1.Effects of Bone Marrow Mescenchymal Stem Cells Conditioned-Medium on Cortical Neuronal Apoptosis Induced by Hypoxia-Reoxygenation in vitro in Rats

Objective To observe the effects of bone marrow mesenchymal stem cells(BMMSCs) conditioned-medium (B-CM) on apoptosis in cultured cortical neurons after hypoxia-reoxygenation in vitro in rats.Methods BMMSCs were isolated,cultured and expanded,the culture me-dium was substituted by new medium that contain no serum when cells grew to 90% confluence.After cultured for 24 hours,the medium was collected as B-CM.Cortical neurons of mouse cultured for 8 days were divided into three groups:normal control groups,hypoxic group,B-CM disposed groups.Cell viability were detected by 3-(4,5-dimethlthiazd-2-yl)-2,5-diphenyltetra-zoliu bromidel (MTT) methods and the apoptosis of neurons was examined by flow cytometry and telemicroscopy on hypoxia for 0,6 hours and hypoxia-reoxygenation for 24 hours,respectively.Results Compared with control group,hypoxia for 6 hours could decrease cell viability,hypoxia-reoxygenation for 24 hours increased the cell apoptosis ratio remarkably(P
2.A case design and application of flipped classrooms in stomatology

Flipped classrooms have been developed in recent years at home and abroad although there is no systematic application in Chinese higher education institutions. College of Stomatology Shanghai Jiao Tong University has discussed the application prospect of flipped classrooms in medical education and methods of increasing effect and quality by informational means and classroom reform through a case de-sign and application in stomatology, in which performance comparison of the students in flipped classroom and traditional classroom is done as well as recognition of the students to flipped classrooms by a question-naire survey. Still, procedure, experience, supporting conditions and tips on the design and application have been concluded. The practice in classrooms tells us the students accept flipped classrooms positively and scientific design and application may increase teaching result as an effect method. Education management department of higher education institutions and faculty may take advantage of that method with varying degrees.
3.Effect of bisoprolol combination with perindopril and spironolactone on chronic heart failure caused by rheumatic heart disease

Objective To evaluate the combination treatment effects of bisoprolol,perindopril and spirono-lactone on chronic cardiac failure caused by rheumatic heart disease.Methods 92 patients were randomly divided into the control group (n=45) and observation group (n=47).The observation group received combination usage of bisoprolol,perindopril and spironolactone,while the control group was given regular treatment.Cardiac functions were measured in both two groups.Results The left ventricular end diastolic diameter ( LVEDD) and left ventricular end systolic diameter (LVESD) were significantly decreased in the observation group(t=4.217,5.842,P<0.05),while left ventricular ejection fraction (LVEF) was significantly increased compared with the control group (t=3.164,P<0.05).Conclusion The combination treatment effects of bisoprolol,perindopril and spironolactone has the privilege effects on chronic cardiac failure caused by rheumatic heart disease,which mainly improves cardiac remodeling.

5. Preparation, characterization and evaluation of a novel tumor targeting superparamagnetic iron oxide nanoparticles

OBJECTIVE: To improve superparamagnetic iron oxide nanoparticles (SPIO-NP) stability, biocompatibility and tumor-targeting and evaluate their tumor targeting in vitro. METHODS: The folic acid-carboxymethylchitosan-superparamagnetic iron oxide nanoparticles (FA-OCMCS-SPIO-NPs) were synthesized by "three-steps": at first, superparamagnetic oxide iron nanoparticles were synthesized by coprecipitation, then, O-carboxymethyl chitosan and folic acid were successfully immobilized on the surface of SPIO-NPs in turn. The fourier transform infrared spectroscopy, X-Ray diffraction were used to confirm their synthesis, meanwhile, transmission electron microscope (TEM), dynamic light scattering (DLS), Zeta-potential measurement and vibrating sample magnetometry (VSM) were applied to characterize their physicochemical properies. Ferrozine assay and Prussian blue staining were used to evaluate their tumor targeting in vitro. RESULTS: X-Ray diffraction showed the crystalline powder of FA-CMCS-SPIO-NPs agree with the standard Fe3O4, Fourier transform infrared results showed O-carboxymethylchitosan and folic acid were covalently modified on the surface of SPIO-NPs successfully, TEM showed all synthesized SPIO-NPs were almost spherical or ellipsoidal. Their sizes were less then 20 nm, dynamic light scattering (DLS), Zeta-potential results demonstrate the intensity particle size distributions of FA-OCMCS-SPIO-NPs were (41.4±0.132)nm, Zeta potential were (-21.36±15)mV. The surface modification may lead to decrese of magnetisms Ferrozine assay and Prussian blue staining results showed FA-OCMCS-SPIO-NPs had good tumor targeting and the tumor targeting had good relations with the amount of FR on surface of tumor cells. CONCLUSION: FA-OCMCS-SPIO-NPs with strong superparamagnetic property, excellent stability, and good folate receptor targeting is successfully synthesized, which demonstrated the potential for tumor MRI diagnose and therapy.
6. The immune-enhancing effect of Ar-He targeted cryoablation for the treatment of advanced non-small cell lung cancer

Objective: To investigate immune-regulating effects of Ar-He targeted cryoablation for the treatment of the advanced non small cell lung cancer (NSCLC). Methods: One hundred NSCLC patients were enrolled in this study. The serum levels of IL-2, TNF-α and IL-12 were measured by enzyme-linked immunosorbent assay (ELISA) before and 10 days after Ar-He targeted cryoablation. Low-density mononuclear cells (NINC) were separated by Ficoll-Hypaque as effector cells. Human lung cancer SPC-A1 and K562 cells were selected as target cells. The activities of natural killer (NK) cells and cytotoxic T-lymphocytes (CTL) were determined by MTT assay. Results: Of the 100 patients, 3 cases achieved complete remission (CR), 34 cases achieved > 50% remission (PR), 47 cases had stable disease (SD), and 16 cases had progressive disease (PD). The one-year survival rate was 43% and the median survival time to progression was 4 months. The non specific tumor-killing ability of NK cells increased by 83% and the specific tumor-killing activity of CTL increased by 67%. The serum levels of IL-2, TNF-α, and IL-12 were elevated significantly. The positive expression of CD3+ and CD4+ on T cells and the ratio of CD4+/CD8+ also increased. Conclusion: The effectiveness of Ar-He targeted cryoablation for the local treatment of NSCLC could be observed by imaging analysis. Ar-He targeted cryoablation regulates and stimulates self immunological function of NSCLC patients. Ar-He targeted cryoablation provides an effective method especially for old and surgery nontolerant patients.
