Objective To investigate the drug resistance situation and the epidemiological characteristicin the tuberculoupatientin ordeto provide the reference foeffective prevention and treatmenof T.MethodTotally 2 271 patientwith posi-tive sputum smeapulmonary tuberculosi(TB) in the Jiangbei Branch Hospital of Chongqing Zhongshan Hospital from January 2009 to Decembe2013 were performed the drug resistance detection of 10 kindof anti-Tdrugs(isoniazid ,streptomycin ,rifampi-cin ,ethambutol ,para-aminosailcyliacid ,kanamycin ,pasiniazid ,protionamide ,capreomycin ,and ofloxacin) .ResultOf all 2 271 ca-seof positive sputum culture pulmonary T,the total drug resistance rate and total multi-drug resistance rate were 61 .91% and 26 .29% ,respectively ;the total drug resistance ratein initial treatmenand retreatmenpatientwere 52 .85% and 78 .06% respec-tively ,and the multi-drug resistance ratewere 16 .70% and 43 .38% ,respectively ;the top five in the average drug resistance ratefrom high to low were streptomycin(39 .45% ) ,isoniazid(33 .20% ) ,rifampicin(32 .94% ) ,para-aminosailcyliacid(31 .40% ) and pasiniazid(21 .53% );the anti-Ttreatmenhistory ,gende,age and occupation were significantly associated with the drug resist-ance occurrence (P<0 .05) .Conclusion The drug resistance situation of the Tpatientin the local areof Chongqing iseriouin the recen5 year.The standardized managemenand treatmenon the Tpatientwith drug resistance should be strengthened .

Objective:To explore inhibition of telomerase activity and malignant phenotype on breast cancer cells MCF7 by Dominant Negative human telomerase reverase transcriptase gene(DN-hTERT).Methods:DN-hTERT eukaryotic expression vector DN-hTERT-IRES2-EGFP and empty vector(I) IRES2-EGFP were transfected into MCF7 by lipofectamine2000,after being selected by G418,positive clones were obtained;Tthe transfected cells growth was observed with inverted fluorescence microscope.The expression levels of hTERT mRNA of transfected cells were determined by reverse transcriptase polymerase chain reaction(RT-PCR).Telomerase activity of transfected cells was measured by TRAP-ELISE.Results:The proliferation of MCF7/DN cells were inhibited by DN-hTERT transtection;The expression levels of hTERT mRNA increased in MCF7/DN.Telomeric length was shorter in MCF7/DN than that in MCF7/I.The telomerase activity measured by TRAP-ELISE was 2.36?0.12 in MCF7/DN and was 3.32?0.14 in vacant vector MCF7/I.The telomerase activity in MCF7/DN was significantly lower than in vacant vector transfected MCF7/I(P
3. Combination of magnesium ion with mineralized collagen intervenes osteogenic differentiation of mouse preosteoblasts

BACKGROUND: Preliminary study has shown that the composite materials composed of magnesium-based materials and mineralized collagen have a good supporting effect on repairing the critical defects, which can improve the mechanical strength of mineralized collagen and premature collapse during bone healing to some extent. However, magnesium-based metals degrade fast in chloride-containing solutions (including human body fluids or plasma), and the effects of releasing magnesium ions on the proliferation and differentiation of osteoblasts are unknown. OBJECTIVVE: To investigate the effects of magnesium ion combined with mineralized collagen on osteogenic differentiation of mouse preosteoblasts in vitro. METHODS: Mineralized collagen extracts were prepared from complete medium with magnesium ion concentration of 0, 5, 10, and 20 mmol/L. Mouse preosteoblasts were cultured with four mineralized collagen extracts, respectively, which were divided into mineralized collagen group, and 5, 10 and 20 mmol/L Mg2++mineralized collagen groups. The mouse preosteoblasts cultured in complete medium were used as control group. The cell morphology, proliferation, apoptosis, intracellular microfilament actin, and the activity of alkaline phosphatase and expression level of the osteogenic gene Runx2 after osteogenic differentiation were detected. RESULTS AND CONCLUSION: (1) After 24 hours of culture, the cells in the mineralized collagen group, and 5 and 10 mmol/L Mg2++ mineralized collagen groups adhered well, which showed no significant difference from the blank control group, and the elongated spindle cells with many synapses linked to the adjacent cells were observed. The cells in the 20 mmol/L Mg2++mineralized collagen group showed obvious pyknosis. (2) After 1, 3 and 5 days of culture, the cell viability in the 10 mmol/L Mg2++mineralized collagen group was significantly higher than that in the other four groups (P < 0.05). There was no significant difference among mineralized collagen, 5 mmol/L Mg2++ mineralized collagen and blank control groups (P > 0.05). The cell viability in the 20 mmol/L Mg2++mineralized collagen group was significantly lower than that in the mineralized collagen group (P < 0.05). (3) After 3 days of culture, DAPI staining showed that 20 mmol/L Mg2++mineralized collagen group had obvious nuclear disintegration, the other four groups had no obvious nuclear disintegration. (4) After 24 hours of culture, phalloidin staining showed that except the blank control and 20 mmol/L Mg2++mineralized collagen groups, the other three groups showed completely extended cell structure, and clear actin microfilaments, especially the 10 mmol/L Mg2++mineralized collagen group. (5) After 7 days of osteogenic differentiation, except for 20 mmol/L Mg2++mineralized collagen group, the activity of alkaline phosphatase and the expression level of Runx2 gene in the other three groups were significantly higher than those in the blank control group (P < 0.05), and those in the 10 mmol/L Mg2++mineralized collagen group was significantly higher than those in the 5 mmol/L Mg2++mineralized collagen and mineralized collagen groups (P < 0.05). (6) These results suggest that the combination of magnesium ion with mineralized collagen should be applied with appropriate concentration range of magnesium ion (≤ 10 mmol/L).
5.Sampling results and quality assessment of recombinant human interferon α1b injection

Objective To evaluate the quality status of recombinant human interferon α1b injection and find out some quality problems.Methods Totally 31 batches of recombinant human interferon α1b for injection and 11 batches of recombinant human interferon α1b injection from two enterprises were examined according to Chinese Pharmacopoeia Volume Ⅲ (2010),and the quality status of recombinant human interferon α1b injection was evaluated by statistical analysis of the results.Results All 42 batches of samples were qualified.The production process of each enterprise was steady.Conclusion At present the quality of recombinant human interferon αlb injection is generally good.The current standards are feasible,but the specified standard of osmolality needs to be improved.

Objective To evaluate the effect of rehabilitation training on rats with spinal cord injuries by Meta-analysis. Methods Ar-ticles were searched from PubMed( ~2013),CNKI(1989~2013), WanFang Data( ~2013),VIP(1989~2013),quality of included article was assessed with Jadad scale,and available data was analyzed with RevMan 5. 0 software. Results 287 related articles were identified,but only 11 eligible articles were included. The Meta-analysis of BBB score indicated that the rehabilitation training groups were better than con-trol groups. The BBB score[weighted mean difference(WMD) =1. 87,95%CI(1. 50,2. 33),Z=10. 02,P<0. 01]. There was significant diffence between two groups. Conclusion Rehabilitation training can improve the recover of hindlimb function.
7.Pregabalin attenuates docetaxel-induced neuropathy in rats.

Chemotherapy-induced neuropathy is a serious clinical problem for patients receiving cancer treatment. The aim of this study was to investigate the potential efficacy of pregabalin in chemotherapy-induced neuropathy in rats. A total of 35 male Sprague-Dawley rats were randomly divided into 5 groups: group 1, naive control; group 2, treated with pregabalin (30 mg/kg p.o., for 8 days); group 3, docetaxel was given by single intravenous infusion at 10 mg/kg; groups 4 and 5, pregabalin at 10 mg/kg and 30 mg/kg respectively was orally administered for 8 days after the docetaxel treatment. On day 8, behavioral test was performed, and substance P and CGRP release in dorsal root ganglion (DRG) and sciatic nerve were analyzed by electron microscope. Our results showed that docetaxel induced mechanical allodynia, mechanical hyperalgesia, heat hypoalgesia, cold allodynia, and sciatic nerve impairment and substance P and CGRP release in DRG. However, oral administration of pregabalin (10 mg/kg and 30 mg/kg) for 8 consecutive days significantly attenuated docetaxel-induced neuropathy by ameliorating heat hypoalgesia, cold allodynia, impairment of sciatic nerve and reducing the release of substance P and CGRP. The findings in the present study reveal that pregabalin may be a potential treatment agent against chemotherapy-induced neuropathy.
8.Diagnosis of infective endocarditis by Duke criteria and a new national criteria in a cohort of 205 patients

Objective To compare the value of the new national criteria (2 major or one major plus 3 minor criteria) with the Duke criteria for diagnosis of infective endocarditis (IE). Methods A total of 205 patients with clinical diagnosis of IE admitted at West China Hospital of Sichuan University were included in this study. Among them, IE was pathologically confirmed in 97 patients. The sensitivities of both criteria for the diagnosis of IE were compared. Results In 205 cases, the same microorganisms were detected twice in blood cultures in 13 cases (8.3%). Vegetations were detected by echocardiography in 183 patients (89.3%). In 97 cases with pathologically confirmed IE, the same microorganisms were detected twice in blood cultures in 6 cases (6.2%). Vegetations were detected by echocardiography in 89 patients (91.8%). IE diagnose was made in 44 (45.5%) and 86 (88.7%, P < 0.05 vs. Duke criteria) out of 97 pathologically confirmed IE patients by the Duke criteria and new national criteria, respectively. The specificities were 100% and 95.7% by Duke and new national criteria, respectively (P > 0.05). Conclusion With the addition of echocardiographic evidence of endocardial involvement and 2 minor criteria as definite diagnostic criteria, the sensitivity of the new national criteria is superior to that of the Duke criteria for diagnosing IE and the specificity for the diagnosis of IE between the two criteria is similar.
9.Surrogate end points for clinical trials on chronic kidney disease and research of Chinese medicine.

Chronic kidney disease is a kind of disease with the condition always worsening over time passing through a sequence of stages, and the evaluation on its clinical treatment is mainly by observing the speed of renal function deteriorating and the time of terminal renal failure occurrence. In order to conduct the trial go on wheels, the authors proposed that the "surrogate end points (SEP)" should be introduced. It is the biologic mark for substitute the clinical terminal point (event), formed depending upon the scientific evidences of epidemiology, pathophysiology, drug-therapy and other scientific evidence, which could be used for predicting the efficacy or damage of a certain measure, present or absent. This article aimed to explain the definition of SEP and to discuss the usable SEP for clinical trial on chronic kidney disease, such as proteinuria, declination of glomerular filtration rate and its slope coefficient as well as the time of terminal occurrence. Moreover, through analyzing the existent problems in clinical researches concerning TCM treatment of chronic kidney disease, the authors suggested that some improvements, chiefly the utilization of SEP for efficacy evaluation, are necessary in the clinical observation methodologies for chronic kidney disease.

10.Effect of carbenoxolone on expression of Fos, NMDAR2 and GFAP in the hippocampus of pentylenetetrazo-kindled epileptic rats.

OBJECTIVEGap junctions, the clusters of intercellular channels, play an important role in synchronizing electrical activity. This study investigated the effect of gap junction blocker carbenoxolone (CBX) on epileptic activity in pentylenetetrazo (PTZ)-kindled rats.
METHODSThirty adult male SD rats were randomly divided into three groups: control, PTZ-kindled and CBX-treated groups (n=10 each). The rats from the PTZ-kindled and the CBX-treated groups were intraperitoneally injected with PTZ (35 mg/kg x d) to induce epilepsy. After epilepsy kindling, they were intraperitoneally injected for 3 days with CBX (10 mg/kg) (CBX-treated group) or with normal saline (PTZ-kindled group). The control group received intraperitoneal injections of normal saline. Anti-GFAP, anti-Fos, and anti-NMDARZ immunohistochemical ABC methods were used to detect the expression of GFAP-Li, Fos-Li and NMDAR2-Li in the hippocampus respectively.
RESULTSSpontaneous seizures occurred in PTZ-kindled epileptic rats. CBX administration reduced spontaneous seizures. The NMDAR2-Li and Fos-Li neurons as well as GFAP-Li astrocytes in hippocampi increased in PTZ-kindled epileptic rats compared with controls. The numbers of Fos-Li (93.75 +/-7.94 vs 165.25 +/-15.87, P < 0.05) and NMDAR2-Li neurons (61.47 +/-3.62 vs 148.72 +/-14.53, P < 0.01) in the CBX-treated group were significantly less than in the PTZ-kindled group. There were no significant differences in the GFAP-Li expression between the CBX-treated and the PTZ-kindled groups.
CONCLUSIONSCBX may inhibit spontaneous seizures and decrease the numbers of Fos-Li and NMDARZ-Li neurons, thus providing anti-epileptic effects.
