Objective To study the changes and significance of Bid gene expression in hypoxic-ischemic brain damage (HIBD) in neonatal rats.Methods The animal models of HIBD were made. Total RNA from ipsilateral cerebral hemisph ere was extracted.RT-PCR was used to evaluate the levels of Bid gene expression at different time points after hypoxic-ischemic. Results The levels of Bid mRNA were higher in HIBD rats than those in normal rats.The ex pression of Bid mRNA in ipsilateral hemiphere following cerebral hypoxic-ischem ic began to increase at 6 h, increased to the highest level at 24 h, and then gr adually returned.Conclusion The upregulation of Bid mRNA can play a role in the induction of apoptosis fol lowing cerebral hypoxic-ischemic. 2.Effect of zinc gluconate combined with Danshen injection on the plasma lipid level following exposure to noise in rats

Objective To investigate the effects of zinc gluconate combined with Danshen injection on the plasma lipid level following exposure to noise in rats. Methods Fifty female SD rats were randomly divided into a control group, a noise exposure group (model group), a zinc gluconate + noise exposure group (zinc gluconate group), a Danshen injection+noise exposure group (Danshen injection group) and a zinc gluconate+Danshen injection+noise exposure group (combination group), 10 rats in each group. The zinc gluconate group received intragastric administration of zinc gluconate 8-10 mg/(kg?d), the Danshen injection group received intraperitoneal injection of Danshen injection 6 mg/(kg?d), the combination group received intragastric administration of zinc gluconate 8-10 mg/(kg?d) and intraperitoneal injection of Danshen injection 6 mg/(kg?d). On the day 10, except in the control group, the rats in the rest of the groups were continuously exposed to high frequency steady noise for 7 days (4 000 Hz, 95 dB, 3 h/d). The serum levels of total cholesterol (TC), triglycerides (TG), high density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were detected. Results The serum levels of TC (1.88 ± 0.17 mmol/L vs. 1.55 ±0.09 mmol/L), TG (1.45 ± 0.38 mmol/L vs. 1.01 ± 0.27 mmol/L), LDL-C (0.29 ± 0.04 mmol/L vs. 0.23 ± 0.03 mmol/L) in the model group were significantly lower than those in the control group (P<0.01 or P<0.05), while the serum HDL-C level in the model group were significantly lower than that in the control group (0.55 ± 0.08 mmol/L vs. 0.62 ±0.06 mmol/L;P<0.01). The serum TC level in the zinc gluconate group was significantly lower than that in the model group (1.71 ± 0.11 mmol/L vs.1.88 ± 0.17 mmol/L;P<0.05). The serum levels of TC (1.73 ± 0.21 mmol/L vs. 1.88 ± 0.17 mmol/L) and LDL-C (0.25 ± 0.03 mmol/L vs. 0.29 ± 0.04 mmol/L) in the Danshen injection group were significantly lower than those in the model group(all P<0.05). The serum levels of TC (1.57 ± 0.21 mmol/L vs. 1.88 ± 0.17 mmol/L), TG (0.84 ±0.40 mmol/L vs. 1.45 ± 0.38 mmol/L) and LDL-C (0.24 ± 0.05 mmol/L vs. 0.29 ± 0.04 mmol/L) in the combination group were significantly lower than those in the model group (P<0.01 or P<0.05), and the serum HDL-C level (0.61 ± 0.07 mmol/L vs. 0.55 ± 0.08 mmol/L) in the combination group was significantly higher than that in the model group (P<0.05). Conclusions Noise exposure may induce dyslipidemias in rats. Zinc gluconate and Danshen injection can regulate the plasma lipid level following exposure to noise in rats, and combination of zinc gluconate and Danshen injection may be superior to zinc gluconate or Danshen injection alone.
3.Protective effect of Zinc and Salvia on the immune system caused by noise in rats

Objective To observe the protective effect of the zinc gluconate combined with Salvia injection on noise induced serum immunoglobulin(IgG,IgM,IgA) and immune molecules (IL-2,IL-4) in rats.Methods 50 female SD rats were randomly divided into control group,noise group,zinc gluconate noise group(plus zinc group),Salvia injection noise group (plus Salvia group) and Zinc gluconate oral liquid and Salvia injection noise group(combined group),10 rats in each group.Except the control group,rats in the other groups were continuously exposed to high frequency steady noise for two weeks.The concentrations of serum immunoglobulin (IgG,IgM,IgA) and immune molecules (IL-2,IL-4) after the intervention of noise were compared.Results Serum levels of IgG,IgM,IL-2,IL-4of the control group were significantly higher than the noise group (all P < 0.05),plus zinc group (all P < 0.05),plus Salvia group(all P <0.05),the differences were statistically significant.Serum level of IgA of each group was not statistically different compared with control group (all P > 0.05).Serum levels of IgG,IgM,IL-2,IL-4 in the noise group were significantly lower than those in the control group (all P < 0.05),combined group (all P < 0.05),the differences were statistically significant.Serum levels of IgG,IgM,IL-2 in plus zinc group were significantly higher than the noise group(all P < 0.05).Serum IgA concentration of each group showed no significant difference compared with noise group(all P > 0.05).Conclusion Zinc gluconate oral solution and Salvia injection have protective effect on immune system affected by noise in rats,the protective effect of combination of zinc gluconate oral solution and Salvia injection is stronger than the individual.
4.Protective effect of shenfu injection against ischemia-reperfusion injury due to pancreas transplantation in rats.

OBJECTIVETo investigate the protective effect of Shenfu Injection against ischemia-reperfusion (I/R) injury due to pancreas transplantation in rats, and explore its possible mechanism.
METHODSSix normal SD rats with sham operation were taken as the normal control group, 24 steptozozin-induced diabetic SD rats were randomly divided into 4 groups, with 6 in each group. Except I/R group, the rats in the other groups were intravenous injected with Shenfu Injection (SF,10 mg/kg), Hongshen Injection (HS, 9 mg/kg) and Fuzi Injection (FZ 1 mg/kg) respectively at the day and 30 minutes before pancreas transplantation performed in the SF group, HS group and FZ group, respectively. At the same time, rats in the normal control group and in the I/R group were intravenously injected the same volume of normal saline. The blood glucose was detected before and after reperfusion, and 2 hours later after reperfusion, the contents of serum nitric oxide (NO) and tumor necrosis factor-alpha (TNF-alpha), the concentrations of malondialdehyde (MDA) , superoxide dismutase (SOD) , and myeloperoxidase (MPO) in the transplanted pancreas tissues were detected. The cell apoptosis of the transplanted pancreas tissue was determined by TUNEL, and the bcl-2 and Bax protein expression was determined by Western blot.
RESULTSAfter reperfusion, the levels of blood glucose and TNF-alpha decreased and the concentration of NO increased in the SF group, HS group and FZ group, compared with those in the I/R group. The activity of SOD, bcl-2 expression and the ratio of bcl-2 and Bax were higher, while the content of MDA, the activity of MPO, apoptotic indexes, and Bax expression were lower in the SF group, HS group and FZ group than those in the I/R group.
CONCLUSIONShenfu Injection can protect L/R injury due to pancreas transplantation in rats, the possible mechanism may be related to promoting activity of SOD, increasing synthesis of endogenous NO, decreasing the excretion of TNF-alpha, alleviating conglutination and aggregation of polymorphonuclear neutrophils (PMNs) in pancreas, as well as up-regulating Bcl-2 gene expression and down-regulating the Bax gene expression.

5.Effect and mechanism of epidermal growth factor on proliferation of GL15 gliomas cell line.

The effects of epidermal growth factor (EGF) on proliferation of G15 glioma cells and the possible mechanisms were investigated. GFAP and EGFR expression was detected by immunohistochemical method. After the cells were treated with EGF at different concentrations, cell count method was used to determine the proliferation of glioma cells, cell cycle and apoptosis were analyzed by flow cytometry (FCM), and laser scan confocal microscope (LSCM) was used to measure the cytoplasmic free calcium. The results showed that GFAP was diffusedly expressed in GL15 cells and EGFR was over-expressed. EGF at doses of < or =1 ng/mL could significantly stimulate cell proliferation, cells in phase G0/G1 decreased, and those in phase S increased. EGF at doses of 10 and 100 ng/ml could inhibit the cell proliferation significantly, and the apoptosis ratio in high dose of EGF group was higher than in control group. EGF could significantly induce a quick rise of intracellular free calcium, but the peak value of intracellular free calcium activated by high dose of EGF was higher than by low dose of EGF. It was suggested that EGF had a dual effect on gliomas: low dose of EGF could stimulate the cell proliferation of gliomas, but high dose of EGF could induce the cell apoptosis and inhibit the proliferation of gliomas, which might be contributed to the difference of intracellular free calcium.

Objective To explore role of 64-slices spiral CT in differetiation of acute chest pains.Methods Thirty six patients with acute chest pains were performed 64-slices spiral CT chest angiography.Two-dimensional and three-dimensional reconstruction was performed in all patients by means of multiplanar reconstruction(MPR)(coronal,sgittal oblique),curved planar reformation(CPR), maximum intensity projection(MIP),and volume rendering(VR).All images were blindly reading by two experienced radiologist.DSA were performed at the same time in 16 cases.Results The coronary artery branches,pulmonary artery and aortic artery in all patients were showed clearly,The acute myocardial infarction were showed in 10 cases,The pulmonary artery embolism in 14 cases,The aortic dissection in 6 cases respectively,The Coronary embolism in One case ,pneumothorax In One case The constrictive pericarditis in 1 case respectively.Normal findings in 4 cases.Conclusion 64-slices spiral CT is a useful and noninvasive examination in acute chest pain.
7.Effects of RNAi-mediated gene silencing of LRIG1 on proliferation and invasion of glioma cells.

The effects of RNAi-mediated gene silencing of LRlG1 on proliferation and invasion of the human glioma cell line U251-MG and the possible mechanisms were explored in this study. The plasmids pGenesil2-LRIG1-shRNA1 and pGenesil2-LRIG1-shRNA2 were transfected into U251-MG glioma cells respectively by using Lipofectamine 2000 and the transfected cells in which the LRIG1 expression was stably suppressed were selected by G418. The cells transfected with negative shRNA served as control. The expression levels of LRIG1 mRNA and protein were measured by qRT-PCR and Western blotting, respectively. The cell cycle was analyzed by flow cytometry. The results showed that LRIG1 mRNA expression was reduced by 70% and 58% and LRIG1 protein expression by 58% and 26% in U251-MG cells transfected with pGenesil2-LRIG1-shRNAl and pGenesil2-LRIG1-shRNA2 relative to the negative shRNA-transfected U251-MG cells. The proliferative capacity of the LRIG1 specific siRNA-transfected cells was stronger than that of control cells. Cell cycle analysis showed that silencing LRIG1 significantly increased the percentage of S phase cells and the proliferation index (P<0.01). Moreover, silencing LRIG1 could promote the invasion of U251-MG cells (P<0.05). These findings suggested that LRIG1-targeting siRNA can exert a dramatically inhibitory effect on RNA transcription and protein expression of LRIG1, and LRIG1 down-regulation could promote the proliferation of U251-MG cells, arrest U251-MG cells in S phase, and enhance the invasion of U251-MG cells.
8.Maternal periodontal disease and risk of preeclampsia: A meta-analysis.

Research on the association between maternal periodontal disease and the risk of preeclampsia has generated inconsistent results. This meta-analysis was conducted to evaluate the association between maternal periodontal disease and the risk of preeclampsia. A literature search of PubMed and Embase was performed to identify relevant papers published before March 2013. Only observational studies that assessed maternal periodontal disease and the risk of preeclampsia were selected. Patients' periodontal status was examined at different time points during pregnancy or after delivery (at 14-32 weeks of gestation, within 48 h prior to or within 5 days after delivery). Pooled odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were calculated for cases and controls. Cases were defined as women with concurrent hypertension and proteinuria after 20 weeks of gestation. Eleven studies involving 1118 women with preeclampsia and 2798 women without preeclampsia were identified and analyzed. Women with periodontal disease before 32 weeks of gestation had a 3.69-fold higher risk of developing preeclampsia than their counterparts without periodontal disease (OR=3.69; 95% CI=2.58-5.27). Periodontal disease within 48 h prior to delivery was associated with a 2.68-fold higher risk of preeclampsia (OR=2.68; 95% CI=1.39-5.18). Pregnant women with periodontal disease within 5 days after delivery had a 2.22-fold higher risk of preeclampsia than women without periodontal disease (OR=2.22; 95% CI=1.16-4.27). In conclusion, this meta-analysis suggests that maternal periodontal disease is an independent predictor of preeclampsia.
9.MRI diagnosis of cavernous transformation of the portal vein

ObjectiveTo investigate the value of MRI in the diagnosis of cavernous transformation of the portal vein (CTPV).MethodsPlain MRI,dynamic enhanced and (or) dynamic contrast enhanced magnetic resonance angiography(DCE-MRA) findings in 30 patients of clinical-proved CTPV were retrospectively analyzed.ResultsAmong 30 CTPV patients on plain MRI,obliteration of main and (or) branched portal vein were found,and mass-like or reticular abnormal soft-tissue signals were around the vein,which were produced by collateral vessels.On dynamic enhanced MRI,abnormal hepatic perfusion during arterial phase and abnormal enhanced collateral veins during portal phase could be seen.The above signs became more obvious on DCE-MRA.ConclusionsMRI and DCE-MRA can clearly visualize the anatomical features of CTPV.It is important and can provide the reliable evidence for planning properly therapeutic protocol to recognize and directly evaluate the CTPV.
10.Ursolic acid down-regulates COX-2 expression by suppressing the activation of ERK in A549 cells

The antitumor effect and the mechanism of action of ursolic acid in A549 cells(human non-small-cell lung cancer cells) was investigated in this paper. Firstly, MTT assay was performed to test whether ursolic acid could inhibit the growth of A549 cells. Secondly, Western blot was utilized to measure the expression level of COX-2 and the activation of MAPKs. The MTI assay revealed that ursolic acid inhibited the growth of A549 cells. The result of Western blot suggested that ursolic acid inhibited the expression of COX-2 and activation of ERK and ERK specific inhibitor PD98059 suppressed the expression of COX-2 synergistically with ursolic acid. Taken together, our data suggest that ursolic acid can suppress LPS-induced COX-2 expression in A549 cells, which could be due to the inhibition of the activation of ERK.
