1.Bone marrow mesenchymal stem cells combined with chitosan for articular cartilage injuries

BACKGROUND:Trauma easily leads to the emergence of articular cartilage defects, which is a difficult problem in the orthopedics field. Tissue engineering technology provides a new method for cartilage repair. OBJECTIVE:To explore the feasibility of combining chitosan and bone marrow mesenchymal stem cels to repair injured articular cartilage in rabbits. METHODS: Cultured rabbit bone marrow mesenchymal stem cels were seeded onto chitosan scaffold, and then the composite material was implanted into the defect as experimental group. Rabbits with no treatment served as control group. Gross observation and toluidine blue staining were carried out at 6 and 12 weeks after operation. RESULTS AND CONCLUSION:At 6 weeks after operation, the control group had only fibrous tissue hyperplasia, and in the experimental group, cartilage-like tissues were generated at the defect site. At 12 weeks after operation, a smal amount of hyaline cartilage-like tissues were observed in the control group, and the defects in the experimental group were covered with smooth and hyaline cartilage tissues. After 12 weeks, the toluidine blue staining was light in the control group with a smal amount of cartilage tissues; in the experimental group, the toluidine blue staining was remarkable, and the defects were completely covered with hyaline cartilage tissues, and cartilage cels were increased in number. The findings indicate that chitosan-bone marrow mesenchymal stem cels composite material is better to induce cartilage tissue formation and promote cartilage defect repair.
2.ABC Analysis of Non-antimicrobial Temporary Purchased Drugs in a Hospital in 2014

OBJECTIVE:To provide reference for scientific and standardized management of temporary purchased drugs. METHODS:By using ABC analysis method,the data of non-antimicrobial temporary purchased drugs in a hospital in 2014 were analyzed statistically with ABC analysis in terms of quantity,the number of types and departments. Class A drugs were analyzed es-pecially. RESULTS:A total of 152 kinds of drugs were purchased temporarily,including 20 464 boxes,20 kinds of class A drugs, which accounted for 70.56%. Main types were antitumor drugs and nutraceuticals by pharmacology;26 departments temporarily purchased drug,mainly in tumor department and hematology department. CONCLUSIONS:The types of temporary purchased drugs meet the clinical demand in the hospital basically. The necessary type A drugs used frequently can be used as reference of tem-porary drug purchase;the temporary purchased drugs can be managed scientifically and normatively based on ABC analysis.
3.Quantitative analysis of several alkaloids in Fritillaria L. by capillary electrophoresis

D glucoside in Fritillaria L by capillary electrophoresis Methods 9 species of bulbus Fritillaria L. were analysed with internal standard by the mode of capillary zone electrophoresis Results The 3 alkaloids were separated completely with the method which showed good recovery and reproducibility Conclusion The method was proved to be quick, simple, and efficient, and provided a reliable basis for the quality control and evaluation of this plant medicine
4.Proliferation and differentiation of mesenchymal stem cells from the synovial tissue in patients with osteoarthritis

BACKGROUND:With respect to mesenchymal stem cels from other sources, synovial mesenchymal stem cels are rich in source, and moreover, the synovial tissue can regenerate quickly after partial hepatectomy and lead to fewer complications, in recent year, which have become a hot spot in stem cel research. OBJECTIVE:To observe the proliferation and directional differentiation of synovial mesenchymal stem cels from osteoarthritis patients. METHODS:Synovial mesenchymal stem cels were isolated and cultured. MTT assay was used to detect cel proliferation ability. Alkaline phosphatase activity was detected quantitatively at 7 days of osteogenic induction, and osteogensis-related gene expression was measured at 7, 14, 21 days of osteogenic induction. Alizarin red staining was performed at 21 days of induction. RESULTS AND CONCLUSION:(1) Passage 3 synovial mesenchymal stem cels proliferated faster, which were in latent period at 1, 2 days after inoculation, in logarithmic growth phase at 3-6 days, and then entered into the plateau phase at 7 days. (2) The activity of alkaline phosphatase was significantly higher in the induction group than the control group at 3, 7, 10 days after osteogenic induction (P < 0.05). The cels were positive for alizarin red staining at 21 days of osteogenic induction, and there were calcium deposits and calcium nodules in the extracelular matrix. (3) Bone-binding protein and Runx2 were visible at 7 days of osteogenic induction, and reached the peak at 21 days. These findings indicate that synovial mesenchymal stem cels from patients with advanced osteoarthritis have strong proliferation ability, which can differentiate into osteoblasts under in vitro induction.
5.Role of modified Seldinger technique combined with vascular ultrasonography in the placement of peripherally inserted central catheters in patients with breast cancer undergoing postoperative chemotherapy

Objective To investigate the role of modified Seldinger technique combined with vascular ultrasonography in the placement of peripherally inserted central catheters (PICC) in patients with breast cancer undergoing postoperative chemotherapy.Methods Totally 120 patients with breast cancer undergoing postoperative chemotherapy were equally divided into the observation group and the control group using the table of random number.In the observation group,PICC were placed using the modified Seldinger technique combined with vascular ultrasonography; in the control group,the PICC were placed using the traditional blind puncture technique.The success rate of first puncture,success rate of first placement,and the incidences of mechanical phlebitis and thrombosis were compared between two groups.Results The success rate of first puncture,success rate of first placement,and the incidences of mechanical phlebitis were 91.67%,100%,and 3.33%,respectively,in the observation group and 76.67%,86.67%,and 45%,respectively,in the control group (P =0.0061,P =0.0434,and P < 0.0001).However,no significant difference was found on the incidence of thrombosis between the two groups (P =0.1187).Conclusion Modified Seldinger technique combined with vascular ultrasonography is efficient and safe for PICC placement for patients with breast cancer undergoing postoperative chemotherapy.
6.Effects of astragalus polysaccharides on lipid metabolism and PPAR-α gene expression in myocardium of diabetic hamsters

Objective This article was designed to observe the effects of astragalus polysaccharides (APS) on glucose and lipid metabolism, and on expressions of proxisome proliferator activated receptors-α (PPAR-α) and its downstream genes in diabetic hamsters cardiomyopathy. Methods Forty-five hamsters were divided into 3 groups randomly: normal control group (15 normal hamsters), diabetic control group [15 streptozotocin (STZ)-induced diabetic hamsters], and astragalus polysaccharides (APS)-therapy group (15 STZ-induced diabetic hamsters administered with APS 2 g/kg per day orally for 10 weeks). The levels of insulin, C-peptide, myocardial enzymes, glycosylated serum protein (GSP) and lipoprotein of all hamsters were measured. The ultrastructure of myocardium was studied, and the gene and protein expressions of PPAR-α, FATP and ACS were also detected by fluorescent quantitative RT-PCR and Western blot. Results It was shown that Compared with DM group, the levels of GSP, myocardial enzymes and lipoprotein of hamsters in APS-therapy group were lower, the myocardial ultrastructure of hamsters in APS-therapy group was well-protected, and the gene and protein expression of PPAR-α, FATP and ACS of hamsters in APS-therapy group were higher. Conclusions APS is partly effective in treating diabetic cardiomyopathy.
8.Abnormal DNA-binding abilities of NF-?B and glucocorticoid receptor in cortex of kindey from rats with Adriamycin-induced nephrosis

Objective To explore the role of transcription factors such as nuclear factor-kappa B(NF-?B) and glucocorticoid receptor(GR) in the pathogenesis of Adriamycin(ADM)-induced nephrosis in rats and the therapeutic effects of dexamethasone(Dex) and cyclosporin(CsA) on these animals.Methods The DNA-binding abilities of NF-?B and GR in cortex of kindey were examined with electrophoretic mobility shift assay(EMSA) and isotopic radioautography on the 7th,14th,21th and 28th day after a single intravenous injection of ADM,and the therapeutic effects of Dex and CsA were estimated.The biochemistry parameters from blood and urine of rats and the urine protein excretion were also measured.Results The NF-?B DNA-binding ability was significantly increased after 7 days and achieved maximum after 28 days(P0.05).Conclusion The DNA-binding ability of NF-?B is abnormally increased and that of GR is decreased in cells from cortex of kindey in Adriamycin-induced nephrotic rats.The NF-?B DNA-binding ability can be inhibited and the urine protein excretion is decreased by the treatment of CsA.

Object To study the chemical constituents in the lipophilic portion of Panax notoginseng (Burk.) F H Chen Methods Through monitoring each fraction based on antimicrobial experiment of Staphylococcus aureus Rosenbach, the constituents were isolated on silica gel column chromatography and preparative TLC, and the chemical structures were identified by means of spectral data (IR, MS, 1HNMR, 13 CNMR, DEPT, HMQC, HMBC) Results Three compounds were isolated from its petroleum ether extract and identified as: panaxynol (Ⅰ), panaxydol (Ⅱ), and ? sitosterol (Ⅲ) Conclusion Compounds Ⅰ, Ⅱ having strong antimicrobial activity against S aureus were obtained from this plant with the yields of 0.01% and 0.033%

Objective Purpose understanding the prevalence of diabetes(DM) of Xuzhou mine workers and impacts on the incidence of risk factors, and discussing the prevention and treatment.Methods From November 2007 to March 2008,1180 Xuzhou mine workers were determinated by height,weight,abdominal circumference,blood pressure,fasting plasma glucose and lipid four(including triglycerides ,total cholesterol,low-density lipoprotein cholesterol, high-density lipoprotein cholesterol), OGTT test.Results DM prevalence rate is 5.25 percent,more men than women;prevalence rate under 40 years old is lower,the prevalence rate from 40 to 50 year-old began to increase, to the top of 50-year-old.Overweight and obesity people took the most part.Conclusion DM is a common diseases among Xuzhou mining area in-service workers with high rates of prevalence,it connected with age,family history, occupation,body mass index(BMI),etc.
