1.NORETHISTERONE OXIME: ESTROGENICITY AND ANTI-IMPLANTATION
Chinese Pharmacological Bulletin 1986;0(04):-
Several comparative studies on anti-implantation and estrogenicities of noret histerone oxime(NETO)and norethisterone(NET) or ethynyl estradiol (EE2)were carried out in order to clarify their relationships. It was observed that NETO was showed much stronger potency than NET, which the anti-implantation, uterotrophic and vaginal cornifi-cation effect were 25, 26, and 21 times, respectively. The ratio of anti-implantation to vaginal cornification of NETO was 14-fold stronger than that of EE2. These results suggest that the increased antiimplantative activity of NETO is related to its stronger estrogenic activity, but other mechanisms contribute to the anti-implantation besides estragenicity.

Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2)and matrix metalloproteinase-9(MMP-9) in MIA PaCa-2 cells blocked by AS-ODN cultured in hypoxia. Methods Heparanase(Hpa) expression of MIA PaCa-2 cells was blocked by AS-ODN and cultured in hypoxia. The expression of MMP-2 and MMP-9 mRNA and proteins in cell lysate was evaluated by RT-PCR and Western blot respectively, and the enzymatic activities of MMP-2 and MMP-9 in supernatants were detected by gelatinase activity assay. Results Hypoxia stimulated mRNA and protein expression of MMP-9 in cultured MIA PaCa-2 cells and elevated at 6h,12 h(P <0.05)and 24 h(P < 0.01).When Hpa expression was inhibited by AS-ODN,the expression of MMP-9 mRNA and protein as well as the gelatinase activity in supernatant decreased dramatically at 12 h and 24 h,especially at 24h(P <0.01),however,no significant difference of MMP-2 expression and gelatinase activity was observed after AS-ODN transfection.(P > 0.05).Conclusion In hypoxia,MMP-9 expression, either mRNA or protein in cultured MIA PaCa-2 cells, increased gradually accompanied with elevated gelatinase activities. When the heparanase expression was inhibited, the MMP-9 mRNA and protein,as well as the gelatinase B activity in supernatant, were decreased dramatically at 12h and 24h,however,no significantly differences of MMP-2 expression and gelatinase A activity were observed after the AS-ODN transfection.

OBJECTIVE:To evaluate the stability of 12 kinds of submicro emulsion in market, and screen the test method for the stability. METHODS:12 kinds of submicro emulsion in market were selected, high pressure sterilization (121 ℃,30 min), high speed centrifugation(4000 r/min,15 min),accelerated test [placing 6 months under temperature of(40±2)℃,relative hu-midity of (75 ± 5)%] were conducted to investigate the pH,particle size and other indexes, and SPSS 22.0 was used to analyze the distribution variance and chi-square test, and investigate the correlation of 3 evaluation methods. RESULTS:In terms of stabili-ty investigation, the pH value of 12 kinds of submicro emulsion decreased to some extent after accelerated test, average particle size of 6 kinds of submicro emulsion samples were greater than 300 nm,the variance of the particle size distribution of 9 kinds ap-peared in 0.05-0.15,the chi-square test results of 8 kinds distributed below 1. The average particle size of 4 kinds of submicro emul-sions changed more than 10 nm after accelerated test. In terms of stability test method,Pearson chi-square progressive significance of high pressure sterilization and accelerated test was 0.665,which was higher than 0.05,indicating there was no correlation (no significance),the stability results of high pressure sterilization can not represent the results of accelerated test;that of high speed centrifugation and accelerated test was 0.004,which was lower than 0.05,indicating stability results between high speed centrifuga-tion and accelerated test results were significantly correlated. CONCLUSIONS:The submicro emulsion in market can meet the re-quirements of stability. To a certain extent, high speed centrifugation can replace the acceleration test.

Objective To study the regulation of heparanase expression by hypoxia and its correlation to the invasiveness of tumor cells. Methods BxPC-3 cells were cultured in hypoxia in vitro and the heparanase mRNA and protein expression were detected by reverse transcriptional polymerase reaction chains (RT-PCR) and western blot respectively. Matrigel invasion assay was used to observe the invasive abilities of tumor cells in hypoxia and in the status of heparanase was inhibited by antisense oligodeoxynucleotide (AS-ODN) targeting to the heparanase gene promoter. Results In normoxia, there was a relatively low level of heparanase mRNA and protein expression in cultured BxPC-3 cells. In hypoxia, heparanase expression, mRNA and protein which expressed consistently, were inhibited slightly at 3h and upregulated significantly at 6h, 12h, 24h and 48h. When the heparanase expression was inhibited by AS-ODN, the heparanase mRNA and protein maintained low in hypoxia, however, the nonsense oligodeoxynucleotide (NS-ODN) did not block upregulation of heparanase expression. In matrigel assay, after 48h incubation, number of BxPC-3 cells that penetrated the Matrigel-coated filter of transwell chamber was increased 96.2% in hypoxia (P<0.01), the Hpa AS-ODN (400 nmol/L) inhibited the invasive cells by 37.2% (P<0.05). Conclusions BxPC-3 cells invasion ability is enhanced by hypoxia through upregulation of heparanase mRNA and protein expression in BxPC-3 pancreatic cancer cell lines.

Chinese Journal of Biotechnology 2013;29(8):1185-1192
Sclareol is a member of labdane type diterpenes mostly used as fragrance ingredient. To enable microbial production of sclareol, synthetic pathways were constructed by incorporating labdenediol diphosphate synthase (LPPS) and terpene synthase (TPS) of the plant Salvia sclarea into Saccharomyces cerevisiae. It was found that sclareol production could be benefited by overexpression of key enzyme for precursor biosynthesis, construction of fusion protein for substrate channeling, and removal of signal peptides from LPPS and TPS. Under optimal shake flask culture conditions, strain S6 produced 8.96 mg/L sclareol. These results provided useful information for development of heterologous hosts for production of terpenoids.
