1. Generation of brevican knockout mice

Objective To generate brevican knockout mutant mice. Methods The targeting construct to inactivate the brevican gene was made by using the ET cloning technology. The construct was linearized and electroporated into embryonic stem (ES) cells. Homologous recombinants were identifed by PCR after selecting the stable transfectants. The positive clones were then injected into C57BL/6 blastocysts, and the injected blastocysts were transferred into pseudopregnant foster mothers. Heterozygote and homozygote were multiplied by procreation mosaic. Progeny gene type was assayed by PCR. Results A neomycin resistance expression cassette under the control of the phosphoglycerate kinase (PGK) promoter was flanked by a 2.4-kb 5-arm fragment and a4.8-kb 3-arm fragment, thus introducing the NEO gene into exon 3 and deleting the encoding region from exon 3 to exon 8 after homologous recombination. Fourteen clones of gene-targeted ES cells were identified and three male chimeras with a higher than 50% chimeric ratio were produced. Six brevican-null mice were generated after outbred and inbred. Conclusion Genetically modified ES cells have been successfully generated by homologous recombination, and brevican-deficient mouse strain has been successfully generated.

3.The Clinical Application of MRCP in Diagnosing Cholangiocarcinoma

Objective To study the diagnostic value of MRCP in cholangiocarcinoma.Methods MRCP in 39 patients with cholangiocarcinoma comfirmed by surgery and pathology were performed on a GE 1.5T superconductive MR imaging unit before operations.Results MRCP imagings with diagnostic value were obtained in 39 cases,MRCP accurately showed the obstructive positions confirmed by surgery and pathology in all cases (100%).The qualitative rate of cholangiocarcinoma was 97%(38/39).Conclusion MRCP can not only accurately show the position of cholangiocarcinoma but also make accuracy diagnosis.
4.Patent applications in hospitals of Guangdong Province since 2000

The relation between patent applications and published papers in hospitals of Guangdong Province since 2000 on China Intellectual Property Network was analyzed , which showed that the quality and quantity of patents were lower in hospitals of Guangdong Province than in those of Shanghai and Beijing , the quality of patents was re-lated with the medical service level in hospitals, the patent applications were centralized in large class A hospitals of Guangzhou , and inventive patents were those of medical preparations .Suggestions were thus put forward for hos-pitals of Guangdong Province to improve their awareness of patent application, the quality of patents, transformation of patents, cooperation with patent service organizations, research of patent application, and balance of patent dis-tribution in different areas.
5.The correlation study of beta-ifbrinogen gene polymorphisms and plasma ifbrinogen concentration in patients with colorectal cancer

Background and purpose:Patients with colorectal cancer are often accompanied by the increase of plasma ifbrinogen concentration. This study aimed to investigate the distribution characteristics of beta-ifbrinogen gene-448G/A, -148C/T, -1420G/A and -854G/A polymorphism and plasma ifbrinogen (Fg) concentration in patients with colorectal cancer. Furthermore, we analyzed their effects on the occurrence and development of cancer.Methods:The level of plasma Fg was quantiifed by using Clauss clotting method.FGBβ gene polymorphisms were identiifed by re-al-time lfuorescence quantitative PCR (RTFQ-PCR) in 194 colorectal cancer patients and 74 healthy controls.Results:The plasma Fg levels in tumor metastasis group and non-metastasis group were signiifcantly higher than that in control group, respectively (P<0.05). Compared with control group, the frequencies of -148T allele and mutation genotype were notably higher in disease group (P<0.05). In all the groups, the plasma Fg levels of those with -148T allele were higher than those without -148T allele (P<0.05). In stageⅣ patients, there was no difference in PFS between -148T wild gen-otype group and mutation genotype group (P>0.05).Conclusion:Plasma Fg concentration in patients with colorectal cancer was signiifcantly raised, which suggests that Fg may play a role in the occurrence and development of colorectal cancer. The beta-ifbrinogen gene -148C/T polymorphism is one of the reasons that cause plasma Fg elevation, but has no correlation with prognosis of patients with stageⅣ colorectal cancer.

7.The rapid prediction of GVHD by three-dimensional structure differences of HLA

Objective Trying to predict the degree of GVHD after partly matched hematopoietic stem cell transplantation. Methods Analysis of the relationship between three-dimensional structure differences of donor-patient unmatched HLA and the GVHD levels after hematopoietic stem cell transplantation. Results GVHD levels were related to donor-patient unmatched HLA structure differences. The HLA structure differences forⅠ - Ⅱ degree GVHD were much smaller than that for Ⅲ - Ⅳ degree GVHD. Conclusion Prediction of GVHD by HLA structure differences is simple, rapid, specific and could help select proper conditioning regimens before transplantation and the proper immune suppressive agents after transplantation.
8.Clinical study of the effect of cardiac resynchronization therapy in patients of chronic heart failure

Objective To investigate the effect of cardiac resynchronization therapy on improving heart function and reversing left ventricular remodeling in chronic heart failure patients. Methods Eleven patients with NYHA class Ⅲ to Ⅳ heart failure, ejection fraction ≤35% and QRS duration ≥130 ms were implanted synchronous biventricular pacemaker. Serial assessment was performed before and up to 1-36 months after pacing. Results There were significant improvement of LVEF、 LVFS; and decrease in MR、 LVED and QRS duration. The NYHA class of heart function was improved from Ⅲ-Ⅳ to Ⅰ-Ⅱ. Six-minute hall walk distances were also improved. Conclusion For patients with NYHA class Ⅲ to Ⅳ heart failure and ventricular conduction delay, cardiac resynchronization therapy could reverse left ventricular remodeling and improve heart function.
9.Content Determination and Quality Evaluation of Gallic Acid in Gentiana Rigescens from Different Areas in Guizhou Province

Objective To determine the content of gallic acid and evaluate the quality of Galla chinensis of different habitats in Guizhou Province. Methods HPLC method was used to determine the content of Gallic acid on Alltima C18 column (4.6 mm?250 mm, 5 ?m) with the mobile phase of methanol-water-phosphoric acid (15∶85∶0.085). The flow rate was 1.0 mL/min and the detective wave length was 270 nm. Results The contents of Gallic acid in Galla chinensis of 10 different habitats in Guizhou Province were 51.43%~ 60.25%. Conclusion The quality of Galla Chinensis in different habitats in Guizhou Province are up to the Chinese Pharmacopoeia, and the individual varibility is significant.
10.THE APPLICATION OF MOLECULAR MODELING OF HLA ANTIGENS IN UNMATCHED STEM CELL TRANSPLANTATION FOR GVHD PREDICTION

To explore the pathogenic mechanism of GVHD, attempting to forecast the degree of GVHD after stem cell transplantation, and to enhance the therapeutic effectiveness of clinical transplantation. The conventional microlymphocytotoxicity and sequencing methods were used in typing the HLA. The degrees of GVHD were estimated by molecular modeling of HLA antigens and veryfied the estimation by comparing the clinical results with anticipated degrees. In 8 recipients, three were transplanted with half matched stem cells. Among these 3 pationts, two developed IV degree GVHD, and one developed II degree GVHD. In the other 3 patients, cells of two unmatched HLA antigens were transplanted, and among them one developed I degree GVHD, and two developed II degree GVHD. In two patients who were transplanted with cells of one unmatched HLA antigens, I and II degrees GVHD developed respectively. Second, the correlationship analysis showed that degrees of GVHD had positive correlation with the RMSD (relative mean square deviation) between different HLA antigens. These results indicated that the degrees of GVHD after stem cell transplantation were related with the difference of three dimensional structures of unmatched stem cell HLA antigens; molecular modeling might be used to predict the prognosis of clinical stem cell transplantation.
