1.Significance and expression of p-STAT3 and survivin in primary gallbladder carcinoma

Objective To detect the expression of p-STAT3 and survivin in primary gallbladder carcinoma (PGC) and explore its significance for the genesis and development of PGC.Methods The expression of p-STAT3 and survivin were determined with immunohistochemistry in samples from 45 PGC tissues and 20 chronic cholecystitis.The relationship between the expression of these proteins and various clinicopathological factors was evaluated.Results The expression rate of p-STAT3 and survivin in 45 PGC tissues was 55.6% ( 25/45 ) and 64.4% ( 25/45 ),respectively,which was significantly higher than those in 20chronic cholecystitis tissues 5.0% ( 1/20),10% ( 2/20 ) ( P < 0.01 ).pSTAT3 expression was correlated with survivin ( r =0.830,P <0.01 ).p-STAT3 and survivin protein expression were significantly associated with histopathological grading,lymph node metastatis,nevin staging and 3-year survival rate.Conclusions p-STAT3 and survivin might play a vital role in the development of PGC.The expression of p-STAT3 or survivin was an independent prognostic factor in PGC.
2.Diagnosis and treatment of intraarticular calcaneal fractures

Fractures of the calcaneus are the most common type of tarsal bone injuries, and about 75% of total calcaneal fractures are intraarticular. Despite increasingly more experiences with this type of fractures, however, there is considerable debate regarding their diagnosis and management. Controversies involve the most appropriate classification system, treatment options, indications for surgery, surgical approaches, and postoperative management. On the basis of current main views on the diagnosis and treatment of intraarticular calcaneal fractures, this article holds that for displaced intraarticular calcaneal fractures, operative treatments are superior to conservative ones.
3.Study on the interference therapy of mild hypothermia and correlation between ICAM-1 expression and leukocyte infiltration on cerebral ischemia-reperfusion in rats

4.Association of allogeneic peripheral blood stem cell transplantation with a low incidence of relapse

Objective To evaluate the relationship between transplantation of allogeneic peripheral blood stem cells (allo-PBSCs) and low incidence of leukemic relapse. Methods Thirty-seven adult patients with acute myeloid leukemia in first remission ( n =16) and chronic myeloid leukemia in chronic phase ( n =21) underwent allo-PBSCT from June 1997 to December 1999. A median number of CD34 + cells, CD3 + cells, CD4 + cells and CD8 + cells infused was 7.3 ?10 6/kg, 364?10 6/kg, 210?10 6/kg and 137?10 6/kg, respectively. Busulfan and cyclophosphamide were used as conditioning regimen (BU-CY2). All patients received cyclosporine and methotrexate for GVHD prophylaxis. Results Engraftment of neutrophil and platelet was achieved at a median of day +13(9~28) and day +12(7~19) respectively. Acute GVHD (aGVHD) occurred in 18 of 37 patients ( 48.6% ) with grade II~IV aGVHD ( 24.3% ). Chronic GVHD (cGVHD) developed in 22 (extensive 8, limited 14) out of 32 evaluable patients ( 68.8% ). 28 patients were still alive in complete remission at a median follow-up of 950 days (range 600~ 1 500 days). The main causes of death were aGVHD in two patients, CMV-IP in four patients, cGVHD in two patients, and relapse only in one patient. The probability of disease free survival at 3 years was 75.7% . Conclusion In our series, leukemia recurrence was seen only in one patient ( 2.7% ), suggesting that a high incidence of cGVHD may have an enhanced graft-versus-leukemia effect and hence reduce the incidence of relapse after transplantation.
5.Th1 cell subset、memory、function and tissue distribution

Objective:Understanding of subset、memory cell generation、effector function and organ distribution of Th1 cells in vitro and in vivo.Methods:Different stages of polarized Th1 cells are generated from antigen-specific CD4 +T cells.The expression of cell surface markers and intracellular cytokines(IL-2、IFN-? and IL-4) is determined at a single cell level by FACS. Equal numbers of CFSF-labeled naive CD4 +T cells and different stages of Th1 cells are adoptively transferred into mouse.The production of IFN-? and distribution of Th1 memory cells in lymphoid and non-lymphoid organs are measured. Results:Th1 cell populations can be subdivided into several subsets based upon the production of IL-2 and IFN-?. The percentage of IFN-?-producing cells is increased and IL-2-producing cells is decreased following repeatedly stimulation under Th1 culture condition. Moreover,the expression of CD62L on Th1 cells is reduced after activation and differentiation of CD4 +T cells. 2 weeks post-transfer,the frequency of naive CD4 +T cells is comparable from lymph nodes、spleens and lungs,whereas memory Th1 cells are preferentially detected in spleens and lungs and rapidly produce IFN-? following re-exposure to the same antigen.Conclusion:Th1 cell population is composed of distinct subsets of cells. In comparson with naive CD4 +T cells,memory Th1 cells reveal the changes of surface molecule expression、biologic function and distribution in tissues.
6.CD4~+T cell division is associated with up- and down-regulation of surface molecules and cytokines after stimulation with antigen

Objective:To investigate the correlation between CD4+T cell division and surface molecule expression or cytokine production after stimulation with antigen.Methods:Isolation of CD4+T cells from spleens and lymph nodes of OVA-T cell-receptor transgenic mouse (TCR-Tg ). After stimulation with OVA peptide antigen in the presence of antigen-presenting cells, CD4+T cell division, surface molecule expression and intracellular cytokine production are determined in flow cytometry.Results:CD4+T cells are divided 1-5 times after stimulation for 3 days with antigen. The cell division is associated with both up- and down-regulation of surface molecules such as CD25, CD44, CD62L and CD69 and of cytokines such as IFN-?, IL-4 and IL-10.IL-12 promotes cell division and enhances IFN-? but inhibits IL-4 and IL-10 production.Conclusion:After stimulation with antigen, CD4+T cells are divided in association with the quantitative and qualitative changes of surface molecules and cytokine production.
7.A possible mechanism of relief of symptoms by transmyocardial laser treatment for angina pectoris

Objective To explore the mechanism of relieving symptoms of angina pectoris with transmyocardial laser treatment. Methods Twenty mongrel dogs were divided randomly into 4 groups (5 dogs each): I. sham-operated negative control animals undergoing thoracotomy and pericardiotomy only; II. ligation of the coronary arteries to create acute myocardial ischemia; III. 40 minutes after production of acute myocardial ischemia, transmyocardial punctures were made with high power CO_2 laser; IV. chemical destruction of cardiac nerves by application of 85% phenol to the epicardium after myocardial ischemia. The myocardium was harvested for examination 2 weeks later. Cardiac sympathetic afferent nerve function and innervation was assessed with immunohistochemical staining of tyrosine hydroxylase (TH), which was the specific enzyme of sympathetic nerves, and computer image analysis was done. Histopathological changes in myocardium were also evaluated with haematoxylin-eosin staining. Results Immunohistochemical analysis showed positive staining in I and II groups but negative or weakly positive in III and IV groups. 8.An experimental pathological study of acute intoxication by macleaya cordata

Objective To find out the main target organs, their main pathological changes and to explore the toxic mechanism of the macleaya cordata. Method SD rats were intoxicated by oral administration with extracts of macleaya cordata followed by pathological and electromicroscopic examination of main organs. Results The LD50 was 19.5% when 1.5g/ml macleaya cordata extract was oral administrated. All pocsoned rat's serum GPT, Cr, BUN, CK, CK-MB increased markedly. The EKG were abnormal. The weight of brain, liver, spleen, and kidneys decreased significantly. There were extensive congestion and hemorrhage in heart, brain, liver and kidneys. The mitochondria, endoplasmic reticulum and nuclear membrane of the rat's heart, brain, liver, and kidney were destroyed. Conclusion The results indicated that the toxic target organs of macleaya cordata were the heart, brain, liver and kidneys mainly. The toxic mechanism of macleaya cordata poisoning was the damage of the mitochondrial, endoplasmic reticulum and nuclear membrane directly causd by the macleaya cordata.

Objective: To analyze the prescription dosage for discharged patients in our hospital and investigate the potential risks.Methods: A retrospective research method was adopted to statistically analyze 55 872 discharge instructions from July to September in 2015,and more attention was paid to the prescriptions with dosage over 4 weeks,expecially the ones with 26-week dosage, and the related influencing factors were studied as well.
