatin and emodin in the pieces and dispensing granules, and the difference in the two contents was compared. Results: The content of polydatin and emodin in polygonum cuspidatum pieces met the requirement de-scribed in China pharmacopoeia, which was lower than that in the dispensing granules, and the contents of the two components in the commercial dispensing granules were not equivalent to the labeled amount, which was 1/15 and 1/27 of that in the pieces. Conclu-sion:The transport rate of the traditional preparation process of dispensing granules is with poor efficiency, which should be improved.

Contents and modes are described in detail. It proposes va-rious forms of education should be carried out according to different user groups, so as to further promote the development of evidence-based medicine education.
4.Effect of Electroacupuncture on Experimental Epilepsy: Roles of Different Acupoints and Stimulation Parameters

Objective: The strategies against epilepsy are currently very limited. The author performed this work to determine the effect of electroacupuncture (EA) on epileptic seizures and define the optimal EA conditions (acupoints and stimulation parameters). Methods: Experimental epilepsy was induced by injecting kainic acid into the lateral cerebral ventricle of adult male SD rats. EA with a current of 1 mA/100 Hz or 1 mA/10 Hz was applied to the model starting at 0.5 h after the injection for 30 min. Four pairs of acupoints were tested, including Renzhong (GV 26) + Dazhui (GV 14), Jinsuo (GV 8) + Yaoqi (Ex-B 9), Neiguan (PC 6) + Quchi (LI 11) and Fenglong (ST 40) + Yongquan (KI 1). Results: Behavioral scores showed that (1) either high (100 Hz) or low (10 Hz) frequency EA at different acupoints attenuated epileptic seizures (P<0.05 or 0.01 vs. the control), except EA at Neiguan (PC 6) + Quchi (LI 11) with 1 mA/10 Hz current; (2) EA with low frequency stimulation (10 Hz) induced a better effect at Fenglong (ST 40) + Yongquan (KI 1) than other acupoints (P<0.05 or 0.01); (3) there was no significant difference in EA effects induced by various acupoints when exposed to high frequency EA (100 Hz); and (4) high frequency EA (100 Hz) elicited a better effect than low frequency EA (10 Hz) in all groups (P<0.05 or 0.01) except for that of Jinsuo (GV 8) + Yaoqi (Ex-B 9). EEG showed the similar tendency, i.e., EA generally reduced the amplitude and frequency of epileptic discharges. Conclusion: Electroacupuncture attenuates epileptic seizures, which is relatively specific to EA parameters and acupoints.
5.Correlation analysis of HPV and cervical cancer

Objective To investigate the correlation between HPV positive rate and incidence rate of cervical cancer.Methods 438 women,who reveived cervical Pap smears and biopsy,were included in the present study.Human papilloma virus nucleic acid amplification genotyping assay kit was used to detect 21 kinds of HPV subtypes.Results There were 102 patients with cervical cancer (23.29%) and HPV-positive 94 patients (21.46%),which had no significance difference (x2 =0.421,P =0.517).There were 56 patients HPV16 positive (54.90%),19 patients HPV58-positive (18.63%),12 patients HPV18 positive (11.76%) and 8 patients HPV52 positive (7.84%),which were significantly higher than those of other types of HPV-positive rate(all P < 0.05).The proportion of the patients with invasive cervical cancer in HPV i6,HPV58,HPV18,or HPV52-positive was significantly higher than that of intraepithelial lesions and carcinoma in situ (all P < 0.05).Conclusion HPV16,HPV58,HPV18,and HPV52 have high correlation with uterine cancer.
6.The impact of the Jiawei Xiaochaihu particle and it's prescription on immune function of ITP mice

Objective:To observe the impact of the Jiawei Xiaochaihu particle and it's prescription on immune organ and immune function of ITP mice.Methods:The ITP mice were divided into five groups at random:(1)Nomal group;(2)ITP model group;(3)Jiawei Xiaochaihu particle group;(4)Xiaochaihu particle group;(5)Danshen and Sanqi particle group.All ITP mice were administrated 7days later by 30ml per kilogram the rat's weight once a day for 14 days.21 days after the model mice were made,peripheral blood T lymphocyte subpopulation was detected with flow cytometry.Antibody against platelets was measured by ELISA.Peripheral blood IL-2 and IL-10 were measured by radio immuno assay(RIA).Thoracic gland and spleen were weighted to compute the immune organ index.Results:Spleens of ITP mice had intumesce of spleens and a rise of the index;Peripheral blood platelet antibody was also increased;A disproportion was found in T lymphocyte subpopulation,CD8 upgrade,CD3 and CD4 descent,CD4/CD8 also descent.As one of the Th1 cytokine,IL-2 expression accentuated while IL-10,the kind of Th2 cytokine,was inhibited.There were significances when compared with the nomal group(P0.05).Conclusion:There are indeed dysfunction in immune organ and its function of ITP mice.Jiawei Xiaochaihu particle and Xiaochaihu particle can exert a regulation,but the Danshen and Sanqi particle not.

10.EFFECTS OF HIGH-FAT DIET ON LIPID METABOLISM IN SR-AⅠ/Ⅱ GENE KNOCK-OUT MICE

Objective To observe the alteration of scavenger receptor class A types Ⅰand Ⅱ (SR-AⅠ/Ⅱ) gene knock-out on lipid metabolism in mice fed with high-fat diet, and explore the underlying mechanism. Method The SR-AⅠ/Ⅱgene knock-out and wild-type male mice were fed with normal and high-fat diet for 12 w. Thereafter, the level of lipid metabolism (such as the levels of lipids in blood and liver) was detected with enzyme method or oil red O staining, and the expression of scavenger receptor class B typeⅠ(SR-BⅠ) and CD36 in liver was analyzed by RT-PCR. Results Under high-fat diet condition, as compared with wild-type mice, the levels of TG, TC, LDL and HDL in SR-AⅠ/Ⅱgene knock-out mice were decreased at 3, 6, 12 w (P0.05). Conclusion The alteration of lipid metabolism induced by high-fat diet in SR-AⅠ/Ⅱgene knock-out mice might be relative with the up-regulated SR-BⅠmRNA expression and the counter transport of peripheral lipids to liver.
