1.A study about the association of detoxication gene GSTM1 polymorphism and the susceptibility to aflatoxin B1-related hepatocellular carcinoma.

OBJECTIVESTo investigate the association between susceptibility to aflatoxin B1(AFB1)-related hepatocellular carcinoma (HCC) and the polymorphism of detoxication gene GSTM1.
METHODSThe peripheral white blood cell DNA samples were obtained from all the subjects including 140 HCC cases and 536 controls from an AFB1 high risk area in Guangxi province. The GSTM1 polymorphism was detected using PCR technique.
RESULTS(1) The GSTM1-present was associated with a decreased HCC risk. The GSTM1-null was associated with an increased HCC risk [adjusted OR (95% CI)= 2.07 (1.20-3.57)]. (2) In the cohorts of both low/median and high exposure levels of AFB1, GSTM1-null genotype was associated with a conspicuous significantly increased risk for HCC [adjusted OR (95% CI) = 1.92 (0.92-4.00) and 1.80 (0.77-4.17)].
CONCLUSIONThe results suggest that genetic polymorphism of GSTM1 was susceptible to HCC and individuals who are GSTM1-null have an increased risk of developing HCC. There is evidence of interaction between GSTM1 polymorphism and AFB1 exposure, especially with low/median degrees of AFB1 exposure.

2.Study on the detoxication gene gstM1-gstT1-null and susceptibility to aflatoxin B1 related hepatocellular carcinoma in Guangxi.

OBJECTIVETo study the association between susceptibility to aflatoxin B1 (AFB1)-related hepatocellular carcinoma(HCC) and the null genotypes of detoxication gene gstM1 and gstT1.

RESULTS(1) gstM1- and gstT1-present were associated with decreasing risk of HCC. gstM1- and gstT1-null were associated with the increasing risk of HCC [adjusted OR (95 % CI) = 2.07 (1.20-3.57) and 1.44 (0.85-2.45), respectively]; (2) The appearance of both gstM1- and gstT1-null genotypes were more susceptible to HCC than either one of them(adjusted OR and 95% CI are 2.43 and (1.19-4.97); (3) From low/median to high level of AFB1 exposure, both gstM1- and gstTl-null genotypes were associated with significantly conspicuous increasing risk of HCC [adjusted OR(95% CI) = 12.76(5.38-30.24) and 7.82(3.61-16.90) respectively].
CONCLUSIONIt was suggested that: genetic polymorphisms of gstM1 and gstT1 were susceptible to HCC; individuals who were gstM1- or gstT1-null would have an increasing risk of developing HCC while individuals with both nulls were more susceptible. There was evidence of interaction between gstM1- and gstT1-null and the level of AFB1 exposure which was associated with the increasing risk of HCC.

3.Ultraviolet Mutation Breeding Aspect of the Biological Control of Postharvest Diseases by Kloeckera apiculata in Citrus Fruit

A series of experiments were conducted to study the mutation of Kloechera apiculata by many kinds of treatments such as UV and UV+LiCl.The optimal dosage disposal was determined:15 W 30 cm under Ultraviolet irradiation for 20 s,UV+LiCl under Ultraviolet irradiation for 20 s and added LiCl 0.3% (w/v).One strain(UV20-13)which had obvious physiological characteristic was obtained.the incidence of blue and green mold of citrus was reduced by 25.56%and 10.00%in vivo experiment after 7 days respectively. The strain UV20-13 was tested by the experiments of subculture and dynamic growth,and the results showed that the strain UV20-13 was better than K.apiculata in the growth characteristics,and it did not appear retrogression,reversion mutation ect after subculturing 10 generations.Therefore the strain UV20-13 had genetic stability.
4.Relationship between Resting Heart Rate and Coronary Syndrome

Objective To investigate the relationship between resting heart rate (RHR) and coronary heart disease (CHD) and to explore the value of RHR in predicting the occurrence of acute coronary syndrome. Methods 445 patients with CHD were divided into stable angina group and acute coronary syndrome group. RHR, risk factors for coronary heart disease and their correlation were analyzed. Results RHR was higher in the acute coronary syndrome group than in the stable angina group (P<0.01). Logistic regression analysis showed that RHR (OR =1.052, 95% CI: 1.009~1.097, P=0.017), systolic blood pressure (OR=1.027, 95% CI: 1.003~1.053, P=0.031) and hyperglycemia (OR=2.743, 95% CI: 1.207~6.233, P=0.016) were independent risk factors for acute coronary syndrome. Conclusion RHR is an independent risk factor for incidence of acute coronary syndrome.
5.Clinical study of complete dentures supported by two endosteal magnetic-attachment implants.

OBJECTIVETo evaluate treatment outcomes of mandibular complete denture supported by two endosteal magnetic-attachment implants in edentulous patients with severe mandibular alveolar ridge absorption.
METHODSEight edentulous patients with severe mandibular alveolar ridge absorption were chosen. Each of them was treated with a complete denture supported by two CDIC implants. The implants were inserted at the areas of two the first premolars respectively. After five months, two magnets were adhered to the inside of the complete dentures. Retention, masticatory efficiency and maximal masticatory force were measured before and after the magnets were adhered and six months later.
RESULTSStatistical analysis revealed that retention, masticatory efficiency and maximal masticatory force were all increased after the magnets were adhered (P < 0.01) and six months later (P < 0.01).
CONCLUSIONRetention, maximal masticatory force and masticatory efficiency were significantly increased after the endosteal magnetic attachment implants were applied. It is concluded that mandibular complete denture supported by two endosteal magnetic attachment implants is an effective method for severe alveolar ridge absorption cases.

6.Association of the Thr241Met polymorphism of DNA repair gene XRCC3 with genetic susceptibility to AFB1-related hepatocellular carcinoma in Guangxi population.

OBJECTIVETo explore the association of the Thr241Met polymorphism of X-ray cross-complementing group 3 (XRCC3) gene with genetic susceptibility to aflatoxin B1(AFB-1)-related hepatocellular carcinoma (HCC)in Guangxi population.
METHODSWe conducted a hospital-based case-control study, including 257 HCC cases and 711 controls without cancers or liver diseases. The XRCC3 Thr241Met polymorphism was analyzed by PCR.
RESULTSThe XRCC3 genotypes XRCC3-Thr/Met or XRCC3-Met/Met were related with an elevated risk of HCC. The risk of HCC was associated with the number of mutant Met copies (adjusted OR were 2.20 and 8.56 for XRCC3-Thr/Met and Met/Met, respectively); moreover, there seemed to be combined effects for HCC risk between the variant genotypes and AFB1-DNA adduct levels from peripheral blood leukocytes (adjusted OR was 2.34 to 20.44, P < 0.01).
CONCLUSIONThese results suggested that XRCC3 polymorphism may be associated with the risk of AFB1- related HCC among the Guangxi population, and interacts with AFB1 exposure in the development of HCC induced by AFB1.

7.Expression of cyclin A in childhood acute leukemia and its significance.

OBJECTIVETo investigate the expression of cyclin A protein in childhood acute leukemia (AL) and its significance.
METHODSBy using Western blotting analysis, cyclin A protein in bone marrow mononuclear cells from 47 newly diagnosed AL children and 33 non-hematological malignancy children was detected.
RESULTSThe expression of cyclin A in AL group (0.38 +/- 0.20) was higher than that in control group (0.03 +/- 0.15) (P < 0.01). The expression of cyclin A in high risk acute lymphocyte leukemia (ALL) group (HR-ALL) (0.62 +/- 0.38) was higher than that in standard risk ALL group (SR-ALL) (0.33 +/- 0.33) (P < 0.05). The expression of cyclin A in WBC > or = 50 x 10(9)/L group and in WBC < 50 x 10(9)/L group was (0.64 +/- 0.36) and (0.39 +/- 0.38), respectively (P < 0.05). Eight (44.4%) out of 18 patients with positive cyclin A expression achieved complete remission (CR). The CR rate was lower than that of patients with negative cyclin A expression (100%) (P < 0.01).
CONCLUSIONSThe higher expression of cyclin A may predict a poor prognosis for childhood ALL.

8.Effects of acupuncture on body mass index and waist-hip ratio in the patient of simple obesity.

OBJECTIVETo observe the therapeutic effect of acupuncture for slimming.
METHODSEighty cases of simple obesity were randomly divided into an acupuncture group and a medication group, 40 cases in each group. The acupuncture group were treated with body acupuncture, electroacupuncture and auricular acupoint taping and pressing, with Tianshu (ST 25), Guanyuan (CV 4), Sanyinjiao (SP 6) etc. selected for body acupuncture, and auricular acupoints Shenmen, Neifenmi (endocrine), Pi (spleen) etc. selected for ear acupoint taping and pressing. And the medication group were treated with oral administration of Sibutramine. Body weight, body mass index, waist and hip circumferences and waist-hip ratio were determined before and after treatment to evaluate therapeutic effect of slimming.
RESULTSThe total effective rate of 87.5% in the acupuncture group was similar to 82.5% in the medication group (P > 0.05). After treatment, body weight, body mass index, waist and hip circumferences and waist-hip ratio significantly decreased in the two groups, but the improvement of waist circumferences and waist-hip ratio in the acupuncture group was better than that in the medication group (P < 0. 05).
CONCLUSIONAcupuncture has a definite therapeutic effect in slimming and it can effectively improve body weight, body mass index, waist and hip circumferences and waist-hip ratio in the patient of simple obesity.

10.Improvment effect of metformin on ameliorates hyperglycemia induced by lipopolysaccharide in diet induced obese mice

Objective To observe the impact of porphyromonas gingivalis (PG) lipopolysaccharide (LPS) on blood glucose of high-fat (HF) diet-induced obese (DIO) mice and the antagonistic effect of metformin on diabetes model mice.Methods The C57 mice were divided into 8 groups(n =6-8 in each group).Apart from the normal control,mice were fed with HF diet for 16 weeks (including 3 groups) or 21 weeks (including 3 groups) for modeling.Besides the HF control group,the mice of diabetes model group and metformin group were given PG LPS 100 μg · kg-1 with intraperitoneal injection and simultaneously administrated with 100 mg · kg-1 metformin by gavage daily for 3 weeks,respectively.Blood glucose was measured with rapid glucometer.The plasma insulin and tumor necrosis factor-α(TNF-α) were tested by enzyme linked immunosorbent assay.The expression of AMP-activated protein kinase (AMPK) pathway protein in livers were detected by Western blotting.Results In 21 weeks DIO mice,the blood glucose levels in the HF control group,diabetes model group and metformin group were (6.46±0.31),(7.82 ±0.35),(6.23 ± 0.21) mmol · L-1,respectively.Comparison between HF control group and diabetes model group,the difference was significant (P < 0.05);comparison between diabetes model group and metformin group,the difference was significant (P < 0.01).Expression of AMPK-phosphorylation status (p-AMPK)in the diabetes model group and metformin group were 1.03 ± 0.04,1.62 ± 0.21,respectively.Comparison between diabetes model group and metformin group,the difference was significant(P <0.05).Conclusion Metformin is a preferred therapeutic agent for hyperglycemic patients with obesity and periodontitis.
