1.Sequence analysis,cloning expression and immunogenicity analysis of lactate dehydrogenase gene from Taenia solium

The structure and properties about encoding protein of lactate dehydrogenase A from Taenia solium(Ts LDH-A)were analyzed and predicted by bioinformatics in this study.The immunological characteristics of this novel gene were also analyzed by cloning and expressing.The full-length cDNA encoding Ts LDH-A was identified from the cDNA plasmid library by blastx and rpsblast programs provided by NCBI.The physico-chemical properties and structures of Ts LDH-A were analyzed by tools provided by ExPASy.And the B cell epitopes of Ts LDH-A were predicted by the B Cell Epitope Prediction Tools provided by IEDB Analysis Resource.The PCR amplified coding region of Ts LDH-A was cloned into the prokaryotic expression vector pET-28a (+) and expressed in E.coli BL21 with IPTG induction.The immunogenicity of the purified recombinant protein was analyzed by Western Blotting.It was demonstrated that the amino acid sequence of Ts LDH-A had identity with that of LDH-A from other specie and there was a conserved LDH domain in the deduced amino acid sequence.The full-length cDNA sequence encoding Ts LDH-A included a complete open reading frame(ORF)of 1332 bp and coded to a putative protein with 331 amino acids.The molecular weight of Ts LDH-A was predicted to be 35461.1 Da and the coding protein was demonstrated to contain 3 trans-membrane regions and 4 main B cell epitopes.The active site of L-lactate dehydrogenase located at the epitope aa190-199.The 3 key residues in the catalytic site of enzyme were conserved in different species and located near to each other in spatial position.PCR,double enzyme restriction and DNA sequencing were used to identify pET28a (+)-Ts LDH-A.The recombinant protein could react with the rat's sera as well as the sera from the patients and the swine infected Taenia solium.It is clear that the full-length cDNA sequence encoding Ts LDH-A can be screened from the cDNA library of adult Taenia solium by bioinformatics analysis and can be used to investigate the structure and properties about gene and encoding protein of Ts LDH-A as well as the immunological activities of gene expression in the prokaryotic system.
2.Sperm morphology does not influence fertilization rate in vitro.

OBJECTIVETo determine the correlation of the rate of morphologically normal sperm, sperm head defect, teratozoosperm index (TZI) and sperm deformity index (SDI) with the fertilization outcome of IVF and intracytoplasmic sperm injection (ICSI).
METHODSWe detected normal sperm morphology, sperm head defect, TZI and SDI by Krüger's strict criteria and compared them with the rate of fertilization.
RESULTSIn either IVF or ICSI, the fertilization rate did not show any correlation with the normal sperm morphology rate, the head defect rate, TZI and SDI, nor any statistic differences were found in the rates of fertilization, clinical pregnancy and quality embryos either between the SDI > 1.6 and the SDI < 1.6 groups (P > 0.05) or between the normal sperm morphology rate > 15% and < 15% groups (P > 0.05).
CONCLUSIONSperm morphology has no predictive value for the rate of fertilization in vitro.

3.Clinical effect of trimodal pre-rehabilitation strategy in patients undergoing pancreatoduodenectomy

Objective To observe the clinical effect of trimodal pre-rehabilitation strategy in pa-tients undergoing pancreatoduodenectomy.Methods A total of 110 patients undergoing elective pan-creaticoduodenectomy were selected as the study objects.Patients were randomly divided into control group and observation group according to the odd-even order,with 55 cases in each group.The con-trol group was given routine nursing during perioperative period,and the observation group was given trimodal pre-rehabilitation nursing before surgery based on the control group.The psychological state,quality of life,postoperative indexes and complications of the two groups were observed and com-pared.Serum albumin(ALB),prealbumin(PA)and transferrin(TRF)levels were detected in two groups.Results Postoperative exhaust time,getting out of bed time,first oral feeding time and hos-pital stay in the observation group were significantly shorter than those in the control group(P<0.05).At 1 day before surgery and 14 days after surgery,the levels of ALB,PA,TRF and 36-item Short Form Health Assessment Scale(SF-36)in the observation group were significantly higher than those in the control group,and the Hospital Anxiety and Depression Scale(HADS)scores were signifi-cantly lower than those in the control group(P<0.01 or P<0.05).The incidence of pulmonary infec-tion and venous thrombosis in the observation group was significantly lower than that in the control group(P<0.05).Conclusion Trimodal pre-rehabilitation nursing intervention can effectively im-prove the perioperative nutritional status and psychological state of patients with pancreaticoduodenectomy,improve their quality of life and reduce the incidence of complications.

Lymphangioleiomyomatosis (LAM) is a rare, multisystemic, low-grade neoplasm character-ized by diffuse cystic lesions in the lung.In recent years, emerging imaging examination such as 68Ga-NEB PET-CT scan provides efficient and precise non-invasive diagnostic methods to detect lymphatic circulation abnormalities in LAM patients. The long-term efficacy and safety of sirolimus for LAM has accumulated further evidence, and genetic profiling studies have unveiled more information of genetic mechanisms. Prognosis of LAM has been much improved. We briefly reviewed the research advances of LAM in China and other countires.
