1.Antitumor effects of curcumin combining with adriamycin in vitro

AIM To investigate antitumor effects of curcumin (Cur) combining with adriamycin(ADM) on human tumor cell lines in vitro . METHODS The antitumor effects of the drugs on tumor cell lines in vitro were determined with MTT method. The Jin's formula was used to analyse the effect of drug combination. RESULTS In simultaneous adiministration Cur 2 04 ?mol?L -1 ~16 29 ?mol?L -1 combining with ADM 0 70 ?mol?L -1 ~5 52 ?mol?L -1 produced a simple addition or potentiation effect. In sequential adiministration the first adiministration of ADM followed by Cur resulted in an antagonistic effect, while the change of the order of adiministration produced an simple addition effect. CONCLUSION Simultaneous adiministration of Cur and ADM produces synergistic effect but sequential adimistration of the drugs only produce a unidirectional synergistic effect.
2.Effects of deoxycytidine on DNA methylation state and expression of mRNA and protein of pl6 gene in human squamous lingual carcinoma SCC-9 cells

Objective To study the effects of deoxycytidine (5-aza-2 deoxycytidine, DAC) on DNA Methylation state and expression of mRNA and protein of pl6 gene in human squamous lingual carcinoma SCC-9 cells in vitro. Methods The SCC-9 cells were divided into four groups, group 0, 1, 2 and 3 which processed using three gradients concentration of DAC. The group 0 without DAC was as the control group. Q-MSP was used to detect the state of methylation of the p16 in SCC-9 cells treated by DAC after 48 hours. Real-time fluorescence quantitative PCR was used to detect mRNA expression level changing of the p16 in SCC-9 cells treated by DAC. Immunohistochemical method was used to detect the expression of p16 protein. Results The hypermethylation and non-methylated p16 gene in SCC-9 was mixed with the results of Q-MSP. The results of Real-time PCR showed that mRNA expression of p16 in SCC-9 cells which treated by the different concentration of DAC for 48 hours was higher the control group. And difference was statistically significant (P < 0.05). The high expression of p16 protein was found in the experimental group with immunohistochemical method. Conclusion The p16 gene methylation states of SCC-9 may be suppresses and the recovery of mRNA and protein expression of p16 gene must be prompted by DAC.

5.A comparative study of p1 6 gene methylation status in primary tongue squamous cell carcinoma and cervical lymph node metastasis

DNA from 32 cases of tongue squamous cell carcinoma(TSCC)specimens and the neck lymph node metastasis specimens were processed by bisulfite treatment.The methylation of the specimens was examined by Q-MSP amplification.The consistency of p1 6 methyla-tion of primary TSCC with that of lymph node metastasis tumor was 90.62%(P <0.05).We assumed that p1 6 gene promoter CpG island methylation rates in primary TSCC and metastatic lymph nodes are consistent,p1 6 gene methylation status can be a methylation predictor be-tween premary and lymph node metstasized TSCC.
6.Study on the Relation of Glycyrrhizic Glycoside on Hepatitis B Virus Replication in Vitro

OBJECTIVE:To study the relation of glycyrrhizic glycoside on hepatitis B virus(HBV)replication in vitro.METHODS:2.2.15cell line was used and treated with glycyrrhizic glycoside,the contents of HBsAg and HBeAg in the su?pernatant of cell culture with different concentrations of glycyrrhizic glycoside were quantified.RESULTS:The inhibitory rates of glycyrrhizic glycoside on HBsAg were83.8%,71.0%,62.1%,52.0%and29.7%respectively at the concentrations of800,400,200,100and50?g/ml respectively.CONCLUSION:Glycyrrhizic glycoside does not promote HBV replication in vitro experiment.

With the development of recombinant DNA technology, genetically altered mouse models for human cancers are critical to the investigation and characterization of oncogene and tumor suppressor gene expression and function and the associated cancer phenotype. Recently, the inhibition of tumor angiogenesis becomes a new “hot spot” in cancer researches. And the genes involved in the regulation of tumor angiogenesis play an increasingly important role in the field of tumor researches. This review is about the progress of the research in transgenic tumor models and focuses on genes regulating tumor angiogenesis associated with transgenic mice model.
9.Inhibitory effect of EGCG on apoptosis of Langerhans cells after UVB irradiation

Objective To observe the damage to Langerhans cells induced by UVB irradiation,and to evaluate photoprotective effect of these cells from UVB irradiation by epigallocatechin-3-gallate (EGCG).Methods Biopsy specimens were obtained from normal adult foreskin,and epidermal cells were isolated.Density gradient centrifugation and magnetic cell sorting were used simultaneously to purify Langerhans cells from the cell suspension.These cells were then divided into three groups,control (no ir- radiation or EGCG treatment),UVB (irradiation) and EGCG (irradiation+ECCG treatment) groups. The cells in the UVB and EGCG group were irradiated by UVB (30 mJ/cm~2).After the irradiation,the U- VB group was incubated with RPMI-1640 containing 10% bovine serum for 4 hours,while the EGCG group with the same medium containing 200?g/mL of EGCG for 4 hours.Another four hours after the treatment, the cells were collected for the detection of apoptosis rate by propidium iodide staining and flow cytometry. Results Exposure to UVB (30 mJ/cm~2) significantly increased the apoptotic rate of Langerhans cells.The apoptotic rate in EGCG group was significantly lower than that in the UVB group,but was higher than that in the control group.Conclusion Rate of apoptosis of Langerhans cells could be increased by UVB irradia- tion,while EGCG could prevent the increase of apoptosis.
10.Effect of breast serf-examination on early detection of breast cancer and its prognostic value:a meta-analysis

Objective To assess the efficacy of early detection of breast cancer and the prognostic value of breast self-examination(BSE).Methods Randomized controlled trials(RCTs)involving BSE were included the present meta-analysis based on their qualities.The pooled analysis was performed with RevMany 4.2.8.Results Three RCTs were identified and then they were included the present metaanalysis.In women who conducted a BSE.the rate of biopsy fora lump in breast was 1.27%in the first 5 years,2.47%in more than 5 years but less than or equal to 10 years,and 2.50%in more than 10 years, respectively,all of which were significantly higher than in those who could not conduct a BSE(P<0.05) There was no significant difierence in the rate of final diagnosis and the mortality for breast cancer between the two populations in any follow-up time(P>0.05).Conclusion The rate of biopsy for a lump in breast in women could be increased by a BSE.which should not have been discarded from the early detection of breast cancer completely despite it is not helpful in increasing the rate of final diagnosis and decreasing the mortality for breast cancer.
