1.Effects of Ganzhikang Capsule on Ultrastructure Changes in Non-Alcoholic Fatty Liver Rats

Objective: To observe effects of Ganzhikang Capsule on ultrastructures in non-alcoholic fatty liver model rats so as to provide morphological basis for clinical treatment. Methods: Fatty liver model rats were induced by high fat diet, and the effects of Ganzhikang Capsule on microstructures in liver tissue were investigated with Dong Bao Gan Tai used as control medicine. Results: In the model group, serious fatty degeneration of hepatocytes with inflammatory infiltration, degeneration and necrosis, and deforming, swelling of mitochondrion, dilatation of endoplasmic reticulum were found. After treatment of Ganzhikang Capsule, microstructures of hepatocytes restored obviously, fatty degeneration relieved significantly, and Ganzhikang Capsule was superior to Dong Bao Gan Tai in restoration of fatty degeneration (P
2.Curative Effect of Different Biofeedback and Electrical Stimulation Solutions on Female Stress Urinary Incontinence

Objective To investigate the curative effect of different biofeedback and electrical stimulation solutions on female stress urinary inconti-nence(SUI). Methods A total of 40 female patients with mild-to-moderate SUI were randomly divided into two groups(n=20 each group)and treated by the biofeedback and electrical stimulation. In the treatment group,the current increased in the middle of the treatment on the basis of the initial threshold of stimulus intensity to make a new tolerable threshold of stimulus intensity. In the control group,the patients were treated with the initial threshold of stimulus intensity during the whole treatment. The curative effects in these two groups were determined by the comprehensive strength of pelvic floor,the 1 h urine pad test results,and the life quality scores of urinary incontinence. Results After treatment,there were signifi-cant improvements in all the above indicators in two groups(P<0.05),and the results is better in the treatment group than in the control group. The effective rate in treatment group(95%,19/20)was higher than that in the control group(65%,13/20)(P<0.05). Conclusion The biofeedback and electrical stimulation were effective and safe in the treatment of women with SUI. The curative effect of patients treated with increasing current in the middle of the treatment is better,suggesting that it can be widely applied in clinic.
3.Diagnosis and differential diagnosis for acute appendicitis in children

Acute appendicitis is the most common surgical or acute abdomen in childhood.Rapid and correct diagnosis is the premise of rational therapy,but there are still some problems in diagnosis and differential diagnosis.Sufficient comprehension about clinical features and differential diagnostic diseases,typical clinical symptoms,physical examination,laboratory examinations,ultrasonography or CT would make a definite diagnosis.Laparoscopy is also used to make a diagnosis if necessary.
4. Interferon-gamma and systemic lupus erythematosus

The etiology and pathogenesis of systemic lupus erythematosus(SLE) remain unclear. Interferon-γ (IFN-γ), a Th1-type cytokine, may be involved in the pathogenesis and inflammatory process of SLE. This article reviews reports on the serum IFN-γ of SLE patients and mouse SLE models, in vitro secreted IFN-γ, expression of IFN-γ induced protein and IFN-γ related genes, and treatment with traditional Chinese medicines.
5.Azathioprine Treatment in Systemic Lupus Erythematosus: A Double Edged Sword

Extremely severe pancytopenia induced by low dosage of azathioprine in systemic lupus erythematosus patients
is rare. A 40-year-old Chinese female was diagnosed with systemic lupus erythematosus. She suffered worse
erythema, oral ulceration, raised erythrocyte sedimentation rate and high anti-dsDNA in August 2013. Then she
was initiated on oral azathioprine 50mg/d and extremely severe pancytopenia was seen in September 2013. She
was recovered by a series of treatments. Regular monitoring of blood counts is highly recommended to reduce
the possible serious myelosuppression induced by azathioprine.
6.Preparation and identification of monoclonal antibodies against pgp3 of Chlamydia trachomatis

Objective:To express secreted protein-pgp3 of Chlamydia trachomatis(Ct)plasmid,produce monoclonal antibodies (mAbs)and identify their basic biological characteristics.Methods: Construction pGEX-6p2-pgp3 prokaryotic expression vector,then expressed GST-pgp3 fusion protein in E.coli as antigen used to immune BALB/c mice, spleen cells were fused with SP2/0 mouse myeloma cells.The hybridoma cell lines of screening mAbs were secreted by ELISA,and mAbs specificity,type,class and titer were identified.Results:GST-pgp3 fusion protein was successful expressed,5 strains stable hybridoma cell lines that secreted mAbs were screened out,including 4 strains secreted anti-pgp3 mAbs(P1B3,P2A1,P2B6,P2C2),mAbs type were IgG1/κ,the other strain secretion anti-GST mAbs(P3B4),mAbs type was IgG2b/κ.The titer of P1B3,P2A1,P2B6,P2C2,P3B4 were 1∶6 400,1∶3 200,1∶12 800,1∶6 400 and 1∶6 400 respectively.Conclusion:Successful prepared anti-pgp3 mAbs,and lay a foundation for further study the function of Ct plasmid protein pgp3 and the establishment of Ct detection method.
7.Mining analysis on composition and medication of menstruation prescriptions in Fu Qingzhu's Obstetrics and Gynecology.

In this paper, menstruation prescriptions were selected from "Fu Qingzhu's Obstetrics and Gynecology" and analyzed by using GRI algorithm, correlation analysis, hierarchical clustering method through SPSS, Clementine and traditional Chinese medicine (TCM) inheritance auxiliary systems, in order to screen out 15 menopathy prescriptions, which involve 45 traditional Chinese medicine herbs. In the study, blood-tonifying and qi-tonifying herbs were found to be frequent in the prescriptions. The most frequent single herb was white paeony root, accounting for 9.6% in the total number of prescriptions; The most frequent herb pairs were white paeony root-radix rehmanniae preparata and paeony root-angelica sinensis. Among Fu Shan's menopathy prescriptions, 61 herbal pairs showed a correlation coefficient exceeding 0.05, which evolved into 16 pairs of core combinations. The analysis showed that menopathy prescriptions in volume 1 of "Fu Qingzhu's Obstetrics and Gynecology" focused on tonic traditional Chinese medicines involving liver, spleen and kidney and were adjusted according to changes in qi, blood, cold, hot and wet, which could provide a specific reference for further studies on Fu Shan's academic thoughts and traditional Chinese medicine clinical treatment of menopathy.

8.Validity of FASTIN anchor in cervical laminoplasty

[Objective]To evaluate the efficacy and short-term results of FASTIN anchor in cervical expansive open-door laminoplasty(ELAP).[Method]From February 2005 to February 2006,a total of 89 cases of cervical spondylotic myelopathy(CSM) were assessed in this study.Forty patients undergoing expansive open-door laminoplasty(ELAP) by FASTIN anchor were classified as study group.Fouty-nine patients undergoing conventional ELAP served as controls.The clinical results and radiological examinations of both groups were evaluated at 24 months after surgery.[Result]There were no significant differences in operation time,bleeding quantity and recovery rate of Japanese Orthopaedic Association(JOA) scores.The incidences of axial symptoms and C5 palsy in the study group were significantly lower than those in the control group(P
9.EXPERIMENTAL STUDY ON H~+/K~+ EXCHANGE FUNCTION AND cH-K-ATPase mRNA EXPRESSION IN RENAL IMCD CELL MONOLAYER UNDER RESPIRATORY ACID-BASE DISORDERS IN RABBIT

To study the cellular pathophysiological change in kidney IMCD cell under respiratory acid base disorders. 3% or 10% CO 2 was used to initiate chronic respiratory alkalosis(ALG) or chronic respiratory acidosis(ACG) of cultured rabbit IMCD cell. 5% CO 2 was used in control group(CG). Fluorescent probe was used to measure H + /K + exchange function of these cells. In situ hybridization and RT PCR were used to measure cH K ATPase mRNA expression of these cells.The results showed that H + /K + exchange in ACG was significantly higher than that in CG ( P 0 05). It is suggested that chronic respiratory acidosis could induce an increase in H + /K + exchange function and cH K ATPase mRNA expression. However, chronic respiratory alkalosis could induce decreases in H + /K + exchange function and cH K ATPase mRNA expression with no statistical significance, the reason of which calls for further study.
10.Expression and its significance of receptor-interacting protein kinase 3 in ulcerative colitis

Objective To explore the role of receptor‐interacting protein kinase 3 (RIP3)‐mediated necroptosis in ulcerative colitis (UC ) . Methods The colonic mucosa tissues of eighty‐five patients diagnosed with UC were collected .Disease staging and pathological classification of UC patients were evaluated according to Mayo standard and Truelove standard ,respectively .The expressions of RIP3 and tumor necrosis factor (TNF)‐α were detected by immunohistochemistry .The cell inflammation model in HT‐29 cells was induced by interferon (IFN)‐γ.The expression of RIP3 mRNA and interleukin (IL)‐1βmRNA was determined by real time polymerase chain reaction (RT‐PCR) .Twenty healthy individuals were studied as controls .A single factor analysis of variance was performed in the comparison among groups ,and Pearson correlation analysis was used for correlation analysis .Results Among 85 patients with UC , 27 cases were mild ,30 cases were moderate and 28 cases were severe;for pathological classification ,26 were gradeⅠ ,31 were gradeⅡ and 28 were grade Ⅲ .The Mayo score was positively correlated with pathological grade (r=0 .997 , P<0 .05) .The expression of RIP3 and TNF‐α in colonic mucosa tissues of UC group increased along with the pathological classification grade .The expression of RIP3 of gradeⅠto Ⅲ was 1 .81 ± 0 .98 ,2 .77 ± 1 .26 and 4 .86 ± 2 .77 ,respectively ,and the differences were statistically significant (F=26 .10 ,P<0 .05) .The expression of TNF‐αof gradeⅠto Ⅲ was 1 .35 ± 0 .69 ,2 .61 ± 1 .41 and 4 .43 ± 2 .17 ,respectively ,and the differences were statistically significant (F=31 .80 ,P<0 .05) .The expression of IL‐1β mRNA of IFN‐γ group ,IFN‐γ+ z‐VAD group and IFN‐γ+z‐VAD+ TNF‐αgroup was 0 .68 ± 0 .27 ,1 .47 ± 0 .12 and 1 .86 ± 0 .16 ,respectively ,and the differences were statistically significant (F=38 .45 , P<0 .05) .The expression of RIP3 mRNA of IFN‐γgroup ,IFN‐γ+z‐VAD group and IFN‐γ+z‐VAD+TNF‐αgroup was 0 .46 ± 0 .13 ,1 .21 ± 0 .29 and 2 .06 ± 0 .20 ,respectively , and the differences were statistically significant (F= 55 .15 , P< 0 .05) .Conclusions TNF‐α/RIP3 signal pathway is up‐regulated in UC .RIP3‐mediated necroptosis promotes the death of colonic epithelial cells and exacerbates inflammatory response .RIP3‐mediated necroptosis may be an important mechanism in UC .
