1.Kinetics of Fed-batch Cultivation on Recombinant Escherichia coli Containing Human-like-Collagen cDNA

The kinetics of batch and fed-batch cultures of recombinant Escherichia coli to produce humanlike collagen were investigated. Through examining the density of substrate and the amount of mushrooms and the density of products during the process of fermenting, a set of kinetic models are set up. The influence of cell without plasmid was considered. The results show that the kinetic model may well simulate the fermenting process.
2.Pharmacokinetic and distribution of arctiin in rats

OBJECTIVE To study the pharmacokinetic and distribution of arctiin in rats. METHODS Each rat was given a single dose at random by oral administration. The arctiin in serum and organs were determined by use of RP-HPLC. All pharmacokinetic parameters were calculated with a 3P87 program. RESULTS After oral administration of arctiin at the dose of 300mg·kg-1, Arctiin plasma C-T curve conform to open two-compartment model. The Pharmacokinetic parameters were as follow: A=(37.374 5±8.964 7)μg·mL-1;B=(6.210 6±1.489 3)μg·mL-1;α=(0.004 3±0.000 9)min-1;β=(0.000 4±0.000 2)min-1;Kα=(0.420 2±0.167 5)min -1;t1/2α=(115.192 6±14.382 4)min ;t1/2β=(1 485.578 1±161.173 3)min;K10 =(0.001 0±0.000 4)min -1;K21=(0.001 4±0.000 6)min -1 ;K12=(0.002 3±0.001 3)min -1 ;Cmax=(41.786 3±7.521 7)μg·mL-1 ;Tmax=(9.891 9±4.341 4)min;AUC=(22 503.272 7±4 120.182 8)μg·min·mL-1. Liver had the highest concentration of arctiin after oral administration. CONCLUSION RP-HPLC method is rapid, sensitive and specific for the research of arctiin pharmacokinetic and its distribution in rats. Arctiin is distributed and eliminated quickly in rats.
3.Effect of arctiin on hemorheology of experimental rats with blood stasis ayndrone

OBJECTIVE To study influence of arctiin from seeds of Arctium lappa on hemorheology of experimental rats with the blood stasis syndrone. METHODS The blood hemorheology parameters, Fib, aPTT and PT of experimental rats with the blood stasis syndrone were evaluated using semi-automatic biochemical analysis. RESULTS Arctiin obviously decreased their high shear, middle shear, low shear, the blood viscosity, red blood cell aggregation index, red blood cell rigidity index and reductive viscosity. It also significantly prolonged the time of aPTT and PT and lowed the Fib concentration. CONCLUSION Arctiin apparently ameliorated the blood rheology abnormality and enhanced anti-coagulation effect on experimental rats with the blood stasis.
4.Dyuamical studies on metabolic chemistry of lignans from seeds of Arctium lappa.

OBJECTIVETo study the metabolic chemistry and pharmaco-dynamics characters of ligan from seeds of Arctium lappa.
METHODHPLC method was used in the study. The analysis was carried out on C18 column. The mobile phase was CH3CN-0.05% H3PO4 (36:64) with flow-rate at 0.6 mL x min(-1) and wave-length of 210 nm. The column temperature was kept at 25 degrees C.
RESULTThe results indicated that the ligan was detected in plasma and the main organs 5 min after po. The main metabolic production in plasma was arctigenin. In addition, arctigenin and an unknown product were found in metabolic production in the organs.
CONCLUSIONThe method was stable,simple and reproducible. It can be used to determine the metabolic product of the ligan. The metabolic chemistry of ligan in plasma was obviously different from that in the main organs.

6.Analysis of complications of venous indwelling needle in external jugular vein and veins of upper extremity

Objective To detect the complications of venous indwelling needle in external jugular vein and veins of upper extremity in patients with breast cancer.Methods One hundred breast cancer in-patients in Department of Oncology of Nanxishan Hospital of Guangxi Zhuang Autonomous Region(September 2006 to August 2008)were screened and divided into external jugular vein group(47 cases)and veins of upper extremity group (53 cases)according to the indwelling site path.During the indwelling period,phlebitis and leakage in two groups were compared.Results It was showed the complication rate of external jugular vein group(6.4%)was conspicuously lower than that in veins of upper extremity group(54.7%,P < 0.01).Conclusions It is suggested that the venous indwelling needle in external jugular vein is in high achievement ratio,the complication rate of phlebitis and leakage is lower,which could rise quality nursing care for breast cancer patient.
7.Primary lymphoma of breast: a clinicopathologic and prognostic study of 40 cases.

OBJECTIVETo study the clinicopathologic features and prognosis of primary lymphoma of breast.
METHODSForty cases of primary breast lymphoma, diagnosed according to the 2008 World Health Organization classification of hematopoietic and lymphoid tumors, were retrospectively studied. Immunohistochemistry was performed by SP method. The follow-up data were analyzed.
RESULTS(1) All the patients were females and the median age was 47 years. Unilateral and bilateral breast involvement were noted in 36 and 4 patients, respectively. The number of tumor were 31 cases (77.5%, 31/40) less than 3, and 9 cases (22.5%, 9/40) were 3 and more than 3. According to Ann Arbor staging system, 33 cases (82.5%) were in stage I to II and 7 cases (17.5%) in stage III to IV. The level of LDH in 9 cases (24.3%, 9/37) went up. For ECOG scores, 34 cases (85.0%) were 0 to 1 score and 6 cases (15.0%) were more than 2 scores. With respect to international prognostic index, 83.8% (31/37) were of score 0 to 2 and 16.2% (6/37) were of score 3 and more than 3. The axillary lymph nodes of 21 patients (53.8%, 21/39) were involved by the malignancy. (2) Histologically, 38 cases (95.0%, 38/40) were classified as B-cell lymphoma [including 27 cases (67.5%) of diffuse large B-cell lymphoma, 8 cases (20.0%) of mucosa-associated lymphoid tissue lymphoma, 2 cases of follicular lymphoma and 1 case of lymphoplasmacytic lymphoma]. The remaining cases included one case of peripheral T-cell lymphoma and one case of lymphoblastic lymphoma. Immunohistochemically, expression of CD20+/- CD79a were demonstrated in the 38 cases (95.0%) of B-cell lymphoma. The staining for CK was negative in all cases. In 33 cases, the positive rates of MUM-1, bcl-6 and bcl-2 were 57.6% (19/33), 30.3% (10/33) and 72.7% (24/33), respectively. Three cases were germinal center B cell phenotype and 21 cases were non-germinal center B cell phenotype. (3) Follow-up information was available in 37 patients (92.5%, 37/40). Twenty-three patients (62.2%, 23/37) were still alive and fourteen ones (37.8%, 14/37) died. For the 27 cases with diffuse large B-cell lymphoma, the five-year and disease-free survival rates were 48.0% and 36.0%, respectively.
CONCLUSIONSPrimary breast lymphoma is a rare disease entity. Diffuse large B-cell lymphoma is the commonest histologic type and the majority show a non-germinal center B cell phenotype. The level of LDH, number of tumor and international prognostic index are of prognostic significance.

9.Mutation of KLHL-10 in idiopathic infertile males with azoospermia, oligospermia or asthenospermia.

OBJECTIVETo investigate the relationship of the mutation of the spermatogenesis-associated gene KLHL-10 with azoospermia, oligospermia and asthenospermia.
METHODSGenomic DNA was extracted from the peripheral blood samples of 325 patients with idiopathic azoospermia (n = 11), oligozoospermia (n = 196) or asthenospermia (n = 118) and 100 fertile male controls. KLHL-10 mutations were detected for all the DNA specimens by PCR, DHPLC and sequencing techniques.
RESULTSA novel heterozygous mutation (C88 --> A) was identified in exon 1 from 1 oligospermia patient and 3 fertile male controls and another one (C424 --> A) confirmed in exon 2 from 4 fertile controls, 3 oligospermia patients and 1 asthenospermia man. Both of the mutations were synonymous, but neither missense mutation nor microdeletion of the KLHL-10 gene was found.
CONCLUSIONThe KLHL-10 gene is not a major contributor to azoospermia, oligospermia or asthenospermia in Chinese population. The value of this gene in the diagnosis of male infertility remains to be further investigated.

10.Generation and characterization of a cold-adapted attenuated live H3N2 subtype influenza virus vaccine candidate.

BACKGROUNDH3N2 subtype influenza A viruses have been identified in humans worldwide, raising concerns about their pandemic potential and prompting the development of candidate vaccines to protect humans against this subtype of influenza A virus. The aim of this study was to establish a system for rescuing of a cold-adapted high-yielding H3N2 subtype human influenza virus by reverse genetics.
METHODSIn order to generate better and safer vaccine candidate viruses, a cold-adapted high yielding reassortant H3N2 influenza A virus was genetically constructed by reverse genetics and was designated as rgAA-H3N2. The rgAA-H3N2 virus contained HA and NA genes from an epidemic strain A/Wisconsin/67/2005 (H3N2) in a background of internal genes derived from the master donor viruses (MDV), cold-adapted (ca), temperature sensitive (ts), live attenuated influenza virus strain A/Ann Arbor/6/60 (MDV-A).
RESULTSIn this presentation, the virus HA titer of rgAA-H3N2 in the allantoic fluid from infected embryonated eggs was as high as 1:1024. A fluorescent focus assay (FFU) was performed 24-36 hours post-infection using a specific antibody and bright staining was used for determining the virus titer. The allantoic fluid containing the recovered influenza virus was analyzed in a hemagglutination inhibition (HI) test and the specific inhibition was found.
CONCLUSIONThe results mentioned above demonstrated that cold-adapted, attenuated reassortant H3N2 subtype influenza A virus was successfully generated, which laid a good foundation for the further related research.
