Journal of Chinese Physician 2001;0(09):-
Objective To study the effects of D-Ala2-Leu5-enkephalin(DADLE) preconditioning on rabbit donor heart preservation and protective mechanism. Methods 24 rabbits were randomly divided into 4 groups. In group B, after preconditioning with DADLE(1mg?kg -1 ), the donor hearts were then arrested and preserved with Krebs-Henseleit buffer at 4℃ for 4h,and group A without conditioning as controls. Then the donor hearts were transplanted into the abdomen of recipient rabbits. Recovery situation of contraction of the donor heart was compared. The left ventricular tissues were obtained from the donor hearts after 2h of transplantation. The contents of free radicals and ATPase activity were determined and cardiac ultrastructure were observed. Results After 4h cold storage, the heart undergoing DADLE preconditioning in group B showed better recovery of left ventricular development pressure (LVDP).The activity of sodium-potassium ATPase in group B were higher than that in group A. DADLE increased free radical production 2-fold versus group A. Group B showed slighter injury in transmission electron microscopy observation than that in group A.Conclusion Our study demonstrated opioid preconditioning has protective function to rabbit heart injury caused by long term cold storage. The protective mechanism maybe related to increase of free radical content.

Object To research the percutaneous absorption of ?-asarone in various solvents in vitro. Methods To measure the concentration of ?-asarone passed through rat skin in various solvents which contained borneol and azote-ketone by HPLC, count the penetration of accumulation within 24 h and steady velocity. Results 0.1% Borneol could not promote the penetration of ?-asarone; azote-ketone could decrease the penetration of ?-asarone. The penetration of accumulation within 24 h and steady velocity of ?-asarone in 20% ethanol were (352.624?87.049) ?g/cm2, (18.902?4.840) ?g/(cm2?h), respectively. Conclusion The percutaneous absorption of ?-asarone is the best when the solvents contain no promoter.
10.Bone loss (osteoporosis)induced by retinoic acid in mice and its dose-dependent effects
Chinese Pharmacological Bulletin 1986;0(06):-
AIM To study the dose dependent effects on osteoporosis induced by retinoic acid(RA) and to analysis the metabolic characteristics of bone. METHODS Three dosages of retinoic acid were given to the mice for 2 weeks while the body weight(BW) were measured once a week. At the end, femoral dry weight, Bone hydroxyproline, calcium content, Phosphor, Magnesium, Zinc, Manganese and Sulphonium of femora were measured. RESULTS Compared with normal control mice, three dosages of retinoic acid can induce the decrease of femoral dry weight and the short transverse diameter of femoral, Bone hydroxyproline content decreased remarkably, and so did the bone mineral. The Bone HOP and Bone Ca content was coincident with the femoral dry weight, but the lessening of Bone P, Bone Mg, Bone Zn and Bone Mn became inverse ratio with the femoral dry weight, particularly that of the Bone Zn. CONCLUSION Three dosages of retinoic acid in mice can induce osteoporosis with the lessening of bone mass, which characteristics were the losing of Bone HOP and the Bone Ca content in geometric proportion. The levels of Bone P, Bone Mg, and Bone Zn had lost remarkably in the early time, the Bone Zn may play importance function in osteoporosis model induced by retinoic acid.
