1.Effect of hormone replacement therapy on hemorheology in postmenopausal women

Objective Our study is to observe the effects of conjugated equine estrogen (CEE) combined with medroxyprogesterone acetate (MPA) on hemorheology in postmenopausal women in order to evaluate mechanism responsible for the increase in the incidence of cardiovascular disease after menopause and also to provide proper instruction and advice of hormone replacement therapy (HRT) for postmenopausal women Methods 60 healthy postmenopausal women were assigned to 3 groups randomly, 20 cases in each group Group A: CEE 0 625 mg/d+MPA 2 mg/d+calcium carbonate co. (Ca D) 600 mg/d Group B: CEE 0 3 mg/d+MPA 2 mg/d+Ca D 600 mg/d Group C: Ca D 600 mg/d only served as control Measurements of hemorheological parameters were performed before and 6 months after treatment Results After 6 months of HRT, group A: blood viscosity under the shear rate of 94 5 s -1 dropped from (5 23? 0 37) mPa?s to (5 03? 0 43) mPa?s ( P 0 05) Conclusion Both different dosages of CEE have favor effects on blood viscosity and erythrocyte deformabillity, which could play the role in preventing cardiovascular disease in postmenopausal women
2.Formula Optimization of Memantine Hydrochloride Sustained Release Pellets by Box-Behnken Surface Response Design

Objective: To optimize the formula of memantine hydrochloride sustained release pellets and investigate the drug release in vitro.Methods: Memantine hydrochloride sustained release pellets were prepared by a fluidized bed.The factors such as inlet air temperature, spray pressure and feeding rate were optimized by orthogonal tests.The Box-Behnken response surface methodology was used to screen the major influencing factors (ethyl cellulose aqueous dispersion, PEG 6000 and the coating weight) in the release of memantine hydrochloride.The coating formula and coating weight were optimized with the cumulative release rate of memantine hydrochloride in 2, 6,and 12 h as the response values.The drug release in vitro was also studied.Results: The optimum preparation parameters of the pellets were as follows: the inlet air temperature of 45℃, the spray pressure of 1.0 bar, and the feeding rate of 1.5 r·min-1.The best sustained release coating formula was as follows: the content of ethyl cellulose aqueous dispersion of 8.4%, the content of PEG 6000 of 2.3%, and the weight gain of sustained release layer of 16.7%.The memantine hydrochloride sustained release pellets had notable sustained release effect.Conclusion: Orthogonal tests and Box-Behnken response surface method can be used for the formula optimization of memantine hydrochloride sustained-release pellets.The established fitting model is simple with good predictability.
4.Quality evaluation of bletillae rhizoma based on hemostatic biopotency.

This dissertation is to determine the biopotency of hemostat which processed in different places by establishing a bioassay method of Bletillae Rhizoma based on the thrombin time. Contrast test is the main methodology. Specifically, the reference substance of Bletillae Rhizoma is determined by comparing with the control substance of vitamin K1 using thrombin time, which is calibrated the Bletillae Rhizoma. The hemostatic biopotency is calculated by using the method of "parallel line assay method based on quantitative responses" (3.3) from different processed products. It indicates that there is a strong linear correlation between Bletillae Rhizoma and control drugs (Y = 66.332-23.913X, R2 = 0.995 3). The hemostatic biopotency of Bletillae Rhizoma from different processed products ranged between 821.93-1 187.53 U x g(-1) shown in the paper, and all of them can meet the requirements of the test. The methodology has an appropriate instrument precision (RSD 3.8%), intermediate precision (RSD 4.6%), repeatability (RSD 3.2%) and stability (RSD 3.7%). Therefore, it can be turned out that the methodology which established in the dissertation is good at determinating the hemostatic biopotency of Bletillae Rhizoma and it is reliable, simple and repeatable.

7.Effects of Pomegranate Leaf Tennis and Ellagic Acid on In Vitro Primary Rat Adipocyte Transformation and Lipolytic-related Factor Expression

This study was aimed to observe effect of pomegranate leaf tannin and ellagic acid on primary rat adipocyte differentiation and lipid metabolism-related factor expression. Primary rat preadipocyte was in vitro cultured to observe the effect of pomegranate leaf tannin and ellagic acid on lipid fat cells as well as mRNA expression of its related factor. The results showed that pomegranate leaf tannin and ellagic acid had obvious inhibition effect on fat formation in fat cells. It had certain inhibition effect on activities of lipoprotein lipase (LPL) and glucose-3-phosphate dehydrogenase (GPDH). It promoted fat decomposition and reduced intracellular lipid content. It upregulated PPARγ and fatty acid-binding protein (aP2). It downregulated obese (ob) gene level. It was concluded that pomegranate leaf tannin can inhibit fat generation of fat cells and promote fat metabolism. Ellagic acid was its main active ingredient, which had the same effect as pomegranate leaf tannin.
8.Comparison of hollow compression screws and absorbable screws for the treatment of the fifth metatarsal fracture:ankle function and fracture displacement

BACKGROUND:The hol ow compression screws and absorbable screws can be applied for the treatment of the fifth metatarsal fracture. However, little evidence is obtained in China. OBJECTIVE:To compare ankle function and fracture displacement in patients with the fifth metatarsal fracture after interventions of hol ow compression screws and absorbable screws. METHODS:32 patients with the fifth metatarsal fracture were recruited from Zhongnan Hospital of Wuhan University between June 2012 and June 2014. According to the internal fixation method, the involved patients were divided into hol ow compression screws group (n=18) and absorbable screws group (n=14). Clinical outcomes of patients in the two groups were evaluated through observations of the mean healing time, postoperative infection, ankle activity and fracture displacement. RESULTS AND CONCLUSION:Al the 32 patients were fol owed up for 6-18 months and were al healed. Compared with the absorbable screws group, the healing time was shorter (P<0.05), incidence of postoperative screw rupture and fracture displacement was lower (P<0.05), and ankle activity was better in the hol ow compression screws group (P<0.05). It is necessary to enhance the strength, improve screw thread, increase anti-pul ing property, and al ow early ankle activity, thus reducing the risk of clinical application of the absorbable screws.
9.Characteristics of hippocampal 3-dimensional magnetic resonance imaging in amnestic mild cognitive impairment

Objective To identify the characteristics of hippocampal 3-dimensional MRI in patients diagnosed as having subtypes of amnestic mild cognitive impairment(aMCI)using hippocampal surfacebased analytic technique.Methods Fifry aMCI patients and 16 healthy controls who were equivalent in age and education(NC)were recruited.Every subiect carried out a 3-dimensional MRI scan.After the imaging data were acquired.the borders of the hippocampus were manually traced in coronal vlew using the software of InsightSNAP1.4.1. Hippocampal volume was computed automatically and statistically analysed.Hippocampal 3-dimension MRI were transformed into 3-dimension parametric surface mesh models of 400×200 prids.Hippocampal radial distance measures which was the distance from the surface point to the central axis were statistically compared between two groups.The radial atrophy significance maps were acquired and adjusted for multiple comparisons.Hippocampal morphological difference maps of aMCI in contrast with NC were acquired.Results The average normalized volume of left hippocampus were(3247.5±600.2)mm3 in aMCI patients and(3467.9±451.3)mm3 in NC subjects.The average normalized volume of right hippocampus were(3416.8±699.1)mm3 in aMCI patients and(3469.1±358.9)mm3 in NC subjects.Comparison of hippocampal volume did not differ significantly between aMCI patients and NC subjects(t=1.161,P=0.255;U=0.178,P=0.859).By using hippocampal surface-based morphologic analytic technique,3-dimension hippocampal morphological difference maps between two groups were acquired,showing significant atrophy on the lateral and inferior hippocampal surface which corresponded to CA1 and subiculum hippocampal subfields bilaterally in aMCI patients compared with NC subjects. Conclusions aMCI patients do not have significant volume loss in the hippocampus. Through hippocampal surface-based morphologic analyses, partial regional atrophy of hippocampus at some degree is found, mainly localizing in the lateral and inferior hippocampal regions which correspond to CA1 and subiculum hippocampal subfields bilaterally in aMCI compared with NC. These results may reflect the early image marker in aMCI.
