2.The effect and mechanism of epithelial growth factor receptor (EGFR)in inducing vasculogenic mimicry formation

Objective To investigate the effect and mechanism of EGFR in inducing vasculogenic mimicry (VM)formation.Methods Immunohistochemistry was used to detect the expression of EGFR protein and CD34-PAS double staining was used to detect the VM in gliomas paraffin blocks.The migration and VM formation of U87 cell in hypoxic and normoxic groups were detected by transwell and three-dimensional culture.Then we used Western blot to detect the expressions of EGFR,AKT and PI3K protein.Results The positive rate of EGFR expression in VM positive patients was 94.4%,which was significantly higher than that in VM negative patients (71.8%).In hypoxic group,the abilities of migration and VM formation were significantly higher those in normoxic group.The expressions of EGFR,AKT and PI3K were higher in hypoxic group.Conclusion VM formation can be induced by the activation of EGFR/AKT/PI3K signaling pathway.EGFR is very important for VM formation.
3.Effect of Home-based Rehabilitation on Cerebral Palsy

Objective To explore the effect of home-based rehabilitation on cerebral palsy.Methods 60 children with cerebral palsy were assigned to hospital-based rehabilitation group and hospital-home-based rehabilitation group and treated for 1 year.Every child was evaluated before and after the treatment with Bayley Scales of Infant Development(BSID).The Mental Development Index(MDI) and Physical Development Index(PDI) were compared between these two groups.Results After one year,the MDI and PDI improved in both groups(P<0.05),while those of the hospital-home-based rehabilitation group was better than of the hospital-based rehabilitation group(P<0.05).Conclusion Home-based rehabilitation plays an important role in the rehabilitation of cerebral palsy children.

7.Effects of Goserelin on Vascular Growth Factor and Immune Function of Rats with Prostatic Hyperplasia

OBJECTIVE:To study the effects of goserelin on vascular growth factor and immune function of rats with prostatic hyperplasia. METHODS:Rats were selected to establish prostatic hyperplasia model and randomly divided into model group,gose-relin low-dose,medium-dose and high-dose groups (0.4,0.8,1.2 mg/kg);normal rats were selected as normal control group, with 10 rats in each group. Normal control group and model group were given normal saline intragastrically,and goserelin groups were given relevant dose of drugs intragastrically,once a day,for consecutive 25 days. The prostate volume,wet weight,prostatic index of rats were detected as well as positive cell area of VEGF,TGF-β1,FGF,CD4 and CD8 in prostate tissue. RESULTS:Compared with normal control group,prostate volume,wet weight,prostatic index,positive cell area of VEGF,TGF-β1,FGF, CD4 and CD8 were all increased in model group (P<0.05). Compared with model group,the above indexes of goserelin groups were all improved (P<0.05),especially those in medium-dose and high-dose groups were better than in low-dose group (P<0.05);there was no statistical significance between medium-dose group and high-dose group(P>0.05). CONCLUSIONS:Gosere-lin can relieve prostatic hyperplasia of rats,reduce the expression of VEGF in prostate tissue and regulate immune function.

Objective To evaluate the therapeutic effect of botulinum toxin A in conjunction with Wuidatechniques in the treatment of lower limbs spasticity in children with cerebral palsy. Methods Forty patients weredivided randomly into a treatment group and a control group. The 18 patients in the treatment group were treatedwith Botulinum toxin A injection and Wuida rehabilitation techniques, the other 22 patients with Wuida techniquessolely served as control. 10.Adenosine deaminase deficiency associated severe combined immunodeficiency with disseminated varicella infection after vaccination: a case report.

OBJECTIVETo enhance the knowledge of adenosine deaminase (ADA) deficiency associated severe combined immunodeficiency (SCID) with disseminated varicella infection after vaccination.
METHODSWith case report and review of literature, the background knowledge, clinical and laboratory findings, diagnosis and treatment of ADA-deficient SCID were discussed.
RESULTSThe patient had the condition with failure to thrive. The main complaint was more than three weeks of fever and rash. He had received the live attenuated Oka strain varicella vaccination approximately two weeks before the onset of rash. Varicella infection was confirmed with direct immunofluorescence assay. The patient had mild leukopenia, with 3% lymphocytes. The initial immunologic workup included decreased IgG, IgM and IgA, abnormal expanded lymphocyte enumeration which confirmed the reduction of CD3, CD4, CD8, CD19 and CD56. Enzyme testing for ADA activity showed remarkably low level in the hemolysate, as well as increased levels of deoxyadenosine nucleotides.
CONCLUSIONADA-deficient SCID has some characteristic clinical and laboratory findings. Management options for ADA-deficient SCID include hematopoietic stem cell transplantation, enzyme replacement therapy and gene therapy. Immunodeficiency should be considered in children with severe failure-to-thrive. Live vaccine administration should be avoided in patients with immunodeficiency.
