1.Anti-migration and anti-angiogenic effects of tanshinone ⅡA on breast cancer cell MDA-MB435

Aim: To investigate the mechanism of anti-proliferation, anti-migration and anti-angiogenic effects of tanshinone ⅡA. Methods: In this study, MTT, 2-D/3-D migration, tube formation, PCR, chick embryo chorioallan-toic membrane (CAM) were used to evaluate the anti-proliferation, anti-migration and anti-angiogenic effects of tanshinone ⅡA. Results: Tanshinone ⅡA significantly inhibited the proliferation of human breast cancer line MDA-MB-435 with an IC_(50) of 21 nmol/mL And it is showed that breast cancer cells migration was effectively prevented by tanshinone ⅡA at 5 and 6 nmol/mL in a wound healing assay and a transwell migration assay. In ad-dition, tanshinone ⅡA inhibited the tube formation of newborn cattle aortic endothelial cells( NCAECs) after 2 h co-incubation with MDA-MB-435. These effects were not due to the inhibition of NCAECs proliferation; because tanshinone ⅡA non-selectively inhibited NCAECs growth with an IC_(50) of 124 nmol/mL Tanshinone ⅡA showed dose-dependent inhibitory effects on mRNA expression of VEGF and two transcription factors (HIF-1α/c-Myc) . Tashinone ⅡA was also found to inhibit angiogenic in vivo in the CAM assay. Conclusion: These results suggest that tanshinone ⅡA may exert its anti-proliferation, anti-migration and anti-angiogenic effects through down-regu-lating two transcription factors and VEGF. These novel anti-proliferations, anti-migrations, anti-angiogenic activi-ties of tanshinone ⅡA are likely to contribute to its cancer chemopreventive and therapeutic potential, especially in the treatment of breast cancer.
2.Development of Predictive Model for Combined Effect of Temperature and Salinity on the Growth of Vibrio parahaemolyticus

Based on the standard strain-Vibrio parahaemolyticus BJ1.1997, temperature (7?C~43?C) and salinity (0.5%~9.5% NaCl) which affected its growth status was studied with uniform design. The results showed that Logistic equation was optimal in primary models, the second was Gompertz equation, the last was Linear equation, so growth parameters can calculated from Logistic equation. The secondary model was developed by square root models, its r value was 0.9863, the lowest growth temperature was 9.0506?C and the highest growth salinity was 5.93% [the corresponding lowest growth water activity (Aw min) was 0.9227]. Through F test, residual analysis and evaluation by bias factor and accuracy factor, the model can exactly describe the relationship between the growth rate and combined effect of temperature and salinity (water activity).
3.The correlation study of beta-ifbrinogen gene polymorphisms and plasma ifbrinogen concentration in patients with colorectal cancer

Background and purpose:Patients with colorectal cancer are often accompanied by the increase of plasma ifbrinogen concentration. This study aimed to investigate the distribution characteristics of beta-ifbrinogen gene-448G/A, -148C/T, -1420G/A and -854G/A polymorphism and plasma ifbrinogen (Fg) concentration in patients with colorectal cancer. Furthermore, we analyzed their effects on the occurrence and development of cancer.Methods:The level of plasma Fg was quantiifed by using Clauss clotting method.FGBβ gene polymorphisms were identiifed by re-al-time lfuorescence quantitative PCR (RTFQ-PCR) in 194 colorectal cancer patients and 74 healthy controls.Results:The plasma Fg levels in tumor metastasis group and non-metastasis group were signiifcantly higher than that in control group, respectively (P<0.05). Compared with control group, the frequencies of -148T allele and mutation genotype were notably higher in disease group (P<0.05). In all the groups, the plasma Fg levels of those with -148T allele were higher than those without -148T allele (P<0.05). In stageⅣ patients, there was no difference in PFS between -148T wild gen-otype group and mutation genotype group (P>0.05).Conclusion:Plasma Fg concentration in patients with colorectal cancer was signiifcantly raised, which suggests that Fg may play a role in the occurrence and development of colorectal cancer. The beta-ifbrinogen gene -148C/T polymorphism is one of the reasons that cause plasma Fg elevation, but has no correlation with prognosis of patients with stageⅣ colorectal cancer.
5.Research progress of PARP inhibitors in ovarian cancer

Ovarian cancer is one of the highest mortality rate of gynecologic malignant tumors. Chemotherapy can improve the survival rate of the traditional ovary. In recent years, PARP [poly(ADP-ribose)polymerase]inhibitors in breast cancer susceptibility gene (breast cancer susceptibility gene, BRCA) mutations in patients with ovarian cancer can significantly improve the disease-free survival, may change the prognosis of patients with ovarian cancer. This part of PARP [poly(ADP-ribose)polymerase] inhibitors, inhibiting the repairment of DNA damage in tumor cell, causing DNA damage accumulation, eventually killing tumor cells.In breast cancer susceptibility gene 1 (breast cancer susceptibility gene1, BRCA1)/BRCA2 mutation patients with ovarian cancer, PARP inhibitors and BRCA mutation of the synthetic lethal effect provides a new direction for the development of anti-cancer drugs. Now, many highly selective and sensitive PARP inhibitors have been developed and applied in clinical trials.Although PARP inhibitor monotherapy can produce a therapeutic effect in BRCA mutation in patients with ovarian cancer, but the clinical application is still used in combination with other chemotherapy or radiotherapy. This review is focused on the recent progress in clinical trials of PARP inhibitors in combination with common chemotherapeutic agents.
6.Research of anti-tumor peptide neutralizing antibodies activities

The generation and activity of serum neuralizing antibody in cynomolgus monkeys and SD rats undergoing long-term toxicity study with antitumor peptides HM-3 were investigated.The rats were administered intravenously with HM-3 at doses of 4.5 mg/kg,13.5 mg/kg,and 40.5 mg/kg for 6 months,and the cynomolgus monkeys were administrated intravenously with HM-3 at doses of 3 mg/kg,9 mg/kg and 27 mg/kg for 6 months,respectively.Anti-HM-3 antibodies and their neutralizing activities in serum samples taken every month after the administrations were determined by ELISA and cell migration assay,respectively.During the long-term administrations,anti-HM-3 antibodies were generated in some SD rats at high and moderate dose groups,and the antibody-neutralizing activities could be detected in a number of these samples (P <0.05).However,activity could be detected in very few monkeys (P < 0.05),and the antibody titers were not correlated with the neutralizing activities.Therefore,we conclude that the antigenicity of HM-3 was low,but after long-term administration at high dose,low affinity neutralizing antibody could be generated in small number of samples.

The perspective of noninvasive continuous blood pressure measurement is also mentioned.

9.Study on bioactive alkaloids ingredient produced by marine actinomyces N331

Objective To study the antibacterial constituents in the fermentation broth of marine actinomyces N331.Methods The antibacterial components were isolated and purified from the fermentation broth of strain N331 by absorption with macroporous resin and silica gel column chromatography,their physiochemical characteristics and bioactivities were preliminarily investigated.Results A crystal ingredient with bioactivities was isolated from the fermentation broth and it was identified as alkaloids which was composed of several compounds with approximate polarity,and its the minimum inhibitory concentration against sensitive and drug-resistant Staphylococcus aureus was 4?g?mL-1,and its half maximal inhibitory concentration(IC50) was 2.0,3.0?g?mL-1 respectively against KB and lung cancer A549 cells,which was equivalent to cisplatin.Conclusion Marine actinomyces N331 can produce the alkaloidal substances with strong antibacterial activity and cytotoxicity.
10.Clinical value and nursing intervention of prone position ventilation in treating patients

Objective To investigate the effect of position ventilation(PPV) on oxygenation,the airway sputum drainage and hemodynamic of acute respiratory distress comprehensive syndrome(ARDS).Methods From June 2015 to April 2016,13 cases with moderate or severe ARDS were researched.The changes of PaO2,PaCO2,SpO2,PaO2/FiO2 and airway sputum drainage were observed as well as HR,MAP,CVP,CO,CI,SVV,etc.Results Totally 56 PPV were taken.Compared with pre-treatment of PPV,the number of PaO2 and PaO2/FiO2 increased from (74.7±13.18) mm Hg and 168.65±22.12 to (92.13±17.82) mm Hg and 208.80±24.04(P<0.05) respectively;SpO2 and airway sputum drainage increased from (95.32±2.76)% and (7.84±1.68)mL to (97.04±1.84)% and (13.25±3.55)mL(P=0.000).However,there were no significant differences in HR,MAP,CVP,CO,CI and SVV(P>0.05).Conclusion PPV can effectively improve oxygenation in patients with ARDS and promote airway sputum drainage,and hemodynamic effects are not obvious.However,nursing intervention still should be strengthened.
