1.Controlled Clinical Studies on Fluticasone Propionate Spray for Allergic Rhinitis Children Suffering with Adenoidal Hypertrophy

Objective:To research the efficacy and safety of fluticasone propionate spray for allergic rhinitis children suffering with adenoidal hypertrophy. Methods:Totally 160 allergic rhinitis with adenoids growth children were in the observation group, in which 109 cases were diagnosed as adenoidal hypertrophy. Totally 157 adenoids growth children were in the control group, in which 114 cases were diagnosed as adenoidal hypertrophy. The two groups both received fluticasone propionate spray treatment for 3 months, and then the nasal endoscopic adenoidectomy volume, symptom score and adverse reactions were observed and analyzed. Results:Allergic rhi-nitis children suffering with adenoidal hypertrophy were mainly in the 3-6-year-old children. After the treatment, adenoids was shrinked in 88 patients (80. 73%) in the observation group, which was significantly higher than that in the control group [67(58. 77%), P<0. 01]. Adenoidal hypertrophy was still in 44 patients (35. 03%) of the observation group after the treatment, which was lower than that of the control group [55(68. 13%), P <0. 05]. In addition, the integral differences of nasal congestion, snoring and mouth breathing in the observation group after the treatment were higher than those in the control group, and the differences were statistically significant (P<0. 05). Conclusion:The efficacy of fluticasone propionate spray used in allergic rhinitis children suffering with adenoi-dal hypertrophy is more obvious than that in adenoid hypertrophy children, which can more significantly improve the symptoms of nasal congestion, snoring and mouth breathing.
4.Improvement and Application of Drug Procurement Management Module in Our Hospital

OBJECTIVE:To improve the drug turnover rate and the work efficiency of drug storehouse,and to reduce the cost of drug storage. METHODS:Based on ABC dynamic classification,original drug procurement management module in HIS was im-proved. Scientific,accurate and reasonable drug procurement plan was generated by the system automatically through setting pro-curement proportion coefficient. Drug procurement by type was achieved through adding classification scanning function,based on distribution mode and drug property classification. Related index were compared before and after improvement to evaluate the ef-fects of the improvement. RESULTS:Compared with before improvement,average monthly inventory amount of drugs in our hos-pital reduced by 6.44%,turnover rate increased by 47.13%(increasing from 4.18% to 6.15%),and the time for developing pro-curement plan decreased by 75.00%(decreasing from 8 h to 2 h). CONCLUSIONS:The improvement of drug procurement man-agement module effectively reduce drug inventory and improve turnover rate of drugs and work efficiency.
5.Proficiency evaluation of platelet alloantiboby analysis in 42 platelet Immunology laboratories around the world:the 14t~h ISBT Platelet Immunology Workshop and Cooperative Research Project

Objective To organize the 14th ISBT Platelet Immunology Workshop and Cooperative Research Project,and proficiency evaluation on the techniques and quality level of platelet alloantiboby analysis will be taken effect for the 42 platelet immunology laboratories around the world. Methods Organized and confirmed by Nanning Institute of Transfusion Medicine,9 quality control samples contained the Human Platelet Antigen (HPA) specificity antibody have been supplied to all the participated laboratories in this project. The participants could use the In-House technology or/and market kits to test these quality control samples. The forms and sheets have been provided for recording of results. Results There were 36 laboratories from 23 countries participated in this Cooperative Research Project,and 35 laboratories have reported their results. The appraised consistent rate of the 9 quality control samples ranges from 20% to 97.14%,the HPA antibody specificities have been showed as anti HPA-1a,anti HPA-1b,anti HPA-3a,anti HPA-3a,anti HPA-3b,anti HPA-5b,anti HPA-5a,anti GPIV and anti HPA-5b+15b,respectively. Among the appraised consistent rate,the anti HPA-3b were the lowest and the anti HPA-5b and anti HPA-5a was the highest. Over 10 technologies of platelet alloantiboby analysis were used by the laboratories. Conclusion This international cooperative research project has successfully made the proficiency evaluation and report on the techniques and quality level of platelet alloantiboby analysis for the 35 international platelet immunology laboratories.
6.Relationship between plasma level of endogenous secretory receptor of advanced glycation end products and oxidative stress in diabetic patients with hypertension

The relationship between plasma level of endogenous secretory receptor of advanced glyeation end products(esRAGE)and oxidative stress in diabetic patients with hypertension was investigated.The results showed that there was significant negative correlation between esRAGE and body mass index(BMI,r=-0.33), diastolic blood pressure(DBP,r=-O.38)and oxidative stress(all P<0.05).The levels of plasma esRAGE decreased significantly in diabetic patients with hypertension.BMI,DBP and oxidative stress may participate in determinining the esRAGE level in diabetic patients complicated with hypertension.
7.Optimized method for isolating and culturing human nucleus pulposus cells

BACKGROUND: There are different methods to isolate and culture human nucleus pulposus cells, and the differences in digestive enzymes components and digestion time quite are significant. So how to rapidly and efficiently harvest human nucleus pulposus cells has become a research hotspot. OBJECTIVE: To optimize the digestive enzymes components and digestion methods for the preparation of human nucleus pulposus cells. METHODS: Nucleus pulposus tissue specimens were selected from three adult discs in the Department of Orthopedics, China-Japan Union Hospital of Jilin University. The acute traumatic disc tissues that outstanding to the spinal canal were taken under aseptic conditions, and then the peripheral white annulus and jel y-like nucleus pulposus in the center could be seen. According to different mixed enzyme concentration ratio, the samples were divided into two groups. The enzyme Ⅰ group was treated with 0.2% Ⅱ col agenase; and the mixed enzymeⅡ group was digested with 0.25% trypsin for 30 minutes, and then treated with 0.2% Ⅱ col agenase. According to digestion time, each group was divided into three subgroups: 2 hours group, 4 hours group, and overnight group. Final y, suspended cel volume was decided as 2 mL to count cells. Dulbecco’s modified Eagle’s medium containing fetal bovine serum was used for cel culture in vitro. Trypan blue staining was performed to count total cel number and ratio of living cells. Methylthiazolyldiphenyl-tetrazolium bromide assay was used to detect the growth curve of nucleus pulposus cells. RESULTS AND CONCLUSION: Based on the two digestion enzyme concentration, the number of digested cells in the enzyme Ⅰ group was larger than that in the enzyme Ⅱ group after digested for 2 and 4 hours, but the difference was not significant (P > 0.05). Overnight, cellsurvival rate was decreased in the enzyme Ⅰ group after digested for 2 and 4 hours when compared with the enzyme Ⅱ group, and the difference was significant (P < 0.05). After digested for 4 hours, tissue blocks disappeared, and the number of cells reached maximum. The results indicate that enzyme Ⅰgroup composite with Ⅱ col agenase is benefit for the separation of nucleus pulposus cells, and the digestion time is appropriate to 4 hours. This condition has the advantages of simple operation, high efficiency and low cost, and it considered that digestion of nucleus pulposus tissues with 0.2% Ⅱ col agenase for 4 hours is the best condition to obtain nucleus pulposus cells.
8.Effect of cysteine-rich protein 61 on proliferation and cell cycle in human renal tubular epithelial cells

Objective To investigate the effect of cysteine-rich protein 61 (Cyr61) on proliferation and cell cycle in human renal tubular epithelial cells (HK-2).Methods Cyr61 cDNA was cloned into pEGFP-N2,then HK-2 cells were transfected with the recombinant plasmid pEGFP-N2-Cyr61 by Lipofectamine.The cell proliferation was measured by MTT.The expression level of Cyr61,p-FAK and cyclin dependent cyclin-dependent kinase 2 (CDK2) protein were detected by Western blotting.The cell cycle and cell apoptosis were analyzed by flow cytometry.Results The recombinant plasmid pEGFP-N,-Cyr61 could be transfected into HK-2 efficiently.After transfection,the proliferative activity was significantly increased,the proportion of HK-2 cells in G1 phase decreased and in S-phase increased significantly,the level of cell apoptosis decreased markedly (all P < 0.01).The expressions of Cyr61,p-FAK and CDK2 in Cyr61-transfected group were all amplified significantly (all P < 0.01).Conclusions Cyr61 protein over-expressed in HK-2 cells can increase CDK2 expression throngh FAK pathway,resulting in the promotion of HK-2 cells entering into S phase,cell proliferation and the reduction of cell apoptosis.
10.Application of execution sheet for health education among heart failure patients

Objective To explore the effect of execution sheet for health education in chronic heart failure (CHF) patients. Methods About 50 CHF patients hospitalized from October 2015 to December 2015 were assigned as the control group and another 50 CHF patients hospitalized from January to March 2016 were set up as the experiment groups. The control group was instructed by the regular health education and the experiment group was instructed by the execution sheet for health education. Results The executive ability in the experiment group were stronger than that of control group (P<0.01). The level of self management among the patients in the experiment group was higher than those of the that control group. Conclusions The health education execution sheet increase the patients' self-management and strengthen nursing ability for discharged patients. It is worthy of popularization in clinical practice.
