Objective:Toexplore the effectofinsulin(INS) administration on the expression of Bcl-2 protein and the apoptosis of corneal histiocytes following rabbit model of corneal alkali burn.Methods:to establishⅡdegree rabbit model of corneal alkali burn,and respectively to inject normal saline(B group),INS(C group)in the subconjunction each day,and the normal rabbit eyes as controls(A group).The different time after operation the corneal samples were remove and fixed.the apoptosis of corneal histiocytes were detected by light microscope,electron microscope and transferase-mediated dUTP-digoxigenin nick and labeling assay(TUNEL),the expression and alteration of Bcl-2 protein using immunohistochemisty.Results:The TUNEL positive cells were present slightly in the epithelium of normal comea and not present onstroma and endothelium.Bcl-2 were expressed mainly on the normal corneal epithelium,a lower number of Bcl-2 expression on stroma and endothelium.The TUNEL positive cells of group C was stgnificantly lower than that(P

Objective To investigate human immunodeficiency virus type 1 (HIV 1)specific CD8'T cell responses and related human leukocyte antigenⅠ(HLA-I)restriction sites.Methods Four Epstein-Barr virus (EBV)strains-transformed B-lymphoblastoid cell lines (BLCL)from periph eral blood monocytes (PBMC)of 4 healthy individuals were established.Using synthesized peptides in HIV-1 Gag,Vif and Nef regions as epitopes and using BLCL as antigen presenting cells (APC), the frequency of interferon-7(IFN-?)secreting cells in CD8 T cells from a HIV-infected long term nonprogressor by enzyme-linked immunospot (ELISPOT)assay in vitro were assessed.Moreover, matching analysis of HI.A-Ⅰrestriction sites were performed.Results HIV-1 epitopes were presen ted restrictively by HLA-Ⅰmolecules in specific CD8 T cell responses,and this antigen presenting had high specificity.HLA-Ⅰrestriction sitesA03,A26,C04andC08 contributed to presentepitopes in HIV-1 C-ag region,A03 and C08 to HIV-1 Nef as well.Conclusion HLA-Ⅰrestriction sites may include A03,A26,C04 and C08 in HIV-1 Gag region,and A03,C08 in Nef region.
4.Extraction Technology of Volatile Oil from Qingjie Granules and Optimization of Inclusion Technology by Box-Behnken

Objective To optimize the extraction of volatile oil from Qingjie Granules and process of inclusion compound of beta-cyclodextrin. Methods Water Distillation was used for extraction. Extraction time, grinding degree, and the amount of water were set as inspection factors, and volatile oil volume was set as the evaluation index to inspect extraction process of volatile oil from Qingjie Granules. With inclusion rate as the evaluation index, the single factor test and the Box-Behnken combined with the response surface method were used to choose the optimum inclusion process. Results The optimum extraction process for Schizonepetae Herba, Saposhnikoviae Radix and Forsythiae Fructus coarse powder should be with 10 times amount of water, extracting 3 h. Inclusion method should be saturated water solution method, and the inclusion process of volatile oil as feed and beta-cyclodextrin inclusion ratio was 1:12; the temperature was 40 ℃; inclusion time was 3.5 h. By means of TLC, UV and IR spectra, the formation of the inclusion compound of volatile oil in clear solution particles was preliminarily proved. Conclusion The optimum extraction and inclusion process of volatile oil from Qingjie Granules are stable and feasible, which can be used in industrial production.

6.Survey on the levels of high density lipoprotein and low density lipoprotein among the staff in a college

Objective To analyze the levels of high density lipoprotein ( HDL) and low density lipoprotein ( LDL) among the staff in a college .Methods Fasting serum samples were collected from 2 234 paticipants .The lev-els of HDL and LDL in different sex and ages were estimated .Results The average value and abnormal rate of LDL in males were 3.20mmol/L and 49.1%,respectively.The average value of LDL in males was higher than normal val-ue.The average value and abnormal rate of LDL in females were 3.03mmol/L and 42.3%,respectively.The value of LDL in females who less than 54 years old was lower than that in males of the same age ( t=5.33,10.56,all P<0.01).Conclusion The abnormal rate of LDL had significant increase and the level in males was higher than that in females.The health education is necessary to prevent and control the abnormal level of LDL .
7.Exploration on the method for calculating the energy requirement in the adult patients with diabetes mellitus

Objective To search a proper method for calculating the energy requirement in the patients with diabetes.Methods Based on the energy metabolic characteristic of the patients with diabetes and main effective factors of energy requirement,such as age (A),height (H),weight (W),activity factor (AF)and climate factor (CF),we have established a formula:E(kcal)=[(H-A)?6+500?WI 2]?AF?CF.Results This formula is of accuracy and applicabi- lity ,compared with traditional method for calculating energy requirement.Conclusion It is available that this method is used in dietotherapy of patients with diabetes.
8.Applied anatomic study on the temporal flap pedicled with orbicularis oculi muscle

Objective To investigate the anatomic basis of the temporal flap pedicled with the orbicularis oculi muscle. Methods The morphology and the blood supply of the orbicularis oculi muscle were studied on 15 cadavers by surgical dissection and vascular cast.Using digital subtraction angiography (DSA),we observed the branches of the superficial temporal artery in 15 patients.The data was then analysed by the computed image analysis system. Results The orbicularis oculi muscle was wide and thin and located superficially.It was supplied by the branches of many arteries and the branches have numerous connections with the branches of superficial temporal artery.The position of the zygomaticoorbital artery was unvaried .The diameter of the artery was up to 1.0 mm. Conclusion Zygomaticoorbital artery is the most important vessel for the temporal flap pedicled with the orbicularis oculi muscle.
9.The effect of NF-?B decoy on the chemokine expression of human bladder cancer cell line

Objective To investigate the effect of nuclear factor ?B(NF-?B)decoy on the chemokine expression in bladder cancer cell line. Methods Human bladder cancer cell line EJ,NF-?B decoy ODN were used as a NF-?B inhibitor(scrambled NF-?B decoy was used as control).NF-?B DNA binding activity was detected by electrophoretic mobility shift assay (EMSA);and p65 subunit of NF-?B was detected by RT-PCR and Western blot.Chemokines including IL-8,MCP-1,RANTES were detected by RT-PCR. Results EMSA showed that NF-?B decoy inhibited NF-?B activation induced by TNF-?.RT-PCR or Western blot test suggested that p65,IL-8,MCP-1 and RANTES were upregulated by TNF-? and downregulated by NF-?B decoy.However,mutated decoy ODN had no effect on them. Conclusions Chemokines can be detected in bladder cancer.They are activated by TNF-? and inhibited by NF-?B decoy.
10.Establishment of androgen-independent human prostate cancer cell strain,LNCaP C-81

Objective To establish and validate andro ge n-independent human prostate cancer LNCaP cell model. Methods Androgen-dependent LNCaP parental C-33 cells were maintained in a regul ar cell-culture medium,that is,phenol red-positive RPMI 1640 medium supplement ed with 10% fetal bovine serum,1% glutamine and 0.5% gentamicin.Upon continuousl y passaging,androgen-dependence of these LNCaP cells decreased gradually,thus,t he androgen-independent LNCaP cell model which derived from androgen-dependent cells was established. Results Upon continuous passage, the biological behavior of androgen-dependent parental LNCaP C-33 cells (pass age number less than 33) was altered.LNCaP C-81 cells (passage number higher th an 80) clearly exhibited more aggressive growth and lower androgen-dependence t han C-33 and C-51 cells (passage number between 34 and 81) in vitro and in viv o. Conclusions The LNCaP cell model closely recapitulate s the transition of human prostate cancer from androgen-dependent to hormone-r efractory state under the androgen nondeprived condition. This cell model may pr ovide the opportunity to understand the molecular mechanisms involved in the and rogen-independent growth of cancer cells during prostate cancer progression.
