Academic Journal of Second Military Medical University 2012;33(1):53-57
Objective To explore the factors affecting the circadian rhythm of blood pressure in treated hypertensive patients. Methods Based on the clinical data, ambulatory blood pressure, 24-hour mean blood pressure (24hMBP) and circadian rhythm, 125 consecutive patients receiving antihypertensive treatment were divided into the following groups: normal 24hMBP and dipper type (type I), abnormal 24hMBP and dipper type(type E), abnormal 24hMBP and non-dipper type (type I), and normal 24hMBP and non-dipper type (type IT). The clinical data and ambulatory blood pressure of the latter three groups were compared. Non-conditional Logistic regression was used to analyze the factors for ambulatory blood pressure. Results Ninety patients had abnormal 24hMBP (72. 0%), and 106 (84. 8%) had abnormal circadian rhythm. Among these cases, 2 (1.6%) patients had type I, 17 (13.6%) had type TJ, 73 (58.4%) had type I, and 33 (26.4%) had type IT. The incidence of kidney disease in patients with abnormal 24hMBP was significantly higher than those with normal 24hMBP (P< 0. 05). The incidence of diabetes mellitus in patients with abnormal circadian rhythm (non-dipper type) was higher than those with normal circadian rhythm (dipper type), with the highest incidence seen in patients of type IT (P<0. 05); the rates of calcium channel blocker (CCB) use and a daily morning dose administration in type TV patients were higher than those in type II and type III patients (P<0. 05). Type IT group had the most severe abnormal ambulatory blood pressure among the latter three groups. Logistic regression analysis showed that complication with renal diseases (OR=0. 301, 95%CI:0. 124-0. 729, P = 0. 008), use of CCB (OR = 2. 191,95%CI: 0. 967-4. 966,P = 0. 048), and administration of a morning dose (OR = 2. 384,95% CI: 1. 017-5. 591,P = 0. 046) were the factors of ambulatory blood pressure. Conclusion Abnormal circadian rhythm of blood pressure is high in patients receiving antihypertensive treatment. Complication with kidney diseases, use of CCB, and a daily morning dose are the factors for ambulatory blood pressure, indicating it is be reasonable to analyze factors for circadian rhythm by combining the 24hMBP and the components of circadian rhythm.

ObjectiveTo develop three animal models to mimic muscle tremors of Parkinson's disease and observe effects of Chinese herb compounds Jian-Xing on these models.MethodsPure muscle tremor mice models were developed by single intraperitoneal injection of arecoline or oxotremorine. The mitochondria damaged model was developed by intraperitoneal injection of MPTP(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) 20mg/kg for 8 days. Duration time of tremoring was recorded. The climbing time and spontaneous movements of MPTP mice were recorded. HPLC was used to detect the content of dopamine and it's metabolites in MPTP mice.ResultsAfter injection of oxotremorine or arecoline, model mice exhibited significant tremoring, duration time of tremor of mice in Jian-Xing group shortened significantly. As to MPTP model mice, climbing time of model mice prolonged, times of spontaneous movements of model mice decreased and the content of dopamine in striaturn decreased compared with control group. Climbing time of mice in Jian-Xing group shortened, spontaneous movements increased and content of dopamine increased distinctively. ConclusionOxotremorine, arecoline and MPTP all can produce tremor animal models. Chinese herb compounds Jian-Xing can shorten the duration time of tremor. As to MPTP model, Jian-Xing can shorten the climbing time of model mice, increase the spontaneous movements and increase the content of dopamine in striaturn.

Objective To assess the feasibility of heterogeneous acellular dermal matrix(ADM)seeded with adipose derived stem cells(ADSC)for urethroplasty in a rabbit model. Methods ADSC were isolated from a rabbit and expanded in vitro, then identified by flow cytometry. We seeded ADSC onto the ADM and made it into tissue-engineered urethra.12 male rabbits were removed 1 cm urethra and divided into experiment group and control group. There were 6 rabbits in each group.Reconstructed urethra with tissueengineered urethra was used in experiment group, while unseeded ADM were used in control group.Urethrography was performed at 6 months after surgery. The animals were scarified at 3 and 6 months after surgery and the repaired urethra were harvested.H&E staining and immunohistochemical staining were performed with cytokeratin AE1/AE3 and smooth muscle desmin makers. Results The morphology of isolated ADSC was with long spindle cross-links,and had multicentral growth.Flow cytometry showed that the ADSC expressed CD166,CD105,CD90 and CD44,but not expressed CD45 and CD13.The cells could growth well on the ADM and showed good biocompatibility with it.All animals could void normally, urethrography showed there was no significant stenosis.3 months after surgery, the experiment group appeared regenerated smooth muscle but not in the control group, both groups did not regenerate urothelium. At 6 months urothelium and smooth muscle cells could be observed in the experiment group, but only the smooth muscle was evident in the control group. Conclusions By applying tissue engineering methods,we can seed the ADSC onto the heterogeneous ADM and make it into tissue-engineered urethra, which can help improve the reconstructive effect of urethra.
