1.Effect of electrical stimulus at a low physiological frequency on expression of myosin heavy chain in genioglossus of rabbits with chronic hypoxia

Objective To study the effect of chronic electrical stimulus at a low physiological frequency on the expression of myosin heavy chain (MHC) in genioglossus of rabbits with chronic hypoxia.Methods Twenty-four adult rabbits,weighing 2.3?0.1kg,were randomized into control group (A),chronic hypoxia group (B),10 Hz electrical stimulus group (C),and 10+40 Hz electrical stimulus group (D).Rabbits in groups C and D received electrical stimulus of genioglossus at a frequency of 10 Hz and 10+40 Hz,respectively,while those in group B received no electrical stimulus,after they were placed in a hypobaric hypoxia cabin,8 h a day for 5 weeks,with free access to food and water.Expression of MHC in genioglossus of rabbits in the 4 groups was detected by SDS-PAGE and semi-quantitative inverse transcription RT-PCR,respectively.Results The expression level of MHCⅠand MHCⅠmRNA was significantly lower while that of MHCⅡa was significantly higher in group B than in group A [(0.69?0.07)% vs (17.48?4.00)%,(0.69?0.07)% vs (1.05?0.06)%,and (79.89?5.09)% vs (73.33?4.17)%,P
2.Median effective target effect-site concentration of sufentanil when intraoperative wake-up test was successful in patients undergoing scoliosis surgery

Objective To determine the median effective target effect-site concentration (EC50) of sufentanil when intraoperative wake-up test was successful in patients undergoing scoliosis surgery.Methods Ninety ASA Ⅰ or Ⅱ patients,aged 18-64 yr,with body mass index < 30 kg/m2,scheduled for scoliosis surgery under sevoflurane and sufentanil anesthesia,were randomly divided into Ⅰ-Ⅵ groups (n =15 each).Anesthesia was induced with sufentanil,etomidate and cisatracurium,and maintained with sevoflurane,sufentanil and cisatracurium.The patients were tracheal intubated and mechanically ventilated.Before the wake-up test,infusion of cisatracurium was stopped and the end-tidal concentration of sevoflurane was adjusted to 0.The EC50 was determined by the K(a)rber method.The target effect-site concentration of sufentanil was set at 0.19 ng/ml in group Ⅰ,0.18 ng/ml in group Ⅱ and gradually decreased in decrements of 0.01 ng/ml.The wake-up test was performed 5 min later.The EC50 and 95 % confidence interval of sufentanil were calculated by the K(a)rber method.Results EC50 of sufentanil obtained was 0.164 ng/ml and 95% confidence interval of sufentanil obtained was 0.157-0.172 ng/ml when the wake-up test was successful.Conclusion The EC50 of sufentanil is 0.164 ng/ml when the intraoperative wake-up test is successful in the patients undergoing scoliosis surgery.
3.Simultaneous Determination of 5 Components in Lianqiao Baidu Pill by HPLC

OBJECTIVE:To establish a method for the simultaneous determination of chlorogenic acid,paeoniflorin,ferulic ac-id,berberine hydrochloride and baicalin in Lianqiao baidu pill. METHODS:HPLC was performed on the column of Agilent Eclipse XDB-C18 with mobile phase of 0.5% phosphoric acid(triethylamine adjusted to pH 3.0)-methanol(gradient elution)at a flow rate of 1.0 ml/min,the column temperature was 30 ℃,the detection wavelength was 230 nm(for paeoniflorin and berberine hydrochlo-ride),324 nm(for chlorogenic acid and ferulic acid)and 278 nm(for baicalin),and the injection volume was 5 μl. RESULTS:The linear range was 1.884-30.144 μg/ml for chlorogenic acid(r=0.999 9),0.392-6.272 μg/ml for paeoniflorin(r=0.999 7), 0.102-1.632 μg/ml for ferulic acid(r=0.999 7),1.326-21.216 μg/ml for berberine hydrochloride(r=0.999 8) and 1.95-31.2 μg/ml for baicalin(r=0.999 9),respectively;RSDs of precision,stability and reproducibility tests were lower than 2.0%;recoveries were 99.58%-102.47%(RSD=0.93%,n=9),99.21%-102.18%(RSD=0.90%,n=9),98.28%-101.23%(RSD=0.86%,n=9), 99.66%-101.84%(RSD=0.82%,n=9)and 99.18%-101.05%(RSD=0.62%,n=9). CONCLUSIONS:The method is accurate,re-liable,simple and fast,and can be used for the simultaneous determination of chlorogenic acid,paeoniflorin,ferulic acid,berber-ine hydrochloride and baicalin Lianqiao baidu pill.

5.Event-related potentials of impairment of working memory in patients with amnestic mild cognitive impairment.

Objective To find a sensitive index and to investigate the working memory impairment of patients with amnestic mild cognitive impairment.Methods Fifteen patients with amnestic mild cognitive impairment,15 healthy matching aging controls were performed a matching-to-sample task while event-related potential(ERP),reaction time and correct rate were recorded.Subjects were required to press a button in the match condition and another button in the conflict condition.Results In the matching condition,there was no difference between the two groups in distribution(parietal lobe),peak latency and amplitude of P300(F_ 1,28 =1.0324,P=0.3183;F_ 2,42 =0.543,P=0.585).In the conflict condition,the distribution of N270 was fore head,and its latency of patient group were more delayed than the aging group(F_ 1,28 =25.3264,P=0.000),but its amplitude showed no significant changes(F_ 1,28 =0.507,P=0.482).The result of brain mapping showed same change.Conclusion The N270 component is more sensitive than P300 to reflect the central executive function impairment of working memory in patients with amnestic mild cognitive impairment.
6.Experimental study on biodistribution and PET imaging of 3′-deoxy-3′-~(18)F-fluorothymidine in murine model of lung carcinoma

Objective To investigate the biodistribution and positron emission tomography (PET) imaging of 3′-deoxy-3′-~ 18 F-fluorothymidine (~ 18 F-FLT) in a murine model of pulmonary carcinoma, and to evaluate the use of ~ 18 F-FLT as a new PET tracer for diagnosis of pulmonary malignant tumor. Methods 40 T739 mice bearing the pulmonary adenocarcinoma were randomly divided into five groups according to different tracers and time after their injection (n=8/group). The biodistribution of mice for ~ 18 F-FLT was measured with well-gamma detector at 30min, 60min, 90min, 120min after injection via the tail veins. The biodistribution of mice for ~ 18 F-FDG was examined at 60min after injection as controls. In addition, the PET imaging of mice was performed using two tracers. Results In the biodistribution study of ~ 18 F-FLT, considerable radioactive uptake in tumor was observed, and high radioactivity was showed in the kidney and spleen. The T/NT ratios of tumor/blood, tumor/muscle and tumor/lung was all above 2.0. The tumor PET images with ~ 18 F-FLT were clear, as well. Conclusions The uptake of ~ 18 F-FLT in pulmonary adenocarcinoma is higher than that in normal tissues, thus the pulmonary neoplasm could be identified accurately with PET imaging. Our preliminary study of ~ 18 F-FLT in lung carcinoma xenografts is satisfactory, and it provides a basis for further clinical study.
7.Effects of ATRA on interacellular communication function and connexin 43 expressions in acute leukemia bone marrow stremal cells

Objective To observe the effects of all-tram retinoic acid(ATRA)on intercellular communication function and connexin 43 expression in acute leukemia bone marrow stromal cells(ALBMSCs).Methods ALBMSCs were cultured in vitro,and the ATRA(1?10~(-5)mol/L)was added after serial subcultivation.The expression level of Cx43 was detected by immunochemistry and flow cytometry, and the gap junction intercellular communication(GJIC)was examined by using cell scarification with dye transfer(CSDT)technique.Re- sults Before and after addition of ATRA,the positive rates of Cx43 expression as detected by immunochemistry in ALBMSCs were(47.2?2.04)% and(54.5?5.86)%,respectively.The positive rates of Cx43 expression in ALBMSCs as detected by flow cytometry before and after the addition of ATRA were(38.75?23.95)% and(49.5?5.46)%,respectively.The expression of Cx43 in ALBMSCs after adding ATRA was significantly higher than that before the addition of ATRA(P

9.Effect of nanohydroxyapatite/polyamide 66 composite on the vertebral structure and height

BACKGROUND:Nanohydroxyapatite/polyamide 66 composite is very close to the human cortical bone in terms of mechanical properties, and has osteogenic activity, which is a safe and reliable bone material. OBJECTIVE:To investigate the effect of nanohydroxyapatite/polyamide 66 composite bioactive supporting material on the restoration of vertebral structure and height. METHODS: Totaly 177 patients with spinal diseases, 116 males and 61 females, aged 17-81 years, were enroled, including 97 cases of spinal fractures, 5 cases of primary tumors of the spine, 17 cases of spinal tuberculosis and 58 cases of cervical spondylosis. The nanohydroxyapatite/polyamide 66 composite bioactive supporting body was filed into the bone grafts of patients and then subjected to anterior decompression and internal fixation. After 36 months of folow-up, imaging analysis, Frankel spinal cord injury classification, bone graft fusion, visual analog scale scores,Short Form 36 and Japanese Orthopedic Association scores were evaluated. RESULTS AND CONCLUSION:During the folow-up, except one patient with cervical spine fracture appeared to have slight supporting body displacement, there was no supporting body prolapse or rupture phenomena. The bone graft fusion rate was 96.0%, the average supporting body sinking distance was 1.7 mm. After treatment, 97 patients with spinal fractures had different degree of improvement in the neurological function (P < 0.05); the visual analog scale scores, Short Form 36 and Japanese Orthopedic Association scores were improved significantly in al the 177 patients compared with before treatment (P < 0.05). These results demonstrate that the use of nanohydroxyapatite/polyamide 66 composite bioactive supporting material for spinal reconstruction can improve the bone fusion rate, and restore the vertebral structure and height effectively.
