1.Agreement of anterior chamber angle examination by slit lamp optical coherence tomography and gonioscopy

Objective To explore the agreement of anterior chamber angle examination by slit lamp optical coherence tomography (SL-OCT) and gonioscope.Design Case series.Participants Thirteen patients with primary glaucoma (26 eyes) and 8 normal persons(16 eyes).Methods Anterior chamber angle was measured with SL-OCT and gonioscope in turns for temporal,nasal,superior and inferior quadrant.Results of two methods were analyzed and the data were analyzed with Pearson correlation coefficient and Kappa value.Main Outcome Measures Anterior chamber angle.Results The Pearson correlation coefficient of the two methods was 0.86(P=0.00)and the Kappa value is 0.75 (P=0.00).The specificity and sensitivity of detecting occludable angle were 94.7% and 89.4%.Conclusions Anterior chamber angle examination with SL-OCT and gonioscope are well consistent.The specificity and sensitivity for SL-OCT in detecting occludable angle is satisfying.SL-OCT can be regard as an objective assistant method for the diagnosis of angle closure glaucoma.
2.Clinical observation on acupuncture plus subtle adjustment manipulation of the cervical spine for peripheral facial paralysis

Objective: To observe the clinical efficacy of acupuncture plus subtle adjustment manipulation of the cervical spine for peripheral facial paralysis.Methods: A total of 60 patients with peripheral facial paralysis who met the screening criteria were selected and divided into two groups according to the method of random number table, with 30 cases in each group. The patients in the acupuncture plus tuina group received routine treatment of acupuncture and subtle adjustment manipulation of the cervical spine, while the patients in the acupuncture group only received the same acupuncture treatment. Assessment of peripheral facial paralysis with integrated traditional Chinese and Western medicine was adopted for rehabilitation assessment. Results: The total effective rate of the acupuncture plus tuina group was 93.3%, while that of the acupuncture group was 70.0%, and there was a significant between-group difference (P<0.05). After treatment, the total scores of rehabilitation assessment in both groups were significantly higher than those before treatment (both P<0.05), and the score of the acupuncture plus tuina group was significantly higher than that of the acupuncture group (P<0.05). Conclusion: The treatment of acupuncture plus subtle adjustment manipulation of the cervical spine can improve the function of facial muscles in patients with peripheral facial paralysis, and its clinical efficacy is better than that of the acupuncture treatment alone.
3.Prediction of temporal trends in gastric cancer mortality in Linzhou city from 1988 to 2010.

OBJECTIVETo describe the temporal trends in the mortality rate of gastric cancer during the period of 1988 and 2010, and to predict the gastric cancer mortality between 2016 - 2020.
METHODSThe data of gastric cancer mortality in Linzhou city between 1988 and 2010 was extracted from the cancer registry, including a total of 11 714 cases, covering 22 447 073 person-years. The mortality rate of gastric cancer of each 5-year period was calculated by sub-site and gender. Age-standardized rate (ASR) was calculated using the Chinese standard population in 1982. Intrinsic estimator (IE) model was used to fit the mortality trend by sub-site and gender, and to predict the mortality of gastric cancer in Linzhou city between 2016 and 2020.
RESULTSFrom 1988 to 2010, the gastric cancer mortality in Linzhou city was 52.18/100 000 (11 714/22 447 073) with the ASR at 49.23/100 000; the mortality in male was 67.02/100 000 (7678/11 455 512) with ASR at 68.68/100 000 while the mortality in female was 36.72/100 000 (4036/10 991 561) with ASR at 32.12/100 000. The mortality of cardia carcinoma was 27.87/100 000 (6257/22 447 073) with the ASR at 26.37/100 000; while the mortality of non-cardia carcinoma was 24.31/100 000 (5457/22 447 073) with the ASR at 22.86/100 000. The ASR of gastric cancer during 1988 - 1990 was 63.37/100 000 (1653 cases) and decreased by 28.34%, to 45.41/100 000 (2622 cases) during 2006 - 2010. The IE model showed that the birth cohort effect decreased greatly. The mortality risk of cardia carcinoma in population born after 1950s, decreased significantly; and the mortality risk of non-cardia carcinoma in population born in 20 century continually decreased. The death of gastric cancer among the population over 30 years old was predicted to be 3626 cases, increasing by 40.60% compared with the number between 2006 and 2010 (2579 cases). Among them, the mortality of cardia carcinoma increased by 51.89% (predicted number between 2016 and 2020 was 2456 cases, and 1617 cases between 2006 and 2010), and the mortality of non-cardia carcinoma increased by 21.62% (predicted number between 2016 and 2020 was 1170 cases, and 962 cases between 2006 and 2010).
CONCLUSIONThe mortality rate of gastric cancer in Linzhou city showed a decreasing trend during the period of 1988-2010, being mainly attributed to the cohort effect. However, the mortality will still increase in the future, between 2016 and 2020.

4.Construction of pcDNA3.1(+)/A2E eukaryotic expression vector and its expression on K562 cell.

To construct pcDNA3.1(+)/A2E eukaryotic expression vector and obtain a stable expression on HLA-I negative human K562 cell, PCR technique was employed to amplify A2E cDNA from the multi-cistron expression vector pG/A2E carrying HLA-E and HLA-A2 cDNA through internal ribozyme entry site (IRES), the cDNA was subcloned into vector pcDNA3.1(+), thus a eukaryotic expression was constructed and named pcDNA3.1(+)/A2E; then, the recombinant plasmid was transferred into the target cells, followed by screening with G418 and limiting dilution; finally, flow cytometry was adopted to detect HLA-E expression on the target cells. The results showed that HLA-E molecules were successfully expressed on K562 cells transfected with pcDNA3.1(+)/A2E (27.76%) and the expression of HLA-E molecules was not detected on K562 cells transfected with pcDNA3.1(+). It is concluded that the pcDNA 3.1(+)/A2E eukaryotic expression vector was successfully constructed and the HLA-E molecules were expressed on K562 cells. The data presented here would be expected to lay a good basis for the research of the molecular mechanism of HLA-E function and the interaction between HLA-E and the receptor on NK cells, as well as the influence of the expression of HLA-E in vitro on NK cells.

5.Study on the expansion of megakaryocyte progenitors in vitro from cord blood.

This study was aimed to investigate the effect of various cytokines on megakeryocytes expansion in vitro from human cord blood CD34(+) cells in order to establish an optimal culture system for MK expansion. Mononuclear cells were obtained by Ficoll-Hapaque density gradient separation. CD34(+) cells were positively isolated using a CD34 progenitor cell isolation kit. CD34(+) cells were placed into 24 well plates at a concentration of 2 x 10(4) per well. Each well contained 1 ml of IMDM with the present of effective MK cells growth cytokines. Clonogenic potentials of MK progenitor were assayed using a methylcellulose cultures system. The results suggested that four cytokines (IL-3 + IL-6 + TPO + FLT3L) culture system could effectively induce and expand cord blood CD41(+) MK cells. The number of CD41(+) cells expanded 154.67 +/- 32.21-fold on day 7, and 193.23 +/- 25.24-fold on day 14. In conclusion, established expansion system in vitro for MK cells provides experimental foundation for recovery of platelets after cord blood transplantation.

6.Expressions of CD1a and CD83 of Langerhans cells in the local lesions of epidermodysplasia verruciformis patients.

OBJECTIVETo investigate the expressions of CD1a and CD83 of Langerhans cells (LC) in the lesions of epidermodysplasia verruciformis (EV) patients.
METHODSWe used immunohistochemical method to detect the expressions of CD1a and CD83 in the lesions of 10 patients with EV lesions and in the skins of 10 normal subjects.
RESULTSNo CD83 + LCs was detected in all EV patients and normal controls, but CD1a + LC was found in all cases. The quantity of CD1a + LCs in the lesions of EV patients was significantly lower than that in the normal skin (P < 0.01); furthermore, the distribution of LCs in EV lesions was uneven.
CONCLUSIONThe functions of LCs may be inhibited in EV patients.

7.Chemical components in essential oils from tender branches and leaves of Rhododendron.

AIMTo analyze and compare the compositions in essential oils from branches and leaves of Rhododendron simsii Planch. and Rhododendron naamkwanense Merr.
METHODSEssential oils were extracted by water distillation according to Chinese Pharmacopoeia and analyzed by capillary gas chromatography-mass spectrometry as well as chemometrics resolution method and authentic compounds. The relative contents of each component in the essential oils were obtained by normalization of peak areas.
RESULTSA total of 124 components were identified, of which 48 compounds were existed in both of the samples. Ninety four compounds accounted for 84.47% of the essential oil from Rhododendron simsii Planch. and seventy eight components accounted for 90.25% of the total essential oil from Rhododendron naamkwanense Merr. were identified. 72.76% and 88.07% of the components in Rhododendron simsii Planch and Rhododendron naamkwanense Merr., respectively, included oxygen element. They are mainly terpenol, acids and esters. 1-octen-3-ol (4.00%, 7.90%), 1,6-octadien-3-ol, 3,7-dimethyl-(12.60%, 3.48%), 9,12,15-octadecatrienoic acid, [Z, Z, Z]- (1.15%, 45.34%), phytol (15.21%, 8.56%), p-menth-1-en-8-ol (2.15%, 3.29%), and 9,12,15-octadecatrienoic acid, ethyl ester, [Z,Z,Z]- (9.16%, 8.01%) were their common main compounds, which accounted for 44. 27% and 76.58% of the total amount of the two essential oil samples, respectively. In addition, n-hexadecanoic acid (7.73%), 9,12-octadecadienoic acid (1.85%) and tetracosanoic acid, methyl ester (1.38%) were also the main compounds in essential oil from Rhododendron simsii Planch.
CONCLUSIONMuch higher reliability and accuracy were obtained with the help of chemometrics resolution method and authentic n-alkane standard solutions than those of using GC-MS alone.

8.Quantitative study of dynamic magnetic resonance perfusion-weighted imaging in rabbit models of implanted hepatic VX2 tumor.

OBJECTIVETo investigate the possibility of dynamic magnetic resonance (MR) perfusion-weighted imaging (PWI) in rabbit models of implanted hepatic VX2 tumor.
METHODSMRI and PWI images were obtained from 16 rabbit models of implanted hepatic VX2 tumor and 4 normal rabbits at 14, 18, 22, and 26 days after tumor implantation, respectively, and the mean time to enhancement (MTE) and maximum slope of increase (MSI) were estimated and compared with pathological changes.
RESULTSIn the rabbit models of implanted hepatic tumor the MTE tended to increase and MSI to descend 14, 18, 22, and 26 days after tumor implantation, showing significant differences from the control group. Coagulation necrosis and fibrous hyperplasia in the tumors appeared to progress with time as found pathologically.
CONCLUSIONDynamic MR PWI quantitative analysis of rabbit models of implanted hepatic VX2 tumor provides valuable means for observing the growth behaviors of VX2 tumor and may be clinically applicable in the evaluation of hepatic diseases.

9.Clinical trial of fluticasone propionate on children with allergic rhinitis

Objective To observe the clinical efficacy of fluticasone propionate aerosol in children with allergic rhinitis and its effect on osteopontin (OPN),interleukin-4 (IL-4) and intedeukin-5 (IL-5)expression.Methods Sixty children with allergic rhinitis were randomly divided into control group and treatment group,30 cases in each group.Control group was treated with ketotifen 1 mg,twice daily.The treatment group was treated with fluticasone propionate aerosol,100 μg each time,twice a day for 2 months.The levels of interleukin (IL)-4,IL-5 and integral optical density (IOD) of immunohistochemical staining of osteopontin in two groups were observed.The clinical effect and the number of eosinophils,neutrophils and lymphocytes in peripheral blood of 1,3,6 months after operation were observed.Results Mter treatment,the levels ofIL-4,IL-5 and OPN in treatment group were (66.31 ± 18.00) ng· mL-1,(62.12 ± 0.33) ng· mL-1,5589.71 ± 267.00,had significant difference with those in control group,which were (85.32 ± 12.34) ng · mL-1,(30.24 ± 0.30) ng · mL-1 and (15.00 ±0.26) + 103(P <0.05).The number of eosinophils,neutrophils and lymphocytes in 6 month after treatment in treatment group were (0.21 ±0.01) × 109/L,(0.10 ±0.01) × 109/L,(0.14 ±0.01) + 109/L.The number of eosinophils,neutrophils and lymphocytes in control group at 6 months after treatment were (0.21 ±0.02) + 109/L,(0.10 ±0.01) + 109/L,(0.14 ±0.01) + 109/L respectively,all had significant difference with those in 1 month after treatment (P < 0.05),with no significant between the two groups (P >0.05).The total effective rate was 93.33% (28/30 cases) in treatment group and 36.67% (11/30 cases) in control group (P <0.05).The incidence of adverse drug reaction in treatment group was 3.33% (1/30 cases).In control group,there were 4 cases of skin allergy,3 cases of thrush,3 cases of bronchospasm with asthma,theincidence of adverse drug reactions was 33.33 % (10/30 cases,P < 0.05).Conclusion Fluticasone propionate aerosol treatment of children with allergic rhinitis have significant clinical efficacy.
10.Adenovirus-mediated glucose transporter 3 infection in PC12 cells of rats after hypoxiareperfusion injury

[Objective]To investigate the effect of adenovirus-mediated glucose transporter 3 (Ad-GLUT3) infection on PC 12 cells of rats after hypoxia-reperfusion injury.[Methods]PC 12 cells treated with hypoxia and re--oxygenation were prepared;after PC12 cells being infected by Ad-GLUT3 (experimental group),adenovirus encoding green fluorescent protein (Ad-GFP group) and blank plasmid (control group),respectively,flow cytometry (FC) and TUNEL assay were used to analyze the apoptosis of PC12 cells;the glucose uptake rate was detected by isotopic-scintiscan,and the expression of nuclear factor (NF-kB) was detected by Western blotting.[Results] FC indicated that the apoptotic rate in the experimental group (4.7%±1.75%) was significantly lower as compared with that in the Ad-GFP group (14.25%±3.46%) and control group (13.1%±2.98%,P<0.05);TUNEL assay showed that the apoptotic rate in the experimental group (7.7%±3.31%) was significantly lower as compared with that in the Ad-GFP group (17.1%±3.10%) and control group (15.2%±2.70%,P<0.05).And the glucose uptake rate in the experimental group (603%±13.1%) was obviously higher as compared with that in the Ad-GFP group (300%±8.77%) and control group (319%±11.2%,P<0.05);and the NF-k<B level in the experimental group (0.92±0.17) was significantly lower as compared with that in the Ad-GFP group (1.02±0.24) and control group (1.06±0.21,P<0.05).[Conclusion] After being infected by Ad-GLUT3,the PC12 cells treated with hypoxia and re-oxygenation enjoy elevated glucose uptake rate and inhibited apoptotic rate.
