1.Therapeutic effect of pharyngeal treatment machine combined resuscitating acupuncture on dysphagia after stroke

Objective:To explore therapeutic effect of pharyngeal treatment machine (PTM) combined resuscitating acupuncture (RA) on patients with dysphagia after stroke.Methods: A total of 98 stroke patients with dysphagia were selected.According to random number table, they were randomly and equally divided into pure pharyngeal treatment group (received pure PTM therapy based on routine treatment) and combined treatment group (received RA based on pure pharyngeal treatment group).Video fluorography (VFG) score and swallow-quality of life (SWAL-QOL) score before and after treatment, and total effective rate were evaluated and compared between two groups.Results: Compared with before treatment, there was significant rise in VFG score, and significant reduction in SWAL-QOL score in both groups after treatment, P<0.01 all;compared with pure pharyngeal treatment group after treatment, there was significant rise in VFG score [(5.46±2.04) scores vs.(6.54±2.75) scores], and significant reduction in SWAL-QOL score [(741.33±25.92) scores vs.(632.54±26.77) scores] in combined treatment group, P=0.032, 0.001 respectively.Total effective rate of combined treatment group was significantly higher than that of pure pharyngeal treatment group (93.88% vs.75.51%), P=0.014.Conclusion: Compared with pure pharyngeal treatment machine, pharyngeal treatment machine combined resuscitating acupuncture possesses better therapeutic effect for recovery of swallowing function, which is worth extending.

4.Recombinant Lactobacilli for Ethanol Production

Recombinant plasmids pHY-PA, pBBR-PA were constructed in which genes pdc and adhB were placed under the control of tac promoter, respectively, and had successfully expressed in Escherichia coli. Then these recombinant plasmids were electroporated into Lactobacillus strains for ethanol production. Preliminary ethanol fermentation using these Lactobacillus strains and their recombinants was carried out using 42℃ as fermentation temperature. The results indicated that introducing pdc and adhB, ethanologenic pathway was successfully constructed in L.plantanum CICIM B0080. 0.4% (V/V) ethanol was detected at the end of fermentation with 6.7% glucose, and that is 67-fold as the wild-type B0080. Two-fold of ethanol production was detected in L.amylovorus B0112 (pHY-PA) and L.acidophilus B0068 (pBBR-PA). Introducing both pdc and adhB, and meanwhile knock-outing the lactate dehydrogrnase gene may better convert carbon flux to ethanologenic direction.

Signal transducer and activator of transcription 3 (STAT3), an acute-phase response protein, is ac-tivated to over-express by cytokines .STAT3 also acts as a transcriptional factor to regulate the expression of cytokines .O-ver-expression of cytokines is accompanied by STAT 3 activation and over-expression in acute pancreatitis .Meanwhile , the proliferation of pancreatic stellate cells in chronic pancreatitis is mediated by STAT 3.In this review, the research progress in STAT3 function is summarized to elaborate its potential role in the pathogenesis of pancreatitis .
6.X-knife radiosurgery in the treatment of brain metastasis of lung carcinoma

Objective To discuss the X-knife radiosurgery (XKS) in the treatment of brain metastasis of lung carcinoma. Methods A total of 100 patients with similar prognostic factors were divided into two groups with 50 patients in each group, receiving either whole-brain radiotherapy alone (30~40 Gy/3~4 weeks) (Radiotherapy Group) or XKS combined with radiotherapy (Combination Group). In the Combination Group, 27 patients received XKS with single fraction of radiation, with a median prescription dose of 14.2 Gy, and the other 23 patients received multiple fractions of radiation (5~10 Gy/f, 3 times weekly), with a total dose of 15~30 Gy. Results In the Combination Group and the Radiotherapy Group, the median survival time was 16.4 and 10 months, respectively (P=0.0064), the 2-year local tumor control rate was 88% (44/50) and 44% (22/50), respectively (?2=21.569,P=0.000), and the effective rate under CT or MRI scanning at 1~3 months after treatment was 87.5% (35/40) and 52.2% (24/46), respectively (?2=16.497,P=0.001). An analysis on the cause of death showed that 11.9% of patients (5/42) in the Combination Group died from brain metastasis, which was significantly lower than that in the Radiotherapy Group (55.6%, 25/45) (?2=25.908,P=0.000). The incidence of complications was not significantly different between the Combination Group (8%, 4/50) and the Radiotherapy Group (4%, 2/50) (?2=0.709,P=0.400). Conclusions Combined use of X-knife radiosurgery and routine radiotherapy has better therapeutic effects than radiotherapy alone for treating brain metastatic tumor.
7.The changes in serum concentrations of the interferon-?-inducible protein 10(IP-10) in different types of diabetic patients and their significance

Objective To investigate the significance of serum IP-10 levels in different types of diabetic patients. Methods Serum IP-10 level in 78 cases with type 1 diabetes,49 cases with type 2 diabetes and 33 cases of healthy controls was measured with ELISA assays. Type 1 diabetic patients were divided into classic type 1 diabetes (n=39) and latent autoimmune diabetes in adults (LADA,n=39) according to their disease process,and they were also divided into autoimmune type 1 diabetes (n=58) and idiopathic type 1 diabetes (n=20).Type 2 diabetic patients,according to carotid intima-media thickness (IMT),were divided into patients with (n=24) or without atherosclerosis (n=25). Results 1) Serum IP-10 levels were not statistically different among type 1 diabetes,type 2 diabetes and healthy controls. 2) Serum IP-10 concentrations in autoimmune type 1 diabetes with positive islet autoantibody were higher than those in healthy controls(184.96?104.48pg/ml vs 146.10?74.61pg/ml,P=0.03);while there were no difference in type 1 diabetes with variable disease durations. 3) No difference in IP-10 levels was found between type 2 patients with(173.5?69.6pg/ml) and without atherosclerosis (188.5?79.7 pg/ml). Conclusions Serum IP-10 level is augmented in autoimmune type 1 diabetes.
9.Effects of levocamitine cardioplegic solution on cardio-myocyte apoptosis in patients undergoing heart valve replacement with cardiopulmonary bypas

Objective To investigate the effects of levocarnitine used as an ingredient of cardioplegic solution on cardiomyocyte apoptosis in patients undergoing heart valve replacement under cardiopulmonary bypass (CPB) .Methods Twenty-four NYHA grade Ⅱ or Ⅲ patients of both sexes (16 males, 8 females) aged 27-67 yrs undergoing heart valve replacement under CPB were randomly allocated into 2 groups (n = 12): levocarnitine group and control group. In levocarnitine group levocarnitine 6 g was added to 1 000 ml of 4℃ St Thomas Ⅱ cardioplegic solution, while in control group equal volume of normal saline was added instead of levocarnitine. Cardioplegic solution 15 ml?kg-1 was injected to perfuse the heart every 30 min during aortic cross-clamping. The KC1 concentration of the cardioplegic solution was reduced by half for the last injection. During CPB the naso pharyngeal temperature was maintained at 25-27℃. Cardiac index (CI) and left ventricular ejection fraction (LVEF) were measured 1 day before and 7 days after operation using heart color ultrasonography. A small piece of myocardial tissue was obtained from right atrium for assessment of apoptosis using terminal deoxynueleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) . The expression of Bax and Bcl-2 in cardiomyocytes was determined by immuno-histochemistry, and Bcl-2/Bax ratio was calculated. 10.EFFECTS OF EXOGENOUS PTEN GENE ON THE CELL CYCLE AND PROLIFERATION OF SHG44 GLIOMA CELL

Eukaryotic expression vector containing PTEN gene was transfected into SHG44 cell under mediation of lipofectamine, and positive cell clones were selected and amplified. Expression of PTEN gene was detected by in situ hybridization and immunohistochemistry. Flow cytometry, electron microscope and other methods were adopted to measure the effects of PTEN on cell cycle, ultrastructure, and cell growth of the transfected glioma cells, as well as on tumorigenicity in nude mice. The results showed that PTEN gene and protein were expressed in the glioma cells transfected with PTEN gene. The progression of cell cycle was arrested from G1 to S phase. The growth of cells transfected with PTEN gene in nude mice was dramatically inhibited compared with the parent SHG44 cells. This experiment suggested that the growth of SHG44 glioma cells could be arrested at G1/S phase, and it could be significantly suppressed by exogenous PTEN gene.
