Purpose:To investigate the effect of CD40 ligandization on breast cancer cell line and endothelial vein cell line.Methods:The expression of CD40 and its ligand on breast cancer cell line(M231),EVC cell line,fresh clinic breast cancer cell were determined by indirect immunofluorescence assay with flow cytometry(FCM) analysis.Then M231 cells cultured with CD40 agonsitic monoclone antibody,adriamycin alone or in combinations for 72 hours and proliferation of M231cells was determined by MTT assay.FCM was employed to study the cells' death or apopotosis with Annexin V PI assay.Results:M231 and ECV cell line and fresh clinic breast cancer cell are highly expressed CD40 but no CD40L.CD40 ligandization can not only inhibit the proliferation of M231 cell line but also inhibit ECV cell line by promoting the death or apoptosis of these cells.Combined CD40 agonsitic monoclone antibody with adriamycin may obviously inhibit proliferation of M231 and ECV cell line.Conclusions:CD40 ligandization may have double therapeutic effect to breast cancer: inhibit the proliferation of breast cancer cell and inhibit tumor angiogenesis by inhibiting vascular endothelial cell.

Objective To establish a method of isolating and purifying islets from Wistar rat, and to evaluate the biological characters of the isolated and purified islets. Methods The pancreases were surgically removed from adult Wistar rats, carefully minced with scissors as small as possible prior to incubation in 0.9 mg/ml collagenase (Sigma, type Ⅴ) for 10~17min at 37℃. The digested tissue was filtered through a 60 apertures/cm~2 metal sieve and washed with 10% fetal bovine sera (FBS) in Hank’s solution. Purification was performed using Dextran discontinuous density gradient centrifugation to separate the contents of the postcollagenase pellets. Islet samples were stained with DTZ staining for identification. The viability of the purified islet was determined by acridine orange/propidium iodide (AO/PI) double staining. The in vitro function was assayed by different densities of glucose incubating islets for 45min, respectively. Insulin secretion in the supernatant was assayed by radioimmunoassay. Islets were transplanted under the kidney capsule into recipient SD rats that had been rendered diabetic by injecting streptozotocin 50mg/kg, so as to evaluate the in vivo function of islets. Results The islets were recovered between the 11% and 22% Dextran layers, and between the 11% and 1640 layers. The average number of isolated islets was 2181?14 per rat pancreas. After a discontinuous Dextran density gradient purification, about 1826?24 islets were obtained from per rat pancreas, with an average purity over 95%. Islets showed good response to glucose and high efficiency to invert hyperglycemia 7-10d after transplantation. Conclusion These results showed that the digestion methods and purification procedure which we have developed could provide islets from rats for transplantation studies. The biological characters of islets harvested by our method were very well.

Objective To investigate the efficacy and safety of local immune regulation therapy of thyroid disease.Methods Totally 110 patients with confirmed diagnosis of autoimmune thyroid disease were recruited.All patients received thyroid local immune regulation therapy with glucocorticoids 1~3 courses of treatment and followed up for 2~3 years,each being taken good care of during the whole observation.Results About 94.0% of the patients had significantly improved their subjective symptoms,such as pain,fatigue,and lethargy.About 67.7% of the cases had reached normal or markedly improved thyroid function after thyroid local immune regulation therapy.About 92.0% of the volumes of thyroid were reduced.Serum antithyroperoxidase antibody levels were decreased from(338.2?43.2)mU/L to(266.9?42.2)mU/L(P

The extracellular region of gp41 consists of N-terminal fusion peptide and downstream N- and C-heptad repeat (NHR and CHR) regions. The peptidesderived from the NHR and CHR regions, designated N- and C-peptides, respectively, have potent inhibitory activity on the HIV mediated cell fusion. C-peptide T-20 has just got the approval of U.S. FDA, which became the first success of one new class anti-HIV agents, named HIV-fusion inhibitors. However, a relatively long peptide such as T-20 suffers from several limitations including proteolytic sensitivity, large dosage, therefore it is unable to produced by gene engineering. Alternately, shorter peptidic fusion inhibitors and active peptides suitable for gene engineering are pursued. Objective To study the effect of Vincristine-loaded intact human erythrocytes on tumor cell in vitro. Methods VCR was loaded in intact human erythrocytes using a modified hypotonic preswelling technique. The proliferation of K562 cells after incubation with VCR-loaded erythrocytes was detected by MTT. Cellular cycle and apoptosis of K562 cells were analyzed through FACS staining with PI/Rhodamin123, and morphology of apoptotic K562 cells and their nucleus were observed through staining with Hoechest33258. Results VCR-loaded erythrocytes could efficiently inhibit the proliferation of co-cultured K562 cells and induce the cells′ apoptosis in a dosage-dependent manner. Cellular cycle analysis demonstrated that proportion of K562 cells in G 0/G 1 decreased whereas in S+G 2/M increased after incubation with VCR-loaded erythrocytes. [Objective]To evaluate FANCF gene methylation status and its roles in the pathogenesis acute myeloid leukemia (AML).[Methods]Genomic DNA from primary 30 AML patients and 21 AML cell lines were subjected to FANCF methylation analysis by PCR based restriction enzyme digestion assay.FANCF protein expression was detected by Western blot.In addition,FANCF gene methylation status was further analyzed using bisulfate sequencing.[Results] No FANCF methylation was found in primary AML patients.One (4.76 ％) AML cell line contained FANCF methylation in the promoter region.The AML cell line was hypersensitive to MMC with absence of FANCF protein expression.[Conclusion] FANCF methylation is a rare event in AML,and does not contribute to the initiation of AML, but may contribute to the clonal transformation and cellular phenotype maintenance in some AML cell lines.

astaxanthin is an effective antioxidant and natural pigment which has wide application. Phaffia rhodozyma is a good source of astaxantin, but wild strain has limited use in industry because of low production level of astaxanthin. Several mutants of Phaffia rhodozyma were obtained by exerting mutagen N-methyl-N-nitro-N-nitrosoguanidine. The growth curve suggested that pigments were mainly produced in the middle and latter periods of log phase. The pigments were extracted from Phaffia rhodozyma and analysed by esterification, thin layer chromatography and absorption spectrometry. It was proved that astaxanthin, astaxanthin diester and β-carotene were the major components of the pigments produced by Phaffia rhodozyma. We also studied the pigments producing phase of Phaffia rhodozyma. and founded that astaxanthin was stable to light under butylatedhydroxytoluene coexistance.

Objective To analyze effect of three level nursing quality control management model on the quality of the nursing service in the department of general surgery.Methods Analysis in the implementation of nurs-ing quality control before and after nursing quality and the improvement of the examination results of nurses situation in general surgery department of our hospital were compared between the 2012 -2013 without the implementation of three level quality control mode and 2013 -2014 of establishment of the three level nursing quality control manage-ment mode,which was set the nursing quality monitoring nurses,quality monitoring group leader and quality monito-ring nurses in three level nursing quality control system.Results After the implementation of quality control,the quality of basic nursing score[(94.02 ±2.01)points],ward management quality score[(94.13 ±3.04)points],first aid management quality score[(96.22 ±4.06)points]and disinfection and isolation,and the quality score[(95.13 ± 3.04)points]and quality control of basic nursing quality score[(8.121 ±3.31 )points],ward management quality score[(81.35 ±2.34)points],emergency management quality score [(91.42 ±4.36 )points]and disinfection [(81.74 ±3.44)points]degrees and isolation quality scores were significantly higher than those of before three level quality control nursing management model(t =33.07,33.32,8.06,29.17,all P <0.05).Nursing quality control of nursing personnel the technical operation of the average score [(96.35 ±1.03)points],theory test average score [(95.73 ±0.98)points],average and quality control of nursing personnel and technical operation [(89.42 ± 2.49)points],theory test average score[(90.36 ±1.79)points]were obviously higher.The results had statistical significance (t =25.72,26.32,all P <0.05).Conclusion Three level nursing quality control management model established in the general surgery department had remarkable effect on the quality of the nursing service,which is safe and reliable,and improving the quality of nursing.

Objective To investigate the effect of continuous quality improvement nursing on grasping disease knowledge and satisfaction of patients with thyroid tumors.Methods 86 cases of patients with thyroid cancer were enrolled.They were devided into the observation group(42 cases)and the control group(46 cases)according to the date of admission.Patients in control group were treated with routine care,while patients in the observation group were treated with continuous quality improvement nursing on the basis of the control group.The effects of nursing in the two groups were compared and analysized.Results After intervention,the scores of cognitive thyroid tumors,daily preventive health care,thyroid cancer surgery and surgical considerations in the observation group were higher than those in the control group,theses differences were statistically significant between the two groups (t =9.806,10.271, 9.924,12.053,all P <0.05).The rate of satisfaction in the observation group was significantly higther than that in the control group(92.86%/75.00%),theses difference was statistically significant between the two groups (χ2 =9.025,P <0.05).Conclusion Continuous quality improvement nursing can effectively improve the diseases -related knowledge in patients with thyroid benign tumor,which contributes to the smooth operation and improves surgical results.
