1.Effect of Bcl-2 knockdown on apoptosis and drug sensitivity of 5-FU in thegastric carcinoma HGC-27cells

Objective To investigate the effect of Bcl-2 gene knockdown on apotosis , proliferation and drug sensitivity of gastric carcinoma HGC-27 cells. Methods The HGC-27 cells were divided into five groups:the untreated control group , the control siRNA group , the specific siRNA targeting Bcl-2 gene group , 5-FU treated group and the combination group (Bcl-2 siRNA and 5-FU treatment). Then flow cytometry and MTT assays were performed to detect the apoptosis and proliferation of HGC-27 cells. The cysteine protease activityand Cytochrome C release level were tested by ELISA method. Results Bcl-2 knockdown enhanced apoptosis and inhibited proliferation of HGC-27 cells. Comparedwith the 5-FU-treated group , the cell apoptosis level, activities of Caspase-3 and Caspase-9, plasma Cytochrome C were significantly increased in the combination group(P <0.01). Conclusion Bcl-2 gene knockdown induced apoptosis, inhibited cell proliferation and enhanced the drug sensitivity of 5-FU in gastric cancer cells , which might be considered as a potential therapeutic strategy forgastric carcinoma.
2.Clinical research on regional citrate anticoagulation for continuous veno-venous hemofiltration in children

Objective To study the reasonable doses, efficacy and safety of regional citrate anticoagulation (RCA) for continuous veno-venous hemofiltration(CVVH) in children. Methods There were 66 patients hospi-ta-lized in Pediatric Intensive Care Unit of Zhujiang Hospital,Southern Medical University treated with RCA-CVVH that were recruited in the study from October 2012 to July 2014. The patients were divided into 4 groups according to their weight:≤10 kg( group Ⅰ) ,20 kg≥weight>10 kg( group Ⅱ) ,30 kg≥weight>20 kg( group Ⅲ) ,>30 kg( groupⅣ),and each group randomly received 2 different doses of anticoagulant acid citrate dextrose formula A(ACD-A):ACD-A(mL/h)=0. 75×blood flow rate(BFR)(mL/min)(A dose) and ACD-A=1. 5×BFR(B dose). Data of hemo-filter duration, activated partial thromboplastin time( APTT) ( systemic and circuit) , ionized calcium( Ca2+) ( systemic and circuit), blood urea nitrogen(BUN), serum creatinine(Cr), alanine aminotransferase(ALT), aspartate amin-otransferase(AST), blood pH, sodium ion(Na+), bicarbonate ion(HCO3-) were collected and analyzed. Results There was no significant difference in BUN,Cr,ALT,AST and APTT of 2 different doses of ACD-A among the groups (all P>0.05);pH of B dose of ACD-A in group Ⅰwas significantly higher than that in A dose(F=7.384,P=0. 015);pH of B dose of ACD-A in groupⅡwas significantly higher than that in A dose(F=4. 492,P=0. 046),HCO3-of B dose of ACD-A in groupⅠwas significantly higher than that in A dose(F=7. 735,P=0. 013);HCO3-of B dose of ACD-A in groupⅡwas significantly higher than that in A dose(F=4. 644,P=0. 042);hemofilter duration of B dose of ACD-A in group Ⅲ was significantly higher than that in A dose(t=-3. 147,P=0. 016);hemofilter duration of B dose of ACD-A in groupⅣwas significantly higher than that in A dose(t=-6. 342,P=0. 000). Conclusions RCA-CVVH is effective and safe for critical children,and different doses of ACD-A for children with different weight can re-duce metabolic alkalosis and enhance regional anticoagulation.
3.Association between interleukin-10-1082 gene polymorphisms and susceptibility of gastric cancer: a meta analysis

Objective To assess the association between interleukin-10-1082 (IL-10-1082) gene polymorphisms and susceptibility of gastric cancer in the Chinese population.Methods Cochrane systematic evaluation was adopted for the analysis.Articles published in Medline,Embase,Cochrane library,CBM,CJFD and CSJD from 1966 to 2012 were retrieved.Case control studies on the correlation between the 1L-10-1082 polymorphism and gastric cancer in Chinese population were collected.Gastric cancer patients were in the gastric cancer group,and healthy individuals were in the control group.Two researchers extracted data and evaluated the quality of literatures independently.Meta analysis was performed to detect whether there were differences between the gastric cancer group and the control group about the distribution of genotypes of IL-10-1082 gene (GG,AA,AG,AA,alleles G and A).The heterogeneity was analyzed using the Q test or I2 test.Fixed effect model or random effect model was adopted,and the results of the meta analysis were presented with odds ratio (OR) and 95% confidence interval (95% CI).Results Thirteen literatures including 5252 patients were included in the analysis.There were 2077 patients in the gastric cancer group and 3175 patients in the control group.The results of meta analysis showed that population with the genotypes GG and AG have higher risk of having gastric cancer when compared with population with the genotype AA (OR =1.76,95% CI 1.33-2.33 ; OR =2.08,95% CI 1.62-2.66,P <0.05).Population with the allele G have higher risk of having gastric cancer when compared with population with allele A (OR =1.67,95% CI 1.31-2.13,P < 0.05).Conclusion The genotypes GG,AG and the allele G of IL-10-1082 gcne of the Chincse population are significantly associated with the increased risk of gastric cancer.
4.Comparative study of real-time two-dimensional shear wave elastography and real-time tissue elastography in the assessment of liver fibrosis in chronic liver disease

Objective To compare the value of two‐dimensional shear wave elastography (2D‐SWE) and real‐time tissue elastography (RTE) in the assessment of liver fibrosis in patients with chronic liver disease. Methods One hundred and ten patients with chronic liver disease scheduled for liver biopsy were studied. Both 2D‐SWE and RTE were performed on these patients in a same day. The correlation coefficient of liver fibrosis level, receiver operating characteristic (ROC) curve of S≥2, and S=4 of 2D‐SWE and RTE were compared according to pathologic results. Results The correlation coefficient of liver fibrosis level for 2D‐SWE ( r =0 7.76, 95% CI 0 6.83 -0 8.45, P =0.000) was higher than that of RTE ( r =0 5.61, 95% CI 0 4.07-0 6.84, P =0.000)( Z =2 9.3, P =0.003). The area under ROC curve for S≥2 and S=4 of 2D‐SWE were 0 8.66 and 0 9.57 respectively, RTE were 0 7.50 and 0 8.43 respectively, which indicated that 2D‐SWE was better than RTE. Conclusions The diagnostic performance of 2D‐SWE was better than RTE for the assessment of liver fibrosis and cirrhosis.
5.Value of real-time shear wave elastography in quantitative diagnosis of breast solid tumors

Objective To obtain the elasticity value of benign and malignant breast solid lesion with real-time shear wave elastography (SWE),in order to assess the value of quantitative elastography with SWE in diagnosis of benign and malignant breast solid lesions.Methods From June 2012 to November 2012,SWE was performed in 125 breast lesions of 103 female patients with pathologically confirmed results,to obtain the elastic modulus (mean,maximum,minimum).Taking pathologic results as reference,ROC curves were used to assess diagnostic performance.Results 87 lesions of 66 patients were pathologically confirmed benign whereas 38 lesions of 37 patients were malignant.The maximum,mean,and minimum elasticity value of benign lesions was 47.2 (7.4-192.0) kPa,22.4 (1.8-90.7) kPa,36.3 (5.3-124.7)kPa; whereas of malignant ones was 192.0(42.7-300.0)kPa,68.1 (0.1-192.0)kPa,138.0(23.9-271.1)kPa.The maximum,minimum and mean elasticity value between benign and malignant lesions had statistically significant difference(P <0.01).The area under the ROC curve of the maximum,minimum and mean elasticity value was 0.935,0.853 and 0.917(P <0.01).Taking 87.65 kPa as the threshold of the maximum elasticity value,the sensitivity was 91.4% and the specificity was 88.5%.Taking 37.15 kPa as the threshold of the minimum elasticity value,the sensitivity was 80.0% and the specificity was 83.9%.Taking 66.85 kPa as the threshold of the mean elasticity value,the sensitivity was 85.7% and the specificity was 88.5%.Conclusions SWE can assess the stiffness of the breast solid neoplasm by quantitatively measuring the elastic modulus,thus providing diagnostic basis in differential diagnosis of breast lesions.SWE elastic modulus value measurement for breast solid lesions provides a new effective and objective diagnostic method.
6.Quantitative study of normal value in health liver by real-time shear wave elastography

Objective To explore the feasibility of detecting liver stiffness by real-time shear wave elastography (SWE) and establish a reference range of shear modulus in healthy liver of adults.Methods 335 adults underwent SWE detection to obtain the shear modulus of livers.The effects of different gender and age on the shear modulus in normal adult were analyzed.Thirty subjects were performed by two operators respectively to evaluate interobserver agreement.Results 329 health volunteers were detected successfully.The success rate was 98.2 %.The mean value of SWE measurements in normal individuals was (5.023 ± 0.966)kPa.The liver shear modulus of healthy adults had a 95% confidence interval from 4.918-5.128 kPa.There were no significant differences between the mean SWE values in men vs women (P =0.218),also among different age groups (P =0.271).No statistically significant was observed between the mean SWE values obtained by two different operators (P =0.498),interobserver ICC value was 0925.Conclusions SWE is a repeatability and stability method to access liver stiffness on healthy subjects.In our study the mean liver stiffness value obtained by SWE in healthy subjects was 5.023 kPa.The mean liver stiffness value obtained by SWE did not influenced by sex and age.
7.Differential expression of ADD3 splicing isoforms between colorectal cancer and normal mucosa tissues

AIM:To investigate the relationship between the expression of adducin 3 (ADD3) and its splicing isoforms and colorectal cancer (CRC).METHODS:The expression of ADD3, ADD3-Ia and ADD3-Ib in 50 pair of CRC tissues , 20 pairs of colorectal polyp tissues , and 2 CRC cell lines SW480 and SW620 before and after oxaliplatin or fluoroura-cil intervention were detected by real-time PCR.The cell activity was determined by MTT assay , the cell migration ability was evaluated by wound-healing assay , and the cell invasion ability was measured by Transwell assay .RESULTS:The expres-sion levels of ADD3 and ADD3-Ib were decreased in the CRC tissues as compared with the normal mucous (P<0.01), and ADD3-Ia/Ib ratio was increased in the CRC tissues (P<0.01).The expression level of ADD3-Ia was higher in T3-4 group than that in T1-2 group (P<0.05).Reduced expression of ADD3, ADD3-Ia and ADD3-Ib in colorectal polyps was observed compared with the normal tissues (P<0.01).Compared with the SW480 cells, the expression levels of ADD3-Ia and ADD3-Ib were lower (P<0.05) and the ADD3-Ia/Ib ratio was higher (P <0.01) in the SW620 cells.After treated with oxalipla-tin or fluorouracil, the cell activity, migration and invasion in the SW620 and SW480 cells were weakened accompanied by the increases in the expression levels of ADD 3, ADD3-Ia and ADD3-Ib to various certain extents .CONCLUSION:In CRC there is a tendency that ADD3-Ib reduction leads to ADD3 decrease, accompanied by an increased ADD3-Ia/Ib ratio.The expression changes of ADD 3 and its splicing isoforms in the CRC may be relevant to its invasion ability .
8.Measurement of hepatic elastic modulus of health control people and chronic liver disease patients using shear wave elastography

ObjectiveTo explore the feasibility of measurement of hepatic elastic modulus using shear wave elastography (SWE).Methods Hepatic elastic modulus of 95 health control people,70 liver fibrosis patients and 86 liver cirrhosis patients were measured by SWE.ResultsThe success rate of imaging of health control people and liver cirrhosis patients were 100%,the success rate of liver fibrosis patients was a little lower,but there was no significant difference between control people and liver cirrhosis patients or fibrosis patients and liver cirrhosis patients (P > 0.05 ).The modulus of elasticity had a significant difference in health control people,liver fibrosis patients and liver cirrhosis patients (P <0.05).The multiple parameter comparison of mean of three groups also showed a significant difference( P <0.05).The health control people had the lowest modulus of elasticity,the liver cirrhosis patients had the highest modulus of elasticity,there was an obvious upward trend.Conclusions SWE can be applied to the measurement of hepatic stiffness,it can better distinguish above three groups.
9.Efficacy and safety of retrograde flexible ureteroscopic lithotripsy with holmium:YAG laser in the treatment of renal stones

Objective To evaluate the efficacy and safety of retrograde flexible ureteroscopic lithotripsy ( RFUL) with holmium:YAG laser for the treatment of renal stones with a large series.Methods The data of 466 patients who underwent RFUL with holmium:YAG laser for the treatment of kidney stones between January 2013 and December 2013 were collected.The maximum diameter of stone was 23 ±16 mm.The stone free rate, complications, retreatment rate were evaluated.Results Out of the 466 patients, the mean operative time and postoperative hospital stay were 33.5 ±18.8 min and 2.3 ±2.0 d.The stone free rate was 67.6% ( 315/466 ) after single procedure, which increased to 69.5% ( 324/466 ) via re-treatments after 3 months.The retreatment rate was 4.3%(20/466), with a total of 493 RFUL procedures performed and 1.06 times per patient.There were 67 (14.4%) cases undergoing complications.Five cases had false passage of ureter orifice causing slight ureteral wall injuries.Steinstrasse occurred in 9 cases, ureteral perforation in 3 cases and perirenal hematoma in 1 case.Overall postoperative fever rate was 10.7%(50/466) with urosepsis in 16 cases (3.4%).When the stone size was≤10, 11-20, 21-30, 31-40, and >40 mm, the stone free rates after a single procedure were 97.0% ( 65/67 ) , 84.2% ( 160/190 ) , 63.1%(70/111), 29.2%(14/48), 12.0%(6/50) (P<0.001), and the postoperative fever rates were 1.5%(1/67), 9.5%(18/190), 13.5% (15/111), 14.6% (7/48), 18.0% (9/50), respectively. The postoperative fever rates were 19.3%(27/140) and 7.1%(23/326) (P<0.001) in the patient with positive and negative preoperative urine leukocyte, and 22.0% ( 22/100 ) and 7.7% ( 28/366 ) ( P <0.001) in patients with positive and negative preoperative urine culture.Conclusions RFUL with holmium:YAG laser is a safe and effective treatment for kidney stones.The postoperative fever rate would increase and stone free rate would reduce with the increased stone size.

Objective To study the expression and the role of interferon-gamma (IFN-?)in murine systemic infection of Scedosporum apiospermum. Methods The murine models of systemic Scedosporum apiospermum infection was established by inoculation of the pathogenic fungi. ELISA and RT-PCR were applied to detect the expression level of IFN-? protein and mRNA in spleens, respectively. Colony formingunit (cfu) of infected kidneys was determined with the plating dilution method. The mean survival time (MST) of the mice was also recorded. Results IFN-? levels in lethal infection group were lower than those of the normal controls (P 0.05 on day 7). IFN-? levels of sublethal infection group were higher than those of normal controls (P 0.05 on day 7).
