1.Preparation and Application of Naringin Sensor Based on Molecularly Imprinting Technique

The sensor of naringin(NG), a non-electroactive substance, was prepared based on the molecular imprinting technique.Using cyclic voltammetry technique (scan rate is 100 mV/s), the naringin imprinting sensitive film, poly-o-aminophenol was coated on the surface of a graphite electrode in the presence of naringin which was considered as the template, and characterized by SEM and X-ray reflective spectrophotometry (XRR).Using K_3Fe(CN)_6 as an electroactive marker, the electrochemical properties of the NG sensor were investigated by CV, electrochemical impedance spectroscopy (EIS), differential pulse voltammmetric and chronoamperometric.The results showed that the imprinted electrode was significantly different from the non imprinted electrode in morphologies and electrochemical properties, and a linear relationship between the peak current and the naringin concentration was found in the range of 6.0 × 10 ~(-5)-1.4 × 10 ~(-4) mol/L with a detec tion limit of 1.6 × 10 ~(-5) mol/L.Moreover, the imprinted electrode exhibited a good selectivity and rapid response to the naringin template molecules, as well as an excellent reproducibility(RSD = 1.8 %, n=5).
2.Overexpression of S3 ribosomal protein gene is involved in drug resistance in K562/DOX cells.

OBJECTIVETo investigate the effect of overexpression of S3 ribosomal protein (S3rp) gene on the resistance of leukemia cell to antitumor drugs.
METHODSBoth sense and antisense cDNA recombinants of S3rp gene were constructed with pcDNA3.1 expression vector. Subsequently, the sense S3rp cDNA recombinant was transfected into K562 cells while the antisense one into K562/DOX cells (a multidrug resistant cell line). In addition, empty pcDNA3.1 vector was transfected into the corresponding cells as negative controls. The chemosensitivity of cells was evaluated by MTT assay.
RESULTSSense S3rp cDNA transfected K562 cells were 5.8 times more resistant to doxorubicin than control cells did, whereas antisense S3rp cDNA transfected K562/DOX cells were 3.2 times less resistant to doxorubicin than control cells did.
CONCLUSIONOverexpression of S3rp gene plays an important role in the development of drug resistance in K562/DOX cells.

3.Expression and the effect of preoperative radiotherapy of ET-1 and PKM2 in rectal carcinoma

Objective To explore the relationship of expression and the effect of preoperative radiotherapy of endothelin-1 (ET-1) and pyruvate kinase M-2 (PKM2) in rectal carcinoma.Methods Immunohistochemical method was used to detect the expression of ET-1 and PKM2 proteins of rectal cancer tissues in 96 cases.The expressions of ET-1 and PKM2 were analyzed with the effect of preoperative radiotherapy in rectal cancer tissue.Results The high expression of ET-1 protein was 59 cases (61.46%).The high expression of PKM2 proteins was 54 cases (56.25%).The high expressions of ET-1 and PKM2 protein were worsen the effect of tumor regressive grade (TRG) than lower expressions of those after preoperative radiotherapy of rectal cancer tissue (P < 0.05).The protein expression of ET-1 and PKM2 were positively correlated (P =0.006).Conclusions The high expressed ET-1 and PKM2 proteins in rectal cancer are closely related to preoperative radiotherapy resistance.ET-1 and PKM2 proteins are expected to become new targets of radiotherapy sensitivity and radiotherapy sensitization of rectal cancer.

Background and purpose:Neoadjuvant chemotherapy is one of the hot studied area in breast cancer research.Our aim was to explore the relationship between the clinical effect,pathological changes and the neoadjuvant chemotherapy containing THP and docetaxel on breast cancer.Methods:The expression of ER, PR and CerbB-2 in breast cancer tissue of patients who had received neoadjuvant chemotherapy were detected by immunohistochemistry method.The relationship of age,ER,PR,CerbB-2,pathological stage and axillary lymph node metastasis with pathological complete response(pCR) was analysed.Results:For the patients after neoadjuvant chemotherapy,the pCR of the positive ER(0%),PR(9.38%) was lower than that of negative ER(20.27),PR(90.63%) respectively.The pCR of the positive CerbB-2(33.33%) was higher than that of negative CerbB-2(9.74%,P

Objective To investigate whether the spores from mushroom antigen can cause the allergic pneumonia and manufacture allergic animal model in the C57BL/_6 mouse.Methods Aged 6 weeks old,weight 25-30 g C57BL/_6 mice were collected.In the mouse tail injection compound of spore antigen and the Freud′s adjuvant.Then pours into through the trachea the antigen once a week.The mice were divided into 4 groups.Group A was the normal mouse,Group B was given Freud′s adjuvant(the same method)to determine whether there was affect to the mouse.Group C and D were injected spore antigen 2 and 4 times.When the antigen sensitization finished 1 week later group C and D were completely divided 2 groups,among them one group was inhalation 1.5% spore antigen and induce the acute response.Six hours later the bronchoalveolar lavage fluid(BALF) was collected to observe cell change,and excise the lung tissue to manufacture the pathology specimen,another group had not been induced the acute response and collect the BALF and to exsise the lung tissue directly.Group B were inhalted saline later to collect the BALF and the lung tissue.In the mouse blood serun,enzyme linked immunosorbent assay(ELISA) was used to mensurate antigen specific IgG.Results In group C and D,antigen specific IgG significantly inhanced than that in group A and B(all P
6.Study on the genetiey relationship between Chinese and Korean medicinal materials of niuxi with the method of RAPD.

OBJECTIVETo study the hereditary relationship between Chinese and Korean medicinal materials of Niuxi achyranthis root.
METHODTen samples of four kinds of Niuxi was studied with the method of RAPD.
RESULTExpanded product showed total record of 100 spectrum bands, which proved that the hereditary gap between Korean self-produced A. japonica and A. bidentata is smallest.
CONCLUSIONKorean self-produced A. japonica is near to A. Sidentata.

7.Identification of multidrug resistance related genes in leukemia by suppression subtractive hybridization.

OBJECTIVETo clone and screen genes related to multidrug resistance (MDR) in leukemia.
METHODSSuppression subtractive hybridization (SSH) was performed to profile differentially expressed genes between a MDR leukemia cell line (K562/DOX, as tester) and its parent cell line (K562, as driver). Reverse Northern dot blot was carried out to further screen the subtracted cDNA library. The overexpressed cDNA fragments in K562/DOX cells were sequenced and compared with known genes in Genbank. RT-PCR and Northern blot were employed to confirm the differential expression of some identified genes.
RESULTSEleven genes were identified being overexpressed in K562/DOX, including S3 ribosomal protein (S3rp) gene, NADH dehydrogenase subunit 2 (ND2) gene and My023 gene, which have not been reported to be related to MDR in cancer.
CONCLUSIONSeveral genes, which might be involved in MDR were identified, indicating novel mechanisms of MDR in leukemia.

8. Effects of reconstruction with unicondylar osteoarticular allografts with or without prosthesis for bone tumors around knee joint

Objective:
To investigate the survival rate, function outcomes, and complications after using unicondylar osteoarticular allografts with or without prosthesis to reconstruct the knee joint for tumors located in distal femoral or proximal tibial uni-condyle.
Methods:
Twenty-two patients who underwent unicondylar osteoarticular allografts with or without prosthesis composite reconstructions from January 2007 to December 2015 in Department of Orthopaedic Surgery of Xi Jing Hospital, the Fourth Military Medical University were retrospectively reviewed. There were 14 males and 8 females and the mean age was 35 years(8-65 years). There were 12 malignent tumors and 10 aggressive benign tumors. The tumors were located in distal femur in 14 cases and proximal tibia in 8 cases. After tumor excision, the distal femur was reconstructed with unicondylar osteoallograft-prosthesis composite, and proximal tibial plate was reconstructed with unicondylar osteoarticular allograft with the help of computer-assisted navigation system. Function and radiograph were documented according to the Musculoskeletal Tumor Society (MSTS) functional scoring system and the International Society of Limb Salvage (ISOLS) radiographic scoring system. The median follow-up time was 60 months (5-116 months).
Results:
At the latest follow-up, 2 patients had amputation owing to local recurrence in 12 malignant tumors. Three patients had pulmonary metastasis and 1 patient died another 2 alive with disease. Kaplan-Meier analysis indicated that the disease-free survival rate was 73%. There was no recurrence and metastasis in 10 patients with giant cell tumor. The average MSTS score was 26 points and the radiographic score was 78%-94%(average 90%). The complications included superficial infection in 1 patient and screw broken in 1 patient. There was no broken or collapse allograft in all composite reconstruction patients but 6 cases in allograft reconstruction.
Conclusions
Unicondylar osteoarticular allografts or prosthesis composite was feasible reconstruction for tumors in distal femoral uni-condyle. It could provide good functional outcomes and also prevent joint degeneration. Similarly, allograft reconstruction was also a reliable technique for proximal tibial defect.
9.Gene expression of collagen types IX and X in the lumbar disc.

OBJECTIVETo study gene expression of collagen types IX and X in human lumbar intervertebral discs during aging and degeneration and to explore the role of collagen types IX and X in disc degeneration.
METHODSFetal, adult and pathologic specimens were subjected to in situ hybridization with cDNA probes to investigate mRNA-expressions of types IX and X collagen gene.
RESULTSIn fetal intervertebral discs, positive mRNA hybridization signals of type IX collagen were concentrated in the nucleus pulposus and the inner layer of anulus fibrosus. Interstitial matrix of the nucleus pulposus also showed positive type X collagen staining. Positive mRNA hybridization signals of types IX and X were not detected in the middle and outer layers of anulus fibrosus. In adult specimens, expression of type IX collagen mRNA was markedly decreased. No hybridization signals of type X collagen was observed. As for pathological specimens, there was no gene expression of type IX collagen. In severe degenerated discs from adults, there were focal positive expressions of type X collagen.
CONCLUSIONSObvious changes of collagen gene expression occur with aging. Expression of type IX collagen decreases in adult and pathological discs. Results of type X collagen expression suggest that type X collagen is expressed only in older adult and senile discs (i.e., when disc degeneration has already reached a terminal stage), indicating the terminal stage of degeneration.

10.Correlation of blood concentration of tacrolimus with serum cystatin C in renal transplant recipients and effect of tacrolimus on glucose and lipid metabolism.

OBJECTIVETo investigate the correlation between blood concentrations of tacrolimus (FK506) and cystatin C (Cys C) and the effect of FK506 on glycolipid metabolism in renal transplant recipients.
METHODSA total of 325 patients receiving renal transplantation between August, 2014 and September, 2015 in Nanfang Hospital were divided into 4 groups according to the postoperative time (1 month group, 1-3 months group, 4-6 months group, and 7-12 months group). FK506 blood trough concentration was measured at the time of postoperative follow-up, and creatinine (Scr) and Cys C levels were also detected. Results Plasma FK506 concentration decreased with age in the recipients and showed a positive correlation with Cys C (r=0.985, P=0.015) but no obvious correlation with Scr (r=0.259, P=0.741). FK506 had no effect on blood glucose (5.53-5.59 mmol CONCLUSIONFK506 does not affect the level of glycolipid metabolism in patients after renal transplantation. Cys C is positively related to blood concentration of FK506 in the renal transplantation recipients. The rational use of FK506 can improve the effectiveness and safety of the treatment in the recipients.
