1.Confirmatory testing of 120 positive HIV antibody screening samples in Lanxi City

Objective:
To analyze the confirmatory test results of 120 positive HIV antibody screening samples in Lanxi City, Zhejiang Province, so as to provide insights into AIDS detection and control. Methods Western blotting confirmatory assay was performed on 120 positive HIV antibody samples from HIV screening laboratories in Lanxi City from 2020 to 2022. The confirmatory test results were determined according to the “National Technical Guidelines for AIDS Testing (2020 revision)” and “Diagnosis for HIV/AIDS” (WS 293-2019), and the positive rate of confirmatory test and band distribution were analyzed.
Methods :
Western blotting confirmatory assay was performed on 120 positive HIV antibody samples from HIV screening laboratories in Lanxi City from 2020 to 2022. The confirmatory test results were determined according to the “National Technical Guidelines for AIDS Testing (2020 revision)” and “Diagnosis for HIV/AIDS” (WS 293-2019), and the positive rate of confirmatory test and band distribution were analyzed.
Results:
Among 120 positive HIV antibody screening samples, there were 100 HIV-1 antibody positive samples (83.33%) as revealed by Western blotting confirmatory assay, 6 HIV antibody negative samples (5.00%), and 14 HIV-1 indeterminate tests (11.67%). There were 72 men and 28 men tested positive for HIV-1 antibody, with a male to female ratio of 2.57∶1, and there were 88 patients at ages of 20 to 59 years (88.00%), with the highest frequency of gp160, gp120 and p24 bands (100.00%). The most common Western blotting band pattern was p24 among HIV-1 indeterminate tests (78.57%). Among all HIV-1 indeterminate Western blotting tests, 6 cases were tested positive for HIV-1 antibody (gp160+p24) and 8 cases tested negative for HIV antibody during the follow-up period.
Conclusions
The prevalence of HIV-1 antibody was 83.33% in 120 positive HIV antibody screening samples in Lanxi City. The three most common Western blotting band patterns were gp160, gp120 and p24. The follow-up requires to be reinforced in HIV-1 indeterminate Western blotting tests for early identification and early intervention.
2.Effect of rocuronium on spectral entropy during induction of general anesthesia in patients of Uygur nationality

Objective To investigate the effect of rocuronium on spectral entropy during induction of general anesthesia in patients of Uygur nstionality. Methods Forty ASA Ⅰ or Ⅱ patients (Uygur nationality) of both sexes, aged 20-50 yr, weighing 45-70 kg, undergoing elective surgery under general anesthesia, were divided into 2 groups ( n = 20 each): normal saline (NS) group and rocuronium group (group R). Anesthesia was induced with target-controlled infusion of propofol. The initial target plasma concentration wan net at 2 μg/ml. The concentration wan then increased by 0.5 μg/ml every 4 min until response entropy (RE) was decreased to 45 and maintained for 4 min. When the plasma concentration was equal to the effect-site concentration, iv rocuronium 0.6 mg/kg was injected in group R, while group NS received the equal volume of NS instead. Fentanyl 3 μg/kg was injected intravenously at 3 min after recuronium administration. The patients were tracheal intubated and mechanically ventilated. State entropy (SE) and RE were recorded immediately before induction (baseline, To), before rocuronium administration (T1), 2 main after rocuronium administration (T2) and at 0, 1, 2 and 3 min after intubation (T3-6). The difference between RE and SE wan calculated. Results The RE value at T3 and T4 and the difference between RE and SE at T2.5 were significantly lower in group R than in group NS ( P < 0.05). Conclusion Rocuronium can decrease the RE value and degree of increase in the difference between RE and SE during induction of general anesthesia in patients of Uygur nationality, which may affect the accuracy of spectral entropy in monitoring the depth of anesthesia.
3.Establishment of mice model of C6 glioma stem cells and its evaluation

Aim To establish ICR animal model with C6 glioma stem cells, to provide the ideal model for the further study of gli-oma stem cells in brain glioma model. Methods C6 glioma stem cell was cultured in vitro by suspension,and was identified with Nestin antibody. C6 stem cells of ICR mouse glioma model were used to investigate survival state and tumor volume in mice after the operation. HE staining and CD133 immunohistochemi-cal study were adopted to investigate the postoperative pathologi-cal changes in mice. Results The expression of Nestin was 96. 01% in C6 glioma stem cells, and Nestin was highly ex-pressed in the cultured C6 glioma stem cells. Mice were inocula-ted with tumor after loss of appetite, weight, behavior and slow, sluggish reaction. Tumor volume at day 21 after modeling was (9. 77 ± 6. 58) mm3 . After HE staining, the model showed the invasive growth, tumor cell shrinkage and derangement. Immu-nohistochemical CD133 staining revealed that tumor cytoplasm color was brown. Conclusion Glioma model can be established based on glioma stem cells into a high rate of tumor, the tumor cycle is short, which can be used as an ideal model for glioma.
4.Research of Fusion Expression of Human β-Defensin-3 in Escherichia coli

Objective To achieve the fusion expression of the entire human beta-defensin-3(hBD-3) gene. Method We synthesized two oligonucleotide primers accor ding to the codon preference of Escherichia coli. The gene was cloned into p GEX -4T-2 to establish the pGEX-4T-2-hBD-3 as the fusion expression vector by PCR. Transformed into E.coli strain DH5α, the express vector was induced an d ex pressed by IPTG. The fusion protein GST-hBD-3 was obtained by repeated cycles of freezing and thawing, cut by thrombin to attain the recombinant hBD-3 protei n. Result The result of the antibacterial peptide agarose diffu sion assay shows the antibacterial activity of the rhBD-3 against the S.aureu s exists, and it reaches 0.843U. Conclusion The fusion expr ession of the hBD-3 gene is successful.
5.Evaluation of the linearity characteristic of the cone-beam CT fixed on the Varian 23EX linear accelerator

ObjectiveTo investigate the CT number linearity of the cone-beam CT (CBCT) images at the different spatial locations in the scanning area.MethodsThe CatphanS04 phantom at the different locations are scanned repeatcdly using the CBCT on the Varian 23EX linear accelerator.The phantom is located the isocenter point,eccentric 3 cm,eccentric 6 cm,and different points on the z-axis successively on the accelerator.The scanned mode is the standard head mode.The reconstructive thickness is 2.5 cm.The different densities inserts of CTP404 module on the different locations are measured via Eclips treatment planning system (TPS) and computed by Matlab 7.0 and the CT linear fitting are then processed.In order to understand better the linear distribution along with the value of CT in the spatial distribution the results are compared with the fan-beam CT.ResultsPhantom studies show that:CBCT has good linearity performance not only under the standard header (body) of the scanning conditions,but also on such locations including the cross-sectional,the sagittal,the coronal plane and the eccentric position ( R2 > 0.953 ).Bowtie filtration device does not change the CT finearity but changes the value of CT.ConclusionsThe linearity of X-ray CBCT on the Varian linear accelerator is favorable.CBCT will be used in the TPS dose calculation via further correction of the CT value.
6.Clinical analysis of different canalith repositioning maneuver in treatment of apogeotropic nystagmus of horizontal semicircular canal benign paroxysmal positional vertigo.

OBJECTIVE:
To analyze the clinical effect of different canalith repositioning maneuver in the treatment of the apogeotropic nystagmus of horizontal semicircular canal benign paroxysmal positional vertigo (apogeotropic HSC-BPPV).
METHOD:
Forty patients of apogeotropic HSC-BPPV colleted from March 2014 to May 2015 were divided into two groups, each group had 20 cases. The control group was repositioned with the Gufoni maneuver before the Barbecue maneuver, the research group with the improved Nuti maneuver.
RESULT:
The success rate of the research group was not lower than that of the control group, which hadn't statistical (P > 0.05). And after the Barbecue maneuver, all patients have been recovered.
CONCLUSION
Comparing with the Gufoni maneuver, the improved Nuti maneuver designed by us, had the same success rate to treat apogeotropic HSC-BPPV, and is easier to learn,with more compliance,more comfort and more repeatability.

7.Relationship study of blood serum visfatin and iron metabolism in newly diagnosed type 2 diabetes mellitus

Objective To determine the concentration of blood serum visfatin and some iron metabolism‐related indicators:Ser‐um ferritin、hepcidin、serum transferring receptor(sTfR)level in newly diagnosed type 2 diabetes patients and explore the inter‐rela‐tionship between blood serum visfatin and iron metabolism .Methods Eighty‐five patients with newly diagnosed type 2 diabetes mellitus were selected and divided into 2 groups including 43 patients with normal weight (the normal weight of diabetic group , group B)and 42 obese patients (the obese diabetic group ,group D) .Meanwhile ,86 healthy examinees were selected and divided into 2 groups including 43 cases with normal weight (the control group ,group A)and 43 cases with obese (simple obesity group ,group C) .Serum visfatin ,ferritin ,hepcidin ,serum transferring receptor level and other clinical parameters of all groups were determined . Results Blood serum visfatin concentration was not found to be associated with ferritin ,hepcidin ,serum transferring receptor in all the groups(r=0 .111 ,P>0 .05) .Conclusion Blood serum visfatin may be not associated with ferritin ,hepcidin ,serum transferring receptor in newly diagnosed type 2 diabetes patients .
8.Respiratory Failure with Lung Infection in EICU:An Analysis of Etiological Distribution and Drug Resistance

OBJECTIVE To analyze the etiological distribution and drug resistance characteristics of patients with respiratory failure and lung infection in EICU.METHODS The types of respiratory failure,the incidence condition of ventilator-associated pneumonia(VAP),the mortality rate,and the drug sensitivity of 129 pathogen strains which were isolated and cultured from the 110 cases with respiratory failure and pneumonia between Jan 2006 to Aug 2008 in EICU were analyzed.RESULTS Among the 110 patients,48 cases were died(the mortality rate of 43.6%).There were 28 cases(58.3%) of Ⅱ type respiratory failure,20 cases(41.7%) of Ⅰ type respiratory failure.Of 129 pathogens strains,there were 29 Gram-positive bacteria(22.5%) and 100 Gram-negative bacteria(77.5%).The major Gram-negative bacteria were Pseudomonas aeruginosa,Acinetobacter baumannii,and Klebsiella pneumoniae,and they showed generally higher multidrug resistance.Except minocycline,A.baumannii appeared strong multidrug resistance to the other 16 kinds of common antibiotics.The main Gram-positive bacteria were Staphyococcus haemolyticus,S.aureus and coagulase-negative staphylococci(CNS),and no evidence showed they resisted to vancomycin.CONCLUSIONS There are high mortality rate of patients with respiratory failure and lung infection if they suffered from compound infection.The main pathogens of the lung infection are Gram-negative bactiera,such as P.aeruginosa,A.baumanniiand K.pneumoniae in order,with high multidrug resistance.The main Gram-positive bacteria show no evidence resist to vancomycin.
9.Clinical research of music in relieving orthodontic pain.

OBJECTIVETo discuss the effect of music in relieving pain during orthodontic treatment.
METHODSOne hundred and sixty-five cases who wore arches the first time were selected and treated. These patients were randomly divided into two groups: Music group and blank group. The music intervention was used in reducing orthodontic treatment pain, while blank group experienced no intervention measures. Visual analogue scales (VAS) were adopted to record patients' perception of pain, and Eysenck personality questionnaire (EPQ) to evaluate personality traits of all samples.
RESULTSIn 165 patients, 85.45% were effective. The music group showed significantly less pain than the blank group (P < 0.05). In music and blank group, the pain was stronger in the patients with a tendency of introversion than those with a tendency of extroversion, as well as the irritability ones than steady-minded (P < 0.05). In music group, the pain was stronger in the females than the males (P < 0.05).
CONCLUSIONMusic helps to relieve pain during orthodontics treatment. The effect of music to relieve the pain during orthodontics treatment for the males are better than that for the females, extroverted personality ones are superior to introversive personality ones, and the steady-minded patients are better than irritability ones.
