3.Effect of telmisartan and fosinopril on the expression of angiotensin-con-vertion enzyme 2 in cardiomyocyte after myocardial infarction
Journal of Chongqing Medical University 2007;0(11):-
Objective:To investigate the effect of telmisartan and fosinopril on the expression of angiotensin-convertion enzyms 2 (ACE2) in cardiomyocyte after myocardial infarction. Methods:Animal model of acute myocardial infarction were established by ligating the left anterior descending coronary artery of rats. Vehicle, telmisartan, fosinopril,or both drugs combined were giv- en to rats by intubation feeding, from 2 weeks before coronary artery ligation to 2 weeks there after. ACE2 protein expression of myocardium were semi-quantified by SP immunohistochemistry. ACE2mRNA were measured by Reverse Transcriptase Poly- merase Chain Reaction (RT-RCR). Results:Compare with sham operated group,ACE2 protein increased significantly in infarc- tion group, but not the ACE2 mRNA. telmisartan and fosinopril therapy caused a significant increase in ACE2 protein compare to infarction group, but only telmisartan caused a significant increase in ACE2 mRNA though all therapied groups showed higher ACE2 mRNA expression. Conclusion:Both telmisartan and fosinopril induced increases in cardiac ACE2 exprssion, whereas the combination of these two drugs do not associated with higher ACE2 expression.
4.Utllizition of ELISA in determination of monoclonal antibody affinity constant in ascites
Chinese Journal of Immunology 1985;0(02):-
Antibody affinity influences the ant igen-antibody reaction in vivo and in vitro. It is important to routinely determine the antibody affinity in order to elucidate the impacts of this antibody characteristics on the immunoassay and the pathogenetic mechanisms of human diseases. In this paper, a simple method (modified Friguet' s) was reported to directly measure the experimental affinity data of monoclonal antibody in ascites by means of ELISA and to accurately determine the corresponding affinity constants of monoclonal antibody by amendment of underestimation of affinity constants due to the intrinsic properties of Friguet' s, and shows convenience and reliability.
5.Estrogen, anti-estrogen drugs, and thyroid cancer
Chinese Journal of Endocrinology and Metabolism 2014;30(12):1128-1131
Several researches have suggested that estrogen contributes to the initiation and development of thyroid cancer by binding to estrogen receptors (estrogen receptor α,estrogen receptor β,and G-protein-coupled receptor 30),activating gene or non-genomic pathways and regulating proliferation of thyroid cells.Studies on antiestrogen drugs based on inhibiting thyroid cell proliferation may provide a new target in treating thyroid cancer.
6.Coaxial double catheter technique in intracranial posterior circulation stenting
Chinese Journal of Tissue Engineering Research 2009;13(17):3375-3378
BACKGROUND: The posterior circulation artery is characterized by small caliber, long traveling, many curves and branches, so a stronger resistance will be encountered when a stent is delivered.OBJECTIVE: To investigate the effects of coaxial double catheter technique on intracranial posterior circulation stenting.DESIGN, TIME AND SETTING: A retrospective case analysis was performed at the Department of Neurology, Affiliated People's Hospital of Yunyang Medical College between December 2004 and February 2008.PARTICIPANTS: A total of 21 patients, 11 males and 10 females, aged 50-76 years old, were se/ected from the Department of Neurology, Affiliated People's Hospital of Yunyang Medical College.METHODS: All patients were confirmed as posterior circulation intracranial stenosis (stenosis > 70%) by digital subtraction angiography (DSA), magnetic resonance angiography (MRA), or CT angiography (CTA). They underwent cerebrovascular stent implantation, during which, single catheter technique was first applied, if failed, a coaxial double catheter technique was followed. A conventional group (single catheter technique successfully applied) and a double catheter group (coaxial double catheter technique successfully employed) were set. A written informed consent was obtained from each patient.MAIN OUTCOME MEASURES: Therapeutic results and biocompatibility of implanted stents.RESULTS: The single catheter technique was applied in 21 patients and 6 of them (28.57%) had successful stent deployment, The remaining 15 patients were managed by a coaxial double catheter technique, and the stent was successfully released in these patients. There was only 1 patient presenting with subarachnoid hemorrhage, and the success rate of stent deployment was 93.33%. Chi-square tests results showed that the coaxial double catheter technique produced significantly higher success rate of stent implantation than the single catheter technique (P < 0.05). Except this, no other adverse reactions were found. All stents covered the stenosis lesions completely, and fitted well with the blood vessel wall without rejection, showing the good biocompatibility of selected stents with the host.CONCLUSION: The coaxial double catheter technique can increase the longitudinal supporting strength and enhance the success rate of the intracranial posterior circulation stenting. Meanwhile, the risks of this procedure are low. Therefore, the technique deserves to be popularized.
7.Determination of serum osteopontin in pulmonary tuberculosis patients and its clinical significance
Chinese Journal of Postgraduates of Medicine 2012;35(1):36-39
ObjectiveTo investigate osteopontin (OPN) level in pulmonary tuberculosis and its clinical significance.MethodsThe level of serum OPN was determined in 80 pulmonary tuberculosis patients before anti-tuberculosis treatment[patients group, simple pulmonary tuberculosis with 60 cases (simple pulmonary tuberculosis group),pulmonary tuberculosis combined with tuberculous pleurisy with 20cases (pulmonary tuberculosis combined with tuberculous pleurisy group)],and compared with that of 30healthy persons (healthy control group) and 20 patients having received effective anti-tuberculosis treatment and followed-up for 6 months (followed-up group). At the same time, the clinical significance of OPN was studied by other commonly used clinical indicators of tuberculosis.ResultsThe level of serum OPN in patients group [(843.49 ±569.23) ng/L] was significantly higher than that in healthy control group [(352.50 ± 185.02) ng/L,P <0.05].The level of serum OPN after treatment in followed-up group were significantly decreased compared with those before treatment [(494.11 ±352.40) ng/L vs.(1106.60 ± 628.39) ng/L,P< 0.05 ] and there was no significant difference compared with those in healthy control group (P > 0.05 ).The level of serum OPN in pulmonary tuberculosis combined with tuberculous pleurisy group [(1179.80 ±606.33) ng/L] was significantly higher than those in simple pulmonary tuberculosis group [(727.53 ±511.66) ng/L,P <0.05],and they were both higher than those in healthy control group [ ( 352.50 ± 185.02 ) ng/L,P < 0.05 ].Therefore, for further additional subgroup analysis in simple pulmonary tuberculosis group, the level of serum OPN in patients with cavity[ ( 836.51 ± 549.80) ng/L ] was significantly higher than that without cavity [ (535.52 ± 375.95 ) ng/L,P < 0.05 ],while the levels of serum OPN had no statistical difference between pulmonary lesion involved < 3 leaf patients and ≥ 3 leaf patients, sputum mycobacterium tuberculosis positive patients and negative patients, mycoDot test results positive patients and negative patients, erythrocyte sedimentation rate elevated patients and normal patients,PPD strongly positive patients and non-strongly positive patients (P>0.05).ConclusionsOPN activity is closely related with pulmonary tuberculosis.OPN can serve as a judge of pulmonary tuberculosis and the clinical indicators of disease outcome. The level of serum OPN and the severity of pulmonary tuberculosis have some correlations.
8.Diagnosis and treatment of acute superior mesenteric artery occlusion
Chinese Journal of General Surgery 2009;24(9):722-724
Objective To discuss the manifestations, diagnosis and management of acute superior mesenteric artery occlusion (ASMAO). Methods The clinical data of 15 cases of ASMAO admitted in our hospital between 2000 and 2007 were analyzed retrospectively. Results The causes of ASMAO were mesenteric artery embolism in 11 patients and mesenteric artery thrombosis in 4 patients. All cases underwent emergency laparotomy, in which 2 cases underwent removal of the embolus, 11 cases were subjected to resection of necrotic bowel after removal of the embolus, and 2 cases gave up definite surgery. Postoperative heparin anticoagutherapy was used in all patients. The overall mortality rate was 27% (4 cases). Conclusions Acute superior mesenterie artery occlusion should be suspected in a patient with sudden abdominal pain, especially in those who have organic cardiac disease. Early diagnosis and prompt embolectomy are the key to save the patients.
9.The synergistic antitumor effects of berberine alpha-hydroxy-beta-decanoylethyl sulfonate with hydroxycamptothecine and its effect on topoisomerase.
Acta Pharmaceutica Sinica 2011;46(4):390-4
Synergistic antitumor effects of HB (berberine alpha-hydroxy-beta-decanoylethyl sulfonate, houttuyn berberine) with HCPT (hydroxycamptothecine), and its correlative mechanism were studied in vitro. MTT assay was employed to determine the cytotoxicity of HB combined with HCPT in tumor cells culture in vitro, IC50 and combination index (CI value) were used to evaluate the synergistic effects. The supercoiled DNA relaxation mediated by topoisomerase I & II was measured by agarose gel electrophoresis assay, and influence of HB was detected. The results showed that HB could inhibit the proliferation of tumor cells (SGC-7901, SW1116 and SW480) in vitro, and the inhibition ratio was increased, IC50 was reduced when combining with HCPT. CI value of the two drugs was less than 1 in HepG2, SW480, SGC-7901 and SW1116 cells. The lowest value was 0.447, 0.626, 0.161 and 0.178 in these tumor cells, respectively, further indicating HB has synergistic action with HCPT on suppressing tumor proliferation. The agarose gel electrophoresis assay showed HB can inhibit topoisomerase I & II activity of SW480 cells at the concentration of 2.0-8.0 mg x L(-1). HCPT is a typical inhibitor of topoisomerase I , the synergistic action between HCPT and HB on suppressing tumor proliferation is perhaps related to the congenerous inhibition of topoisomerase.
