1.Effect of cyclooxygenase-2 inhibitor in anti-leukemia

Cyclooxygenase-2 (COX-2) plays an important role in the development of leukemia.COX-2 inhibitor plays a certain anti-leukemia role in the process.Many studies show that COX-2 inhibitor plays antileukemia role by inhibiting the proliferation of leukemia cell,promoting apoptosis,affecting cell cycle,inhibiting cell metastasis and reversing cell drug resistance and so on.Therefore,the researches about the anti-leukemia mechanisms of COX-2 inhibitor will provide a new target in clinical prevention and treatment of leukemia.
2.Macrophage migration inhibitory factor in lipopolysaccharide-induced peri-implant inflammation of bone marrow mesenchymal stem cells

BACKGROUND:Currently, a large number of studies have confirmed that macrophage migration inhibitory factor plays an important role in a variety of biological activities, such as tumor development. In recent years, it also plays an important role in the inflammatory process, and has achieved a lot of results. However, it is unclear whether and how the macrophage migration inhibitory factor plays a role around the oral implant under oral environment.
OBJECTIVE:To investigate the effect of macrophage migration inhibitory factor on the inflammation of bone marrow mesenchymal stem cels growing around the titanium implant.
METHODS:First, bone marrow mesenchymal stem cels were seeded onto titanium cel culture disks to simulate the peri-implant environment in the mouth, and then, the cels were divided into four groups: control group, without any stimulation; lipopolysaccharide group, lipopolysaccharide-induced inflammation of bone marrow mesenchymal stem cels; non-specific smal interfering RNA (siRNA)+lipopolysaccharide group, non-specific siRNA-transfected and lipopolysaccharide-induced cels; macrophage migration inhibitory factor siRNA+lipopolysaccharide group, cels under the stimulation of lipopolysaccharide were transfected with macrophage migration inhibitory factor.
RESULTS AND CONCLUSION: Using flow cytometry, the cels expressing over 95% CD29 and CD90 as wel as less than 5% CD 45 were selected in the experiment. Cel counting kit-8 test showed that macrophage migration inhibitory factor siRNA+lipopolysaccharide had no influence on the proliferation of bone marrow mesenchymal stem cels. Lipopolysaccharide significantly stimulated the inflammatory reactions of bone marrow mesenchymal stem cels, which was 15-20 times of the control group (P < 0.01). However, compared with the lipopolysaccharide group, the levels of interleukin-1β, interluekin-6 and tumor necrosis factor-α were increased significantly after transfection with macrophage migration inhibitory factor siRNA+lipopolysaccharide stimulation (P < 0.01). These findings indicate that lipopolysaccharide can promote inflammation of bone marrow mesenchymal stem cels around the oral implant, but macrophage migration inhibitory factor siRNA can, to some extent, inhibit the occurrence of inflammation.

[Results] Improve airway inflammation astragalus polysaccharides from activation and inhibition EOS EOS infiltration reduction, regulating Th1/Th2 imbalance, so Th0 cells to Th1 cell differentiation, and inhibiting the release of inflammatory cytokines and other aspects, APS can reduce airway hyper-responsiveness mitigate aspects peribronchial infiltration of eosinophils and airway smooth muscle cells induced byα-SMA expression and other high alleviate airway remodeling. [Conclusion] The influence of astragalus polysaccharide on asthma gene research will lay the basis for study of the mechanism of treatment of asthma, monomer research provides new ideas for traditional Chinese medicine.

6.Methods and nutritional effects of gastroscopic jejunal access for enteral feeding

Objective:To investigate the methods and nutritional effects of endoscopic jejunal ac cess for enteral feeding.Methods:106 cases placed jejunum feeding tube were randomly divided into the observation group and the control group,54 cases in the observation group and 52 cases in the control group.43 patients underwent nasogastric enteral nutrition were as the nasogastric tube group.The traditional endoscopic jejunal access method was used in control group,and the modified endoscopic jejunal access method was used in observation group.The operation time,the success rate of indwelling,complications and nutritional indexes were compared among these three groups.Results:The operative time of the observation group and nasogastric tube group was significantly shorter than that of the control group (P < 0.05).The success rate of indwelling was significantly lower in the control group (P < 0.05).There was no significant difference in the incidence of postoperative complications among these three groups (P > 0.05).After intubation and nutritional treatment in all the groups,hemoglobin,plasma albumin and serum albumin levels were significantly improved (P < 0.05).There were no significant differences in nutritional indexes between the observation group and the control group (P > 0.05).The levels of hemoglobin,plasma albumin and plasma prealbumin in nasogastric tube group were significantly lower than those in the observation group and the control group (P < 0.05).Conclusion:Modified endoscopic jejunal access for enteral feeding can effectively shorten the intubation operation time and improve success rate and nutritional indexes.
7.Expression of Bcl-w in gastric carcinoma and associated clinicopathologic factors

Objective To discuss the meaning and association with prognosis of Bcl-w expression in gastric carcinoma. Methods By Immunohistochemistry( SP) method was used to detect the expression of Bcl-w and MMP-2 in 100 cases of human gastric carcinoma. Results (1) Among 100 cases of stomach carcinoma, Bcl-w and MMP-2 positive expression rates were respectively 69. 0% , 77. 0% ; (2) The Bcl-w and MMP-2 expression in gastric carcinoma was positively associated with tumor size, invasive depth, histology differentiation and lymph node metastasis(P<0.05) ;(3)The expression of Bcl-w was significantly correlated with MMP-2 in gastric carcinomas (P =0.002) ; (4) Five years survival rates and overall survival of Bclw-positive patients were lower than the Bcl-w -negative patients. Conclusions (1)Bcl-w plays an important role in inhibiting apoptosis of gastric cancer and promoting gastric cancer invasion; (2) Bcl-w-positive expression patients have a bad prognosis and Bcl-w expression in gastric carcinoma can be regarded as a prognosis evaluation index.
8.The stable Expression of VEGF_(165) in Mammalian Cells

Objective To investigate the transfection and expression of pcDNA3 1-VEGF 165 plasmid in mammalian cells. Methods pcDNA3 1-VEGF 165 plasmid was transfected into CHO cells by electroporation and the positive clones were screened by G418. The expression of VEGF 165 gene in the transfected CHO cells were detected by RT-PCR, ELISA and western blotting analysis. The biologic activities of VEGF 165 secreted by the transfected CHO cells were measured by Mile's assay and MTT method. Results G418-resistant CHO cell clones were obtained. RT-PCR, ELISA and western blotting analysis showed that there were the transcription and translation of VEGF 165 gene in anti-G418 CHO cells. The mile's assay showed that the expressed VEGF 165 could increase the vessel permeability, and MTT assay proved that it could promote the proliferation of endothelial cells. Conclusion pcDNA3 1-VEGF 165 plasmid could express VEGF 165 with biological activities in mammalian cells.
9.Risk factors of under staging and under grading of patients undergoing radical prostatectomy

Objective To determine the relevant factors of under staging and under grading in patients undergoing radical prostatectomy and to define important predictors of under staging in this population. Methods The clinical T stages and biopsy Gleason scores were compared with the pathological T stages and prostatectomy Gleason scores in 55 patients. Results Among the 55 cases,50 were preoperatively classified as clinical stages T 1~T 2,of whom 21(42%) were postoperatively classified as pathological stages T 3~T 4.Of the 26 cases who preoperatively had the Gleason scores of 2 to 6,11(42%) had postoperatively pathological grades of 7 to 10.Logistic regression analysis revealed that PSA( P =0.0159)at diagnosis and the percentage of positive biopsies of prostate ≥50%( P =0.0013) were significant predictors for preoperative clinical staging lower than postoperative pathological staging. Conclusions The results demonstrate that patients who were classified as clinical stages T 1~T 2 with PSA≥20 ng/ml and percentage of positive biopsies≥50% may have high possibility of under staging.
10.Role of free hemoglobin and its receptor CD163 in the rat atherosclerosis formation

AIM:To investigate the influence of free hemoglobin on the initiation and progression of atherosclerosis and the role of CD163 in this process.METHODS:Thirty male SD rats were randomly divided into three groups:control group(group C),atherosclerosis group(group A),atherosclerosis and hemolysis group(group P).The hemolysis and atherosclerosis animal model was established.The free hemoglobin(FHb)and MDA levels in plasma,(RAAPIs)and intima area/midmembrane area(I/M)of each group were measured.The expressions of CD163 and heme oxygenase-1(HO-1)in atherosclerosis plaques in group A and P were detected and measured by means of immunohistochemistry and Western blotting.RESULTS:Compared with group C,the FHb,MDA,CD163 and HO-1 in group A and group P increased significantly(P0.05).The FHb level in plasma and the expressions of CD163 and HO-1 in atherosclerotic plaques correlated with each other(r=0.526,r=0.498,r=0.653;P
