Objective To investigate the expression of FSCN1 mRNA in breast carcinoma and benign breast diseases and its relationship to clinicopathological characteristics of breast cancer. Methods Sixty-three cases of breast cancer and thirty cases of benign breast diseases were analyzed by in situ hybridization. Results The expression rates of FSCN1 mRNA in breast cancer tissue and benign breast lesions were 65.1% and 90.0% , respectively (P < 0.05). No relation was found between the expression of FSCN1 mRNA and histopathologic grading, ER, PR, karyokinesis figure, patients′ age, tumor size, lymph node metastasis, TNM stage and c-erbB-2 expression in breast cancer. Survival rates of breast cancer patients with carcinoma expressing FSCN1 mRNA were 92.3% , while those patients with tumors which was negative for FSCN1 mRNA were 82.2%, but there was no significance difference. A significant correlation was observed between FSCN1 mRNA and its protein expression (rs = 0.315, P < 0.05). Conclusion FSCN1 is downregulated in breast carcinoma which may be associated with breast carcinogenesis.

Objective To investigate the expression of p53,VEGF,E-cad,CK20 and its clinical meanings in bladder transitional cell carcinoma. Methods The specimen microarrays and immunohistochemistry were applied to detect selected cancer-related protein expression in 45 cases of bladder TCC and 8 cases of normal control. Results Abnormal expression of p53,E-cad and CK20 were detected 100.0%,55.6%,55.6% respectively in bladder TCC group, which were expressed normally in control group with significant differences. However, there was no VEGF expression differences between those 2 groups (87.5% v.86.7%).Abnormal expression intensity of p53 and expression rate of E-cad were closely related to the pathological features.CK20 expression was co-related positively with disease staging rather than grading.VEGF played less role in co-relating with pathological features. Conclusions p53,E-cad and CK20 play an important roles in the development of bladder TCC,and VEGF plays less.Microarrays is suggested to detect a large scale of specimen for multiple proteins,it is effective.

Chinese Journal of Microbiology and Immunology 2008;28(6):486-491
Objective To study the immunoregulatory effects of bone marrow mesenchymal stem cells (MSC) on allogeneic peripheral B lymphocytes in vitro. Methods MSCs were isolated and cultured from bone marrow by gradient centrifugation. Mononuclear cells were isolated routinely from peripheral blood, then monocytes were eliminated by L-leucine methy ester method. Remained T lymphocytes were eliminated by AET-SRBC rosette method. The action of MSCs and its supernatant on B lymphocytes proliferation in the presence of anti-human IgM goat antibodies (anti-IgM) was investigated by MTT. The IgG, IgM in the supernatant were detected by ELISA. The percent of apoptosis B lymphocytes, co-cultured with MSCs for 24 or 48h, was assayed by FACS. Results MSCs and its supernatant inhibited B lymphocytes proliferation and Ig secretion. The inhibitory effect depended on the amount of MSCs and condition of its supernatant. The date of FACS indicated that the apoptosis ratio of B lymphocytes, co-cultured with MSCs for different times, were non-significant. The inhibitory effect of MSCs on B lymphocytes was temporary and reversible. Conclusion MSCs have immunoregulatory effects on B lymphocytes, and its mechanisms are complex, not only correlating with the concentration of MSCs but also the action between cells and the secretory cytokine of MSCs.
