Objective To investigate the effect of endoplasmic reticulum stress in hyperlipidemia rats with acute pancreatitis(AP).Methods Forty-eight SD rats were divided into AP group(n=18,in- duced by injection of 40?tg/kg caerulein in normal rats twice with 2 hrs interval),normal control(n=6), byperlipidemia group(n=6,fed with a high-fat diet for 8 weeks)and hyperlipidemia AP group(n=18,in- duced by injection of 40?g/kg caerulein in hyperlipidemia rats twice with 2 hrs interval).The rats were sacrificed at 9,12,24 hrs(6 of each),respectively.Serum level of amylase was tested and the pathologi- cal changes of the pancreatic tissues were observed.The index of pancreatic apoptosis was assessed by TUNEL method.The expression of GRP78/Bip(glucose regulated protein)protein was determined by immunochemistry,the endoplasmic reticulum stress related molecules of XBP-1 splicing(X box binding protein),CHOP/GADD153(C/EBP-homologous protein or growth arrest and DNA-damage-inducible gene 153),caspase-12 were analyzed by RT-PCR.The dynamic expressions of GRP78/Bip and caspase- 12 were determine,d by Western blot.Results Artier 8 weeks of fat diet in hyperlipidemia rats,the ser- um levels of triglyceride[(0.99?0.38)mmol/L]and cholesterol[(3.17?0.18)mmol/L] were signifi- cantly increased(P
2. Analysis of sinomenine synthesis pathway based on transcriptome sequencing in Sinomenii Caulis

Objective: In order to explore the expression of sinomenine content control genes, synthetic control sites and expression pathway. Methods: In this study, high performance liquid chromatography (HPLC) was used to determine the content of sinomenine in the roots and stems of 49 Sinomenii Caulis in six populations. Two populations with large multiple differences in sinomenine content were selected, namely Shanxi Baoji and Guizhou Zunyi. The most representative of them were selected, and their roots and stems were taken for transcriptome sequencing and named as HR/LR and HS/LS. Results: Sequencing results showed that 355 201 transcripts were obtained by splicing clean reads, including 275 491 Unigene transcripts. There were 23 562 and 37 143 differentially expressed genes in HR/LR and HS/LS, respectively. GO database analysis showed that the functions of these differentially expressed genes were significantly enriched in aspartic-type endopeptidase activity and aspartic-type peptidase activity, it is speculated that these two enzymes might be encoded. The results of KEGG enrichment explained that the differentially expressed genes were involved in carbohydrate metabolism, protein binding to cell membrane and vitamin C synthesis. The results of qRT-PCR verified the expression of upstream key genes of the isoquinoline alkaloid synthesis pathway and found that it was positively correlated with the accumulation of sinomenine. Conclusion: This study provided a preliminary understanding of the molecular mechanism that caused the difference in sinomenine content, and provided a reference for further understanding of the accumulation rules and synthesis pathways of sinomenine.
3.Applications of PBL combined with role play teaching method in the clinical teaching of emergency internal medicine

Objective To investigate the application effect of Problem based learning (PBL) combined with role play teaching method in the training of emergency internal medicine.Methods Totally 60 medical students were randomly divided into two groups.The students in the observation groop were trained with PBL teaching method while those in control group were trained with traditional teaching method.The practical performances of the students in both groups were evaluated by virtual experience of dealing with patients in the emergency department through the role play teaching method.The grasp of basic knowledge was assessed by writing tests.Finally all the students in the observation group answered a survey about the teaching methods.Results There was uo difference in the scores of the basic knowledge test between the two groups ( P > 0.05 ).The practical ability of the observation group was much better than that of the control group in the role play practice ( P < 0.05 ).Almost all the students in the observation group accept the PBL teaching method.Conelusion PBL combined with role play teaching method is effective based on the characteristics of emergency medicine.
4.Exploration of pediatrics clinical probation teaching methods for eight-year program medical students

5.Development of Ethanol Production from Lignocellulosic Hydrolysates

Ethanol can be produced from lignocellulose by first hydrolysing the material to sugars,including hexose,and pentose,and then fermenting the hydrolysate to ethanol.Hydrolysis using dilute-acid has advantages over other methods.However,compounds which inhibit fermentation are generated during this kind of hydrolysis.Therefore,it is important to focus on microorganisms metabolizing xylose and tolerating/decomposing inhibitors,on detoxification methods of hydroly- sates with low-cost and facilitated to scale-up,and different fermentation modes in ethanol production from hydrolysate.This review summarized the advance in above aspects.
6.Discussion on the teaching design and practice of Clinical Medicine Summary

OBJECTIVETo detect the female carriers from the intron and/or exon-deletion Duchenne/Becker musclular dystrophy (DMD) familial members for prenatal or preimplantation genetic diagnosis.
METHODSUsing method of PCR to five microsatellite markers (located in 5' terminus and intron 44, 45, 49, 50), analysing of the short tandem repeat sequence polymorphism with the genescan and binding with the quantitative polymerase chain reaction, we detected the DMD carriers from 1 intron and exon -deletion family and 1 intron-deletion family.
RESULTSThe STR-50 genotype of II 2 in family 5 was 245/245, so II3 is DMD gene carrier. The STR-45 genotype of II6 and II8 were del/172, III19 was del/178, so they were all DMD gene carriers.
CONCLUSIONThe STR haploid linkage analysis combined with quantitative polymerase chain reaction is accurate and efficient to detect the female carriers from the intron and/or exon-deletion DMD familial members.

8.Carrier detection of Duchenne/Becker muscular dystrophy in Chinese families by microsatellite analysis.

OBJECTIVETo screen and detect the female carriers from the DMD/BMD family members for prenatal or preimplantation genetic diagnosis.
METHODSFor the detection of DMD/BMD carriers from 27 family members in 4 families, PCR to five microsatellite markers(located in 5' terminus and intron 44, 45, 49, 50) and analysis of the short tandem repeat(STR) sequence polymorphism with the use of genescan were implemented.
RESULTSSix of the 17 female members were obligate DMD gene carriers according to the haplotype analysis of the results of the genescan, which conformed with the pedigree analysis. Besides, the authors detected five carriers and five normal females in these families with the use of the haplotype analysis only. The most polymorphic locus was STR 49, and the least was STR 50.
CONCLUSIONThe STR haploid linkage analysis using (CA)n repeats within the human dystrophin gene is a rapid,accurate, objective method and is well suited for routine use in clinical laboratories engaged in DMD/BMD linkage analysis for the detection of carrier.

9.Ubc9 expression predicts chemoresistance in breast cancer.

Ubiquitin-conjugating enzyme 9 (Ubc9), the sole conjugating enzyme for sumoylation, regulates protein function and plays an important role in tumorigenesis. Whether Ubc9 is involved in the chemoresistance of breast cancer remains unknown. In this study, we aimed to evaluate the contribution of Ubc9 in the chemoresistance of breast cancer. Immunohistochemistry (IHC) was used to examine the expression level of Ubc9. Chi-square test, Wilcoxon test, and one-way ANOVA were applied to analyze the relationship between Ubc9 expression, clinicopathologic features, and clinical response to neoadjuvant chemotherapy. The significance of variables for survival was analyzed by the Cox proportional hazards model in a multivariate analysis. Kaplan-Meier survival curves were plotted and log-rank test was performed. The proportion of Ubc9-positive cells was higher in invasive ductal carcinoma than in normal breast tissues [(48.48 ± 17.94)% vs. (5.82 ± 2.80)%, P < 0.001]. High Ubc9 expression was associated with poor differentiation (Χ² = 6.538, P = 0.038), larger tumor size (Χ² = 4.701, P = 0.030), advanced clinical stage (Χ² = 4.651, P = 0.031), lymph node metastasis (Χ² = 9.913, P = 0.010), basal-like phenotype (Χ² = 8.660, P = 0.034), and poor clinical response to neoadjuvant chemotherapy (Χ² = 11.09, P = 0.001). The expected 6-year cumulative disease-free survival rate was 87.32% in patients with low Ubc9 expression compared to 68.78% in those with high Ubc9 expression (Χ² = 4.289, P = 0.038). These data indicate that high Ubc9 expression correlates with poor response to chemotherapy and poor clinical prognosis.

10.A case report of malignant peripheral nerve sheath tumors of the bladder

Malignant peripheral nerve sheath tumors(MPNST) of the bladder is a very rare malignant tumor, usually secondary to neurofibromatosis type 1 (NF-1), with a few sporadic cases. This paper reports a case of 70-year-old man with bladder MPNST who underwent transurethral resection. Adjuvant intravesical instillation with gemcitabine was given after surgery, and there was no local recurrence and distant metastasis after 12-month follow-up. This paper also made a corresponding literature review.
