2.Clinical comparison of different materials as posterior dental implants

BACKGROUND:Studies have shown that selection of implant crowns that directly impact the service life of implants and patient’s periodontal health is very important.
OBJECTIVE:To compare the clinical effects of Lava zirconia crowns, goal-platinum al oy ceramic crowns and silver-pal adium al oy crowns supplemented by dental implants used in the single missing posterior tooth.
METHODS:Sixty patients with first molar missing (120 teeth) were divided into three groups and treated with Lava zirconia, goal-platinum al oy ceramic and silver-pal adium al oy crowns applied to the dental implants for posterior column defects. The clinical restorative effects of three prostheses were compared.
RESULTS AND CONCLUSION:During 6-48 months of fol ow-up, the gingival edge coloring, gingival margin microleakage, the color of prosthesis of Lava zirconia al oy ceramic crowns and gold-platinum al oy ceramic crowns were better than those of silver-pal adium al oy crowns, and the gingival edge coloring and the color of prosthesis of Lava zirconia al oy ceramic crowns were better than those of gold-platinum al oy ceramic crowns, while the gingival margin microleakage of gold-platinum al oy ceramic crowns was better than that of Lava zirconia al oy ceramic crowns. Silver-pal adium al oy crowns were lower than Lava zirconia ceramic crowns and gold-platinum al oy ceramic crowns in the fracture extent. But silver-pal adium al oy crowns had the highest gingival index, the worst gingival health and rapidest and most severe plaque formation. Therefore, Lava zirconia al-ceramic crowns and gold platinum al oy-porcelain crown are ideal prostheses for implant restorations. The former has better biocompatibility, and the latter shows better marginal adaptation.
3.Establishment and identification of humanized SCID mouse model

Objective:To establish and identify humanized-SCID mouse model(hu-SCID).Methods:SCID mouse was treated by CTX to inhibit the hemocytopoiesis. With successive 4-day injection, human peripheral blood mononuclear cells(PBMC) were engrafted into SCID mouse through intraperitoneal injection. After 4, 8 and 12 weeks of engraftment, peripheral blood, spleen and liver tissues of engrafted SCID mouse were harvested. Human CD3~+, CD19~+ cells in peripheral blood were analyzed by inflorescence microscopy and FCM, human CD3~+, CD19~+ cells in spleen and liver tissues were observed by immune histochemistry, and human IgG level in SCID mouse serum was measured by ELISA.Results:After engraftment of 4, 8 and 12 weeks, human CD3~+, CD19~+ cells in SCID peripheral blood were identified by inflorescence microscopy and the percents were 31% and 10% respectively by FCM analysis. And these cells could be evidenced after 12 weeks later. Through immune histochemistry human CD3~+、CD19~+ cells were detected in mouse spleen but not in liver tissue. Furthermore the titer of human IgG in mouse serum was 390,1 100 and 1 040 ?g/ml at each time point respectively.Conclusion:Our experimental results demonstrated that a bona fide humanized SCID model was established.
4. Wnt3-induced epithelial-mesenchymal transition in mouse hepatic progenitor cells

Objective To investigate the impact of wnt3 upon epithelial-mesenchymal transition in mouse hepatic progenitor cells (14-19 cells). Methods Ad-GFP-wnt3 virus and blank Ad-GFP were transfected into 14-19 cells; cell morphology was observed under microscope. The migration ability of 14-19 cells was examined by wound-healing assay and transwell assay. The expression of epithelial and mesenchymal markers in 14-19 cells was detected by real-time PCR and Western blotting analysis. Results Microscopic observation found that 14-19 cells changed from an epithelial characteristic shape into a spindle-like shape. Wound-healing assay and Transwell assay showed that the migration ability of 14-19 cells highly expressing wnt3 was significantly enhanced compared with the blank control group(P<0. 01). Real-time PCR and Western blotting analysis showed that mesenchymal markers N-cadherin, vimentin and twistl were significantly increased at both mRNA and protein levels in Ad-GFP-wnt3 virus group than the blank control group, while the epithelial marker E-cadherin was significantly decreased at mRNA and protein levels(P<0. 01). Conclusion Wnt3 can induce epithelial-mesenchymal transition in mouse hepatic progenitor cells, indicating that wnt3 may take part in the progress of hepatic fibrosis.
5.Effect of Rehabilitation Stroke Unit on Walking in Stroke Patients with Hemiplegia

Objective To investigate the effect of rehabilitation stroke unit on walking in stroke patients with hemiplegia. Methods 86 patients with walking impairment after stroke were randomly divided into control group (n=43), who accepted conventional treatment, and experimental group (n=43), who were incorporated into the rehabilitation stroke unit. They were assessed with Berg Balance Scale (BBS),10 m Maximum Walking Speed (MWS), Fugl-Meyer Assessment (FMA) and modified Barthel Index (MBI) before and 8 weeks after treatment.Results The scores of BBS, MWS, FMA and MBI improved after treatment in both groups (P<0.01), and improved more in the experimental group than in the control group (P<0.05). Conclusion Rehabilitation stroke unit is effective on walking ability in stroke patients with hemiplegia.
6.Morphologic features of the acetabulum bone joint area.

The research on the morphology features of the acetabulum bone joint surface area would be helpful to establishing the acetabulum 3D model for the purpose of the biomechanical analysis of hip joint, and therefore might have its important clinical significance. However, in former studies, the acetabulum was simply considered as a semi sphere. In this study, based on the acetabulum 3D-point data acquired by the 3D laser surface scanner and the reverse engineering technology together with the optimal fit algorithm, two kinds of best-fit model were achieved by a sphere surface and a rotating elliptical surface respectively approaching to the acetabulum bone joint surface. Both fitting errors were then compared and analyzed. The results showed that the fitting error of the rotating elliptical surface was significantly less than that of the sphere surface (P < 0.001). The average radius of fitting sphere was 24.37 +/- 2.22 mm and the average long axis of fitting rotating elliptical surface was 26.02 +/- 2.76 mm while its short axis was 24.17 +/- 2.16 mm. These findings would be helpful to our new recognition of the acetabulum since they were results of the first quantitative analyses for the acetabulum bone surface and also might serve as an important reference base in its further studies and application.

7.Phenotypic characteristics of peripheral blood lymphocytes and its clinical significance in patients with coal arsenic poisoning

Objective To study the change and rule of immunological function among the patients with coal arsenic poisoning in order to provide a basis for tumor risk evaluation and monitoring.Methods Seventy patients with coal arsenic poisoning aged from 24 to 71 years old(44 men,26 women,averaging 41 years old)were divided into 4 groups including 23 cases having a course less than 10 years,21 case8 lasting for 10~19 years,20 cages for more than 20 years,6 cases of cancer,and 26 healthy normal controls.Flow cytometer(FCM)was used to analyze the frequency of CD3+(total T cell),CD3+CD4+(inducer/helper T cell),CD3+CD8+(suppressor/cytotoxic T cell),CD19+(B lymphocyte),and CD56+CD16+(natural killer cell)lymphocyte subsets in the peripheral blood of the subjects and the expression rates of lymphocytic membrane surface molecules of human leucocvte antigen (HLA)-DR,CD25,CD38 were also determined by FCM.Results The pmportions of CD3+cells in periDheral blood of less than 10 years,10~19 years,more than 20 years and cancer groups were (63.76±9.32)%。(55.63± 12.97)%,(51.00±12.23)%and(49.83±,9.89)%respectively,which were significantly lower than that in control group[(68.10±8.62)%],and there was a significant difference between different groups(F=12.862,P<0.05). In less than 10 years,10~19 years,more than 20 years and cancer groups,the proportion of CD3+CD4+cells cells was (31.35±6.62)%,(28.38±8,66)%,(24.13±6.46)%and(19.17±4.96)%respectively,which wag significantly lowerthan that in control group[(34.28±7.32)%],and significant in a-group difference was found(F=10.455, P<0.05).The percentages of CD19+cells in more than 20 yeats and cancer groups[(9.00±5.32)%,(9.00± 3.29)%]were lower than that in control group and less than 10 years group[(11.80±3.43)%,(12.35±4.53)%] (P<0.05),while no statistical difference was found between other groups.The expression rates of CD25 and CD38 in lymphocytes of cancer group[(17.96 ±4.98)%,(41.38±8.54)%]were obviously higher than those in control group[(13.10±338)%,(28.60±5.51)%]and there were statistical differences between the experimental groups(P<0.05).The expression rate of HLA-DR in 10~19 years groups[(18.20±6.25)%]was significantly higher than that in control group[(10.72±7.06)%]and less than 10 years group[(11.78±5.13)%],while it was the same in more than 20 years and cancer group[(20.30±8.01)%,(21.82±10.97)%].Conclusions Reduction of cellular immune function caused by coal arsenic poisoning may be an important mechanism of skin cancer.CelMar immune function may be used as a warning signal of skin cancerization of patients with coal arsenic poisoning.
8.A study on new computer-aided modeling method of hip joint.

The main reason of invalidation of prosthetic hip joint is the prostheses flexibility and shift, dislocation and disjunction. Promoting the long time stability of the prostheses is the key of improving the long term hip joint replacement effect. Former research work was focused on the upper segment of femur, and assumed the acetabulum cup to be a spheric concave, and the external form of acetabulum prostheses was basically semi spheric. This paper presents a method of acquiring the point data on the surface of the hip bone using the reverse engineering technology. By analyzing the acetabulum surface fitting error we use rotating elliptical surface to fit the acetabulum surface, together with the optimal technique to build up the CAD model of acetabulum surface. We compare the fitting error between the sphere fitting and rotating elliptical surface fitting and get the result that the rotating elliptical surface fitting error is smaller than the sphere fitting error, and the rotating elliptical surface can describe the shape of the acetabulum better.

9.Relationship between sCD44v6 expression and TCM differentiation type of gastric carcinoma patients and influence of weitai capsule on the expression.

OBJECTIVETo explore the relationship of TCM type with serum level of soluble CD44v6 (sCD44v6) and different histologic parameters in gastric carcinoma patients and to observe the influence of Weitai capsule (WTC) on the sCD44v6 expression.
METHODSTCM typing and sCD44v6 expression were determined in all the enrolled patients (30 in the control and 32 in the trial group) before operation, and 3-4 courses of chemotherapy was applied to them from 3-4 weeks after operation. To the patients of trial group, oral administration of WTC was given additionally with 4 capsules, 3 times a day for consecutive 3 months.
RESULTSsCD44v6 was significantly positive correlated with the degree of cancer cell differentiation, infiltration and lymph node metastasis; (2) Level of sCD44v6 was the highest in patients of blood stasis type, as compared with that in the patients of Pi-deficiency type or of damp-heat type, the difference was significant; (3) After ending treatment, level of sCD44v6 in the trial group was significantly lower than that in the control group.
CONCLUSION(1) Serum level of sCD44v6 could be taken as the criterion for evaluating the development and prognosis of gastric cancer, as well as the therapeutic target for anti-metastasis treatment; (2) Serum level of sCD44v6 is related to some extent with TCM type of blood stasis and Pi-deficiency; (3) WTC combined with chemotherapy could further inhibit the expression of serum sCD44v6 in gastric carcinoma patients.

10.SRY gene-testing in the diagnosis of disorders of sex development among children.

OBJECTIVETo investigate the value of direct sequencing of sex-determining region Y (SRY) gene, as well as peripheral blood karyotype analysis, in the diagnosis of disorders of sex development (DSD) among children and adolescents with ambiguous genitalia.
METHODSThe karyotypes of 20 children and adolescents with ambiguous genitalia were determined by conventional G-banding analysis. PCR amplification was used to detect SRY gene in these patients, and direct sequencing was used to judge whether there was SRY gene mutation.
RESULTSOf the 20 cases, 17 were positive for SRY gene, and 3 were negative for SRY gene. Direct sequencing revealed no SRY gene mutation in the positive cases, however karyotype analysis found 4 special karyotypes in these patients: 46, XY, del(Y) (q12)/45, X; 46, XY, add(Y) (p11); 46, XY, r(9); 46, XY, 9qh+.
CONCLUSIONSSRY gene detection can help determine the type of DSD among children and has the advantage of quick detection. Used together with G-banding analysis, it is helpful for primary diagnosis of DSD among children.
