1.Analysis of VOCs in the headspace of JEOK non-Hodgkin lymphoma cells

Objective To find lymphoma cell markers characteristic of gas for breath monitoring application and provide experimental evidence for the early diagnosis of lymphoma patients,assessing solid phase micro extraction-gas chromatography / mass spectrometry( SPME-GC / MS)is applied to the lymphatic the feasibility of detecting tumor cells in the headspace. To investigate the value of VOCs in the diagnosis of hematologic malignancies. Meth-ods The air samples from the headspace of non-Hodgkin lymphoma cell JEOK,the human lymphoidcell lines and culture medium were collected by syringes,and then determined by means of SPME-GC / MS in order to have a bet-ter understanding of the concentration distributions and changes of VOCs in JEOK cells headspace. Using Mann-Whitney U test,we found the characteristic volatile markers of non-Hodgkin′s lymphoma cells. Results SPME-GC /MS for JEOK cell lines,human lymphocytecell line and a blank incubation were detected headspace analysis,found JEOK cell culture headspace can be detected dimethyl,sulfide,toluene,o-xylene,1,3-Di-tert-butylbenzene,aceto-pheno-ne,dodecane,representing human lymphocytes increased;and alcohol,benzaldehyde,hexanal is signifi-cantly reduced. Conclusion Non-Hodgkin′s lymphoma cell lines can cause flasks JEOK headspace change in the composition of VOCs,increased alkanes and aromatic compounds,aldehydes and reducing alcohol content,these substances may be used as acharacteristic of lymphoma cells markers;SPME-GC / MS as a trace substance detection method can be used to JEOK cell lines VOCs bottle headspace detection training.
2.Effects of Ursolic Acid on Cholesterol Metabolism in Hepatic Cells

Objective To explore the effects and mechanism of ursolic acid ( UA) on cholesterol metabolism in human hepatocellular carcinoma HepG2 and mouse hepatocyte AML-12. Methods HepG2 and AML-12 cells were treated with different concentrations of UA (0,10,20,40 μmol·L-1)for 24 h, then the mRNA and protein expression of cholesterol 7alpha-hydroxylase (CYP7A1) and intracellular cholesterol level was detected by RT-PCR、Western blotting and enzymatic method, respectively. Results Compared with 0 μmol·L-1 UA, 20 μmol·L-1 and 40 μmol·L-1 UA significantly increased the expressions of CYP7A1 mRNA and protein(P<0. 05), and decreased intracellular cholesterol level in HepG2 and AML-12 cells (P<0. 05). Conclusion A certain concentration of UA can reduce the level of cholesterol in HepG2 and AML-12 cells. CYP7A1 may be involved in the regulation process.
3.An Automated On-line Solid Phase Extraction-Liquid Chromatography-Tandem Mass Spectrometric Method for Six Endogenous Phytohormones Analysis in Rice

An automated on-line solid phase extraction-liquid chromatography-tandem mass spectrometry ( on-line SPE-LC-MS/MS ) method for the simultaneous quantification of six endogenous phytohormones including abscisic acid ( ABA ) , indole-3-acetic acid ( IAA ) , salicylic acid ( SA ) , jasmonic acid ( JA ) , indole-3-propionic acid ( IPA) and indole-3-butyric acid ( IBA) in rice tissues was described. Plant samples were extracted with methanol and on-line SPE procedure was performed with C18 SPE cartridges. Analytes were eluted to C18 analytical column with mobile phase and further analyzed by LC-MS/MS. The performance of the method was fully validated. Linearity showed good correlation (R2≥0. 99). Limits of detections (S/N=3) were in the range of 0. 1-0. 8 μg/kg. Recoveries varied between 71. 2% and 126%. The method was rapid and sensitive to determine multiple phytohormones in rice young panicles and the results were cross-validated with the off-line SPE-LC-MS/MS phytohormone analytical method. Finally, the method was applied to monitor the changes of ABA, IAA, SA and JA in wounded rice leaves. The trends were in accord with plant physiological processes.
4.Hemorrheological changes after tourniquet deflation in rabbits

Objective To evaluate the hemorrheological changes after tourniquet deflation.Methods Twenty rabbits were subjected to lower extremity tourniquet inflation for 3 h. Venous blood samples were taken before tourniquet inflation and 2min after tourniquet deflation for measurement of hemorrheological parameters: low-shear viscosity and high-shear viscosity of whole blood; plasma high-shear viscosity , hematocrit; fibrinogen; aggreability,deformability and stiffness of erythrocyte; and blood sedimentation K value. The levels of parameters before tourniquet inflation served as baseline values. 5.Development and application of morphological analysis method in Aspergillus niger fermentation.

Filamentous fungi are widely used in industrial fermentation. Particular fungal morphology acts as a critical index for a successful fermentation. To break the bottleneck of morphological analysis, we have developed a reliable method for fungal morphological analysis. By this method, we can prepare hundreds of pellet samples simultaneously and obtain quantitative morphological information at large scale quickly. This method can largely increase the accuracy and reliability of morphological analysis result. Based on that, the studies of Aspergillus niger morphology under different oxygen supply conditions and shear rate conditions were carried out. As a result, the morphological responding patterns of A. niger morphology to these conditions were quantitatively demonstrated, which laid a solid foundation for the further scale-up.

6.The Protection against +Gz Afforded by Pressure Breathing with Different Pressure Schedules

Objective System of pressure breathing for +Gz (PBG) has been incorporated into service in the high performance fighter aircraft,but there were significant differences among PBG pressure schedules used in different countries.The purpose of this study was to define an optimal pressure schedule in PBG system.Method Five male subjects wearing GZ-2 anti-G suit and medium-sized bladder vest,plus PBG with 1.6,2.4,and 3.2 kPa/G pressure schedules,respectively,were exposed to rapid onset (3.0 G/s) centrifuge +Gz runs.+Gz protection of PBG with each of the three pressure schedules were measured and the subjective ratings were collected.Result The +Gz protection afforded by PBG with 1.60,2.40,and 3.20 kPa/G pressure schedules were 2.00±0.31,2.54±0.32,and 2.44±0.31 G,respectively.Subjective ratings showed that the PBG with 2.40 kPa/G pressure schedule was better than the other two.Conclusion Our data suggest that a PBG pressure schedule of 2.4 kPa/G in PBG system is optimal.It not only assures the anti-G performance of PBG,but also reduces its side effects.

The cytolytic effect of perforin is a mechanism of anti-virus,killing microbial-infected cells and tumor cells.Perforin is a very important non-specific immune factors in fish.In order to understand the function of perforin,the cDNA of grass carp perforin C-terminal peptide was amplified from grass carp liver and kidney cDNA library.It contains a protein kinase C conserved region 2(C2).The cDNA was connected with pET32a,and transformed to expression bacteria DE3.PFP-C was expressed by a prokaryotic expression system and then purified by affinity chromatography.It showed a significant haemolytic activity when tested with rabbit red cells,the optimal pH for haemolytic activity was 7.5,and its haemolytic function dependents on Ca2+ apparently.

Objective To investigate the vision and the eye conditions of the longevous aged 90 years or over Methods The vision was examined by Snellen chart, and the outer eye, anterior segment, fundus and intraocular pressure were examined by slit lamp, funduscope and non contact tonometer in 135 longevous cases Some related examinations were adopted on suspective cases The refractive errors were corrected in 53 cases who were cooperative to examination Results The visions of right eyes were 0 01~1 0 with the average of 0 46?0 50, and the left eyes were 0 03~1 0, with the average of 0 49?0 50. ObjectiveTo investigate the visual condition and causes of blindness and low vision of ov er 60 years old people found by routine physical examination. Methods A total of 892 cases over 60 years old people took a yearly routine examinatio n, such as far and near vision in daily life, anterior segment and fundus of the eye and intraocular pressure. If the vision were under 0 3, the refractive vis ion needs to be corrected. The blindness and low vision patients were selected a nd taken further related examinations to analyse statistically the causes of bli ndness. Results Among 892 cases investigated, blindness together with low vision were 42 cases (4 71%). The morbidity rate of blindness and low vision were 1 01% (9 cases) a nd 3 70% (33 cases) respectively. Diseases caused blindness were macular lesio ns (57 14%), glaucoma(11 91%), cataract (9 52%), retinal diseases, optic nerv e atrophy, keratopathy etc. Refractory eye diseases were obviously more than t he eye diseases which can be treated. The morbidity rate of over 80 years old pe ople were obviously more than that of under 80 years group. Conclusions With ageing, the old people easily suffer from eye diseases and results in bli ndness. For the prevention and treatment of blindness for the elderly, much atte ntion to treatment of eye diseases should not only paid, active training is also important.
