2.Clinical effect of phacoemulsification combined with intraocular lens implantation in the treatment of senile cataract

Objective The clinical effect of phacoemulsification and intraocular lens implantation in the treatment of senile cataract.Methods120 cases of senile cataract patients in September 2014 ~2016 year in December in our hospital, were randomly divided into two groups: the observation group after phacoemulsification combined with intraocular lens implantation, the control group was treated with small incision non phacoemulsification extracapsular cataract extraction.In the comparison of two groups.ResultsAfter treatment, the observation group was 0.5~0.9 the proportion of 51.7%, visual acuity>1 the proportion of 6.7% were significantly higher than control group (P<0.05);the complication rate of observation group was 43.3% lower than the control group 61.7%, the difference was statistically significant (P<0.05).ConclusionPhacoemulsification combined with intraocular lens implantation in senile cataract patients can effectively improve patients' visual acuity, control the occurrence of complications, and the prognosis is good.
3.Activity and mRNA expression of cytochrome P450 and fentanyl metabolism in rat intestine in anhepatic phase

Objective To investigate the variation of fentanyl concentration and the gene expression and activity of rat intestinal cytochrome P450 3A1 in anhepatic phase. Methods The experiment was comprised of 2 steps. Step 1: The rats were randomly divided into experimental group (group A2, underwent occlusion of the hepatic portal) and control group (group A1), with 10 rats in each group. Fentanyl blood concentration was analyzed by LC/MS/MS. Step 2: The rats were randomly divided into group B1 (control), group B2 and group B3 (the rats underwent devascularization of the hepatic portal for 30 or 60 min). 4.Comparative proteomics analysis of aging rat aorta during the process of increasing age

Objective To study the proteins related to aging in aortic of old rats for laying the foundation of further study of aging mechanism.Methods The rat model of aging was built,and all model rats were divided into 4 groups:the adult group(9 weeks),the old group(12 months)of WistarKyoto (WKY) rats,and the age-matched spontaneously hypertensive rats (SHR).Blood pressure of 4 groups was observed.Morphological change of aorta was observed by HE staining.Differential proteins were identified by isobaric tags for relative and absolute quantitation,(iTRAQ)-coupled liquid chromatography and tandem mass spectrometry technology.Part of differential proteins was subsequently detected by real time PCR and Western blot.Results The mean SBP of the old group SHR was higher than WKY of 97.1% (t=39.00,P<0.05),and the adult group of SHR was also higher than WKY of 5.4%(t=3.64,P<0.05).Compared with the adult group,aging change in the aortic morphology of old SHR and WKY were shown in HE staining,and the change in SHR rats was more marked.7 proteins related to aging were identified by Mass spectrum analysis,and they were Profilin-1,Prelamin A,HSP70,creatine kinase-M,Fibulin-5,eIF5A and Prohibitin.Part of differential proteins was subsequently confirmed by real time PCR and Western blot.Prelamin-A was up-regulated in the old group of WKY and SHR (0.15±0.01 vs.0.45±0.04,0.34±0.02 vs.0.78±0.06) (t=12.67,12.06,all P<0.01),Prohibitin was down-regulated in the old group of WKY and SHR(1.34±0.05 vs.1.01± 0.06,1.24±0.05 vs.0.88±0.08) (t=7.41,7.09,all P<0.01).Profilin-1 was up-regulated in the old group of WKY and SHR (9.12±0.4 vs.20.76±0.8,16.84±0.5 vs.55.16±0.9) (t=22.55,64.46,both P<0.01),and Profilin-1 expression in the old group of SHR was higher thanWKY (55.16±0.90 vs.20.76±0.8,t=49.49,P<0.01).Conclusions Differential proteins of the old rat aorta are identified through the comparative proteomics method.These differential proteins will provide new targets for the prevention and control of vascular aging.
5.Clinical application of in vitro maturation of human immature oocytes for infertile women with polycystic ovary syndrome

Objective To investigate clinical effect and safety of in vitro maturation(IVM)of human immature oocytes in infertile women with polycystic ovary syndrome by comparing with conventional in vitro fertilization(IVF)and intracytoplasmic sperm injection(ICSI).Methods From Jan.2003 to Dec.2009,157 infertile women with PCOS underwent 162 cycles IVM in Center for Reproductive Medicine,the First Affiliated Hospital of Anhui Medical University.In the mean time,109 patients with PCOS underwent 114 IVF/ICSI cycles as control group 1 and 106 patients with other factors underwent 106 IVF/ICSI cycles as control group 2.Treatment and outcome of pregnancy and infant were compared among those 3 groups.Results No statistically significant difference were found in terms of the positive rate of hCG in urine[35.7%(56/157),42.2%(46/109),44.3%(47/106)],the rate of clinical pregnancy[29.3% (46/157),37.6%(41/109),41.5%(44/106)],the rate of entopic pregnancy[1.9%(3/157),1.8% (2/109),0.9%(1/106)],the rate of miscarriage[18.6%(8/43),12.8%(5/39),20.9%(9/43)]and the rate of live-birth[22.3%(35/157),31.2%(34/109),32.1%(34/106)]among three grbups(IVM group,control group 1,control group 2,P > 0.05).The rate of preterm labor,low weight newborn,mean birth weight,ratio of male to female did not show significantly difference among 3 groups(P > 0.05).The average control ovarian stimulation was 6 days,the median dose of gonadotropin(Gn)was 675 IU,and the total hospital cost was(8392 ± 1328)RMB in IVM group,which were statistically lower than those in the other two control groups(P < 0.01).The rate of multiple pregnancy was 4.7%(2/43)and ovarian hyperstimulation syndrome(OHSS)0 in IVM group,which were significantly lower than those in the other control group(P <0.01).Conclusion In vitro maturation is an effective treatment in infertile women with PCOS,it could obtain the similar pregnancy outcome and reduce total cost,the dosage of gonadotropinreleasing hormone and rate of OHSS compared with conventional IVF/ICSI.

7.Prevention and treatment of different artificial tears in dry eye after pha-coemulsification

Objective To investigate the clinical effect and prevention of different artificial tears in dry eye after cataract phacoemulsification combined with intraocular lens implantation. Methods Totally 57 patients (57 eyes) with dry eye after phacoemulsification combined with intraocular lens implantation who were diagnosed with cataract in the Department of Ophthalmology in our hospital from October 2016 to October 2017 were randomly divided into 3 groups including sodium hyaluronate group, polyethylene glycol group and control group. All patients underwent phacoemulsi-fication combined with intraocular lens implantation. The control group was given conventional treatment after surgery. The other two groups were given sodium hyaluronate eye drops and polyethylene glycol eye drops on the basis of conventional treatment respectively. The dry eye symptom score, tear film rupture time, Schirmer I test, corneal fluorescein staining score before surgery, at 1 week, 1 month and 3 months after surgery were observed. The therapeutic effect was evaluated. Results In the three groups, the dry eye symptom score and corneal fluorescein staining score after surgery was lower than that before surgery, and the tear film rupture time and Schirmer I test after surgery increased compared with that before surgery. There was significant difference in the recovery of dry eye between the two groups and the control group, of which the sodium hyaluronate group recovered most significantly. Conclusion Cataract pha-coemulsification has an effect on the stability of tear film, which leads to postoperative dry eye. Artificial tear can be used for prevention and treatment. Sodium hyaluronate eye drops are more effective and worthy of clinical application.
8.Analysis of HLA-DQB1 polymorphism for patients with allergic rhinitis of Uygur and Han people in Xinjiang.

OBJECTIVE:
To investigate the association of HLA-DQB1 alleles and artemisia pollen allergic patients of Han and Uygur people in the Xinjiang area.
METHOD:
PCR-SSP (sequence-specific primer polymerase chain reaction) were used to test fifty Uygur and fifty Han people with allergic rhinitis with hypersensitivity to artemisia pollen in the Xinjiang area. Fifty normal Uygur and fifty normal Han people in the Xinjiang region were enrolled in the control group.
RESULT:
The frequency of HLA-DQB1 * 05 in the Uygur group with allergic rhinitis was significantly higher (19.4%) than that in the Uygur control group (7%) (P < 0.05). There were no significant difference between in the allergic rhinitis group and in the control group.
CONCLUSION
HLA-DQB1 * 05 might be the susceptibility genes of the Uygur patients with allergic rhinitis in the Xinjiang area.

9.Cutaneous and subcutaneous coinfection by Lichtheimia corymbifera and Candida parapsilosis: a case report

To report a case of cutaneous and subcutaneous coinfection caused by Lichtheimia corymbifera and Candida parapsilosis.A 67-year-old female peasant consulted about proliferative granuloma developing on her left forearm after topical application of a Chinese herbal drug and splint fixation for the treatment of suspected fracture of the wrist.Direct microscopic examination showed gram positive budding yeast cells in lesion secretions.Pathological study with periodic acid-Schiff (PAS) and gormori methenamine silver (GMS) staining revealed broad non-separate hyphae in the corneum and dermis.Fungal culture of lesional tissue at 35℃ grew both mould and yeast.The mould was identified as Lichtheimia corymbifera based on morphological findings and sequences of the internal transcribed space (ITS) 1-4 regions.Thermal tolerance study revealed that the isolate grew fast at 37℃ but slowly at 40℃.Under a scanning electron microscope,the acrogenous sporangia were pear-shaped with conical sporangiophores originating from the top of stolon,which were among but not opposite to the rhizoids.The yeast was identified as Candida parapsilosis by Chromagar test and D1/D2 region sequencing.As antimicrobial susceptibility test indicated,the Lichtheimia corymbifera isolate was most sensitive to terbinafine and itraconazole.The proteolytic activity of Lichtheimia corymbifera was higher than that of Candida parapsilosis.The granuloma completely subsided after surgical resection and 6-week treatment with oral itraconazole 200 mg twice a day.No recurrence was observed during a 4-year follow-up.
10.Effect of penehyclidine hydrochloride on hyperoxia-induced acute lung injury and the relationship with endoplasmic reticulum stress-dependent apoptosis in infantile rats

Objective To evaluate the effect of penehyclidine hydrochloride on hyperoxia-induced acute lung injury and the relationship with endoplasmic reticulum stress-dependent apoptosis in infantile rats.Methods Ninety-six clean-grade male infantile Sprague-Dawley rats,weighing 40-50 g,aged 14 days,were allocated into 4 groups (n =24 each) using a random number table method:control group (C group),penehyclidine hydrochloride group (P group),hyperoxia group (HO group) and hyperoxia plus penehyclidine hydrochloride group (HP group).Infantile rats were intravenously injected with penehyclidine hydrochloride (0.3 mg/kg) at the same time point every day for 3 consecutive days in P and HP groups.Infantile rats were injected with the equal volume of normal saline instead of penehyclidine hydrochloride in C and H groups.Acute lung injury was induced by inhaling oxygen at concentration greater than 90% for 72 h starting from 4th day after administration.Infantile rats were sacrificed at the end of inhaling,and lungs were removed for examination of the pathological changes and for determination of weight to dry weight ratio (W/D ratio),index of quantitative evaluation for alveolar damage (IQA),pneumonocyte apoptosis (using TUNEL),expression of glucose-regulated protein 78 (GRP78) and CCAAT/enhancer-binding protein homologous protein (CHOP) protein and mRNA in lung tissues (by Western blot or using reverse transcription-polymerase chain reaction).The mitochondrial injury score was assessed,and apoptotic index (AI) was determined.Results Compared with C group,the W/D ratio,IQA,AI and mitochondrial injury score were significantly increased,and the expression of GRP78 and CHOP protein and mRNA was up-regulated in HO and HP groups (P<0.05),and no significant change was found in the parameters mentioned above in group P (P>0.05).Compared with HO group,the W/D ratio,IQA,AI and mitochondrial injury score were significantly decreased,the expression of GRP78 and CHOP protein and mRNA was downregulated (P < 0.05),and the pathological changes of lung tissues were significantly attenuated in HP group.Conclusion Penehyclidine hydrochloride can mitigate hyperoxia-induced acute lung injury,and the mechanism may be related to inhibiting endoplasmic reticulum stress-dependent apoptosis in infantile rats.
