1.Clinical observation of lafutidine treated in curing peptic ulcer

Objective To observe the clinical efficacy and safety of application of lafutidine treated in curing peptic ulcer.Methods Eighty-four patients with peptic ulcer were randomly assigned to receive either lafutidine(10 mg) twice daily in 42 patients of cured group or lansoprazole (30 mg) once daily for 4 weeks in 42 patients of control group.Ulcer healing rate and clinical performance were examined after treatment.Results The first,third and seventh day of peptic ulcer healing rate were 38.09% (16/42), 71.42% (30/42) and 90.47% (38/42) in the lansoprazole group and 23.81% (10/42), 76.19% (32/42) and 85.71% (36/42) in the lafutidine group.There were no siginificant difference between two groups (P =0.157,0.620, 0.736).Gastroscope examination showed the complete healing rate, average healing rate and general clinical effectiveness were 61.90%(26/42) ,30.95%(13/42) and 97.61%(41/42) in the lansoprazole group,while the number for lafutidine group were 57.14% (24/42), 28.57% (12/42) and 95.23% (40/42).There were no siginificant difference between two groups (P =0.657, 0.811, 1.000).No severe side-effect was occurred in both groups.Conclusion Use Lafutidine to cure peptic ulcer can promote healing effectively, improve patients' clinical symptoms which is similar to lansoprazole.
2.The influence of short-term intensive insulin therapy on ?-cell function in newly diagnosed type 2 diabetes

Objective To observe the influence of short-term continuous subcutaneous insulin infusion(CSII)on ?-cell function in newly diagnosed type 2 diabetes(T2DM).Methods A total of 32 inpatients with newly diagnosed T2DM who had a 14-day course of CSII were randomly assigned to two groups of OGTT and arginine stimulating test(AST).Before and after 3 days and 14 days CSII treatments,OGTT and AST were performed respectively.The changes of the values of plasma glucose,HOMA-IR,HOMA-? and △C30/△G30,△C120/△G120 or arginine-induced acute C-peptide reaction(ACRARG)in two groups were compared.Results In OGTT,the values of plasma glucose and HOMA-IR were decreased,HOMA-? and △C120/△G120 were increased significantly after CSII therapy,both in day 3 and day 14;and △C30/△G30 were unchanged after 3 days but increased markedly after 14 days.However,in ACRARG,the above mentioned parameters almost remained unchanged after CSII treatment in 3 days or 14 days.Conclusions In newly diagnosed T2DM with elevated fasting glucose levels,intensive insulin therapy can partly improve ?-cell function showed by OGTT,not by AST.
3.MRI features and pathological grading of hepatic neuroendocrine neoplasm

Objective To compare the MRI features of hepatic neuroendocrine tumors (NET G1,G2) and neuroendocrine carcinomas (NEC G3),as well as to improve the accuracy in hierarchical diagnosis.Methods Twenty patients with histopathologically proven NET and nineteen patients with histopathologically proven NEC were retrospectively analyzed.The morphological and MR signal features were compared.Results The morphological features of vascular invasion (P < 0.05) and lymphadenectasis or necrosis (P < 0.05),as well as the MR signal features on portal phase (P < 0.05) and delayed phase (P <0.05) were different between the NET group and the NEC group;contrast to noise ratios (CNR) were also different between the two groups (x2 =5.14,P < 0.05),CNR of the NEC group on both arterial phase (Z =121.75,P < 0.05) and portal phase (Z =139.31,P < 0.05) were significantly lower than the NET group;ROC analysis of CNR demonstrated an area under the curve of 0.729 (P < 0.05) on portal phase,when the optimal cut-off value of-61.38 was used,a sensitivity of 90.0% and a specificity of 63.2% can be achieved.Conclusions MRI plays an important role in the hierarchical diagnosis of hepatic neuroendocrine neoplasms.The signs of vascular invasion,lymphadenectasis or necrosis as well as the MR signal features during dynamic enhanced scanning are of great value in differentiating NETs from NECs.
4.Establishment of cell line stably expressing INSIG2 and its effects on fat metabolism

Objective To establish the cell line stably expressing INSIG2 and observe its effecet on fat metabolism after overexpression of INSIG2.Methods The eukaryotic expression plasmid pcDNA3.1(+)-INSIG2 was constructed,which was transfected into 3T3-L1 cells.The expression of INSIG2 and related genes were detected by RT-PCR and immunohistochemistry,the contents of FFA in cell culture medium and adipocyte differentiation were detected by ELISA and Oil Red "O"staining respectively.Results After pcDNA3.1(+)-INSIG2 was transfected into the 3T3-L1 cells,the expression of INSIG1 mRNA and FAS mRNA were down-regulated,the content of FFA in the cell culture medium was decreased and adipocyte differentiation was drepressed.Conclusion The cell line stably expressing INSIG2 was successfully established,the transfected INSIG2 may have a drepressant effect on fat metabolism.

Post-stroke cognitive impairment (PSCI) directly affects the outcome of patients with stroke. Enlarged perivascular spaces (EPVS) suggest the impairment of brain clearance mechanism and may affect cognitive function. More and more studies have confirmed that the presence of EPVS will aggravate PSCI. This article reviews the relationship between EPVS and PSCI.
7.The role of the health education model in pain self-efficacy for patients with cancer pain

Objective To observe the role of the health education model in pain self- efficacy for patients with cancer pain. Methods Sixty- four patients suffering from cancer pain were enrolled into the study and self- contrast experiment was made on each patient. The health education model was set up and health education for each patient with cancer pain was implemented. The pain, self- efficacy, cancer pain knowledge before and after the intervention were observed by Numerical Rating Scale (NRS),Chronic Pain Self- efficacy Scale(CPSS) and Cancer Pain Knowledge Questionnaire. Results NRS score were (5.38 ±0.19) points and (1.05 ± 0.11) points before and after the intervention, and there was significant difference (t =25.288, P = 0.000). Before intervention, pain management self- efficacy, physical function self- efficacy, symptom coping self- efficacy of CPSS scores were (10.38 ±0.37) , (20.97±0.81) , (16.86 ± 0.49) points, while after the intervention, the scores were (19.31± 0.30) , (33.25 ± 0.60) , (29.75 ±0.51) points, there were significant differences ( t = -33.225, -18.236, -18.235, all P = 0.000). Before and after the intervention, the answer rate of Cancer Pain Knowledge Questionnaire was 50.00%(32/64) and 87.50%(56/64), there was significant difference( χ2=20.51, P < 0.01). Conclusions To set up the health education model and implement health education for each patient with cancer pain can improve the patient′s pain management and enhance self-efficacy.
8.Protective Effect of Compatibilities of Ginsenosides Rg1 and Aconitine on Myocardial Cells of in vitro Cul-tured Heart Failure Model

OBJECTIVE:To investigate the protective effect of the compatibilities of ginsenosides Rg1 and aconitine on myocar-dial cell of in vitro cultured heart failure model. METHODS:The myocardial cells of neonate rat were grouped into normal control group,model group,positive control group(Deslanoside injection,1×10-7 mol/L),ginsenosides Rg1 group(1×10-8 mol/L),acon-itine group (1 × 10-9 mol/L) or their compatibilities groups (1∶1,2∶1,1∶2,V/V). Except for normal control group,other groups were given 0.8%pentobarbital sodium to induce heart failure model of myocardial cells. After modeling,each group was given rele-vant medicine for 1 h,and then the activities of T-ATPase,Ca2+-Mg2+-ATPase,Na+-K+-ATPase in cells were all detected. The activi-ties of acyl carrier protein(ACP)and lactate dehydrogenase(LDH),and the contents of brain natriuretic party(BNP),TNF-α and total glycogen were measured in cell culture fluid. RESULTS:Compared with normal control group, T-ATPase and Ca2+-Mg2+-ATPase activities were decreased significantly in model group;meanwhile,Na+-K+-ATPase activity was increased signifi-cantly,and ACP,LDH activities and BNP content in cell culture fluid were increased significantly(P<0.05). Compared with mod-el group,the activities of T-ATPase in treatment groups were increased significantly,while the activity of LDH in cell culture fluid was decreased significantly;when the volume ratio of ginsenoside Rg1 to aconitine was 2∶1,protective effect was the strongest;the activities of Na+-K+-ATPase in aconitine group and compatibility groups were all decreased significantly,with statistical signifi-cance(P<0.05). The activities of ACP and BNP in cell culture fluid were all decreased in treatment groups,but the content of to-tal glycogen in cells and the TNF-α content of in cell culture fluid had no change (P>0.05). CONCLUSIONS:Compatibility of ginsenosides Rg1 and aconitine can improve ATPase activities and membranous permeability,regulate BNP secretion and protect myocardial cell of heart failure model,especially the compatibility of ginsenosides Rg1 to aconitine of 2∶1 ratio.
9.Breeding of Zmu-1:DHP inbred strain guinea pig and preliminary analysis of molecular genetic structure of the strain

Objective To breed a guinea pig inbred strain and set up a method for detection of the microsatellite markers of genetic structure in guinea pigs. Method Using inbreeding methods we try to breed the Zmu-1:DHP inbred strain. With 15 pairs of polymorphism microsatellite primers, the genetic homozygosity of Zmu?1:DHP inbred strain,Zmu?1:DHP outbred strain and Zmu?2:DHP inbred strain ( as control) were examined by PCR. Results After breeding for 13 years, 8 sublines of Zmu?1:DHP inbred strain ( >20 generations) were bred. After identification, the gene frequency of the second subline of Zmu?1:DHP inbred strain was 86. 7%,higher than Zmu?1:DHP outbred strain (6. 7%) and Zmu?2:DHP inbred strain (66. 7%). The average number of loci of Zmu?1:DHP inbred strain was 1. 13,lower than that of Zmu?1:DHP outbred strain (2. 47%) and Zmu?2:DHP inbred strain (1. 33%). The genotypic frequency of Zmu?1:DHP in?bred strain was also higher than that of the other strains. The gene types of Zmu?1:DHP inbred strain were included in the genes of Zmu?1:DHP outbred strain, but Zmu?1:DHP inbred strain was short of 2 characteristic genes. The gene homozy?gous rates of 8 sublines of Zmu?1:DHP inbred strain were different with each other,among them, those of the 2nd and 8th sublines were higher than others. Conclusions There are both homozygosity and specificity in the Zum?1:DHP inbred strain and Zum?1:DHP outbred strain. The second Zum?1:DHP subline becomes a new inbred strain guinea pig. It is es?sential that the subline with the characteristic property is screened from these sublines. The guinea pigs of black Zmu?2:DHP inbred strain carrying microsatellite markers not present in the white strains, may carry optimal genes related with hair color properties.
10.Galectin-3 expression in normal and LPS-stimulated genital system of female mouse

AIM:To study the expressions of galectin-3 and the possible anti-infectious function in genital tract of female mouse. METHODS:The expressions of galectin-3 mRNA in different reproductive tissues were analyzed by RT-PCR. The mouse model of genital tract infection was constructed by injection of LPS and Freunds adjuvant incomplete into the vaginal mucous. The expression pattern of galectin-3 protein was detected by immunohistochemistry. RESULTS:The expression of galectin-3 mRNA was detected in the uterus,vagina,oviduct and ovary of normal adult mouse. The galectin-3 protein was expressed in the genital tract mucous of epithelial cell surface and uterine gland.
